    Treadmill exercise within lower body negative pressure protects leg lean tissue mass and extensor strength and endurance during bed rest.

    Leg muscle mass and strength are decreased during reduced activity and non-weight-bearing conditions such as bed rest (BR) and spaceflight. Supine treadmill exercise within lower body negative pressure (LBNPEX) provides full-body weight loading during BR and may prevent muscle deconditioning. We hypothesized that a 40-min interval exercise protocol performed against LBNPEX 6 days week(-1) would attenuate losses in leg lean mass (LLM), strength, and endurance during 6° head-down tilt BR, with similar benefits for men and women. Fifteen pairs of healthy monozygous twins (8 male and 7 female pairs) completed 30 days of BR with one sibling of each twin pair assigned randomly as the non-exercise control (CON) and the other twin as the exercise subject (EX). Before and after BR, LLM and isokinetic leg strength and endurance were measured. Mean knee and ankle extensor and flexor strength and endurance and LLM decreased from pre- to post-BR in the male CON subjects (P < 0.01), but knee extensor strength and endurance, ankle extensor strength, and LLM were maintained in the male EX subjects. In contrast, no pre- to post-BR changes were significant in the female subjects, either CON or EX, likely due to their lower pre-BR values. Importantly, the LBNPEX countermeasure prevents or attenuates declines in LLM as well as extensor leg strength and endurance. Individuals who are stronger, have higher levels of muscular endurance, and/or have greater LLM are likely to experience greater losses during BR than those who are less fit

    Piloted simulation study of an ILS approach of a twin-pusher business/commuter turboprop aircraft configuration

    A six-degree-of-freedom nonlinear simulation of a twin-pusher, turboprop business/commuter aircraft configuration representative of the Cessna ATPTB (Advanced turboprop test bed) was developed for use in piloted studies with the Langley General Aviation Simulator. The math models developed are provided, simulation predictions are compared with with Cessna flight-test data for validation purposes, and results of a handling quality study during simulated ILS (instrument landing system) approaches and missed approaches are presented. Simulated flight trajectories, task performance measures, and pilot evaluations are presented for the ILS approach and missed-approach tasks conducted with the vehicle in the presence of moderate turbulence, varying horizontal winds and engine-out conditions. Six test subjects consisting of two research pilots, a Cessna test pilot, and three general aviation pilots participated in the study.     Muscleblind-like 3 deficit results in a spectrum of age-associated pathologies observed in myotonic dystrophy.

    Myotonic dystrophy type I (DM1) exhibits distinctive disease specific phenotypes and the accelerated onset of a spectrum of age-associated pathologies. In DM1, dominant effects of expanded CUG repeats result in part from the inactivation of the muscleblind-like (MBNL) proteins. To test the role of MBNL3, we deleted Mbnl3 exon 2 (Mbnl3(ΔE2)) in mice and examined the onset of age-associated diseases over 4 to 13 months of age. Accelerated onset of glucose intolerance with elevated insulin levels, cardiac systole deficits, left ventricle hypertrophy, a predictor of a later onset of heart failure and the development of subcapsular and cortical cataracts is observed in Mbnl3(ΔE2) mice. Retention of embryonic splice isoforms in adult organs, a prominent defect in DM1, is not observed in multiple RNAs including the Insulin Receptor (Insr), Cardiac Troponin T (Tnnt2), Lim Domain Binding 3 (Ldb3) RNAs in Mbnl3(ΔE2) mice. Although rare DM1-like splice errors underlying the observed phenotypes cannot be excluded, our data in conjunction with the reported absence of alternative splice errors in embryonic muscles of a similar Mbnl3(ΔE2) mouse by RNA-seq studies, suggest that mechanisms distinct from the adult retention of embryonic splice patterns may make important contributions to the onset of age-associated pathologies in DM1

    Acetaldehyde and hexanaldehyde from cultured white cells

    <p>Abstract</p> <p>Background</p> <p>Noninvasive detection of innate immune function such as the accumulation of neutrophils remains a challenge in many areas of clinical medicine. We hypothesized that granulocytes could generate volatile organic compounds.</p> <p>Methods</p> <p>To begin to test this, we developed a bioreactor and analytical GC-MS system to accurately identify and quantify gases in trace concentrations (parts per billion) emitted solely from cell/media culture. A human promyelocytic leukemia cell line, HL60, frequently used to assess neutrophil function, was grown in serum-free medium.</p> <p>Results</p> <p>HL60 cells released acetaldehyde and hexanaldehyde in a time-dependent manner. The mean ± SD concentration of acetaldehyde in the headspace above the cultured cells following 4-, 24- and 48-h incubation was 157 ± 13 ppbv, 490 ± 99 ppbv, 698 ± 87 ppbv. For hexanaldehyde these values were 1 ± 0.3 ppbv, 8 ± 2 ppbv, and 11 ± 2 ppbv. In addition, our experimental system permitted us to identify confounding trace gas contaminants such as styrene.</p> <p>Conclusion</p> <p>This study demonstrates that human immune cells known to mimic the function of innate immune cells, like neutrophils, produce volatile gases that can be measured <it>in vitro </it>in trace amounts.</p

    Pulmonary immune responses to Aspergillus fumigatus in an immunocompetent mouse model of repeated exposures

    Aspergillus fumigatus is a filamentous fungus that produces abundant pigmented conidia. Several fungal components have been identified as virulence factors, including melanin; however, the impact of these factors in a repeated exposure model resembling natural environmental exposures remains unknown. This study examined the role of fungal melanin in the stimulation of pulmonary immune responses using immunocompetent BALB/c mice in a multiple exposure model. It compared conidia from wild-type A. fumigatus to two melanin mutants of the same strain, Δarp2 (tan) or Δalb1 (white). Mass spectrometry-based analysis of conidial extracts demonstrated that there was little difference in the protein fingerprint profiles between the three strains. Field emission scanning electron microscopy demonstrated that the immunologically inert Rodlet A layer remained intact in melanin-deficient conidia. Thus, the primary difference between the strains was the extent of melanization. Histopathology indicated that each A. fumigatus strain induced lung inflammation, regardless of the extent of melanization. In mice exposed to Δalb1 conidia, an increase in airway eosinophils and a decrease in neutrophils and CD8(+) IL-17(+) (Tc17) cells were observed. Additionally, it was shown that melanin mutant conidia were more rapidly cleared from the lungs than wild-type conidia. These data suggest that the presence of fungal melanin may modulate the pulmonary immune response in a mouse model of repeated exposures to A. fumigatus conidia

    Supine Lower Body Negative Pressure Exercise Maintains Upright Exercise Capacity in Male Twins during 30 Days of Bed Rest

    Exercise capacity is reduced following both short and long duration exposures to microgravity. We have shown previously that supine lower body negative pressure with exercise (LBNP(sub ex) maintains upright exercise capacity in men after 5d and 15d bed rest, as a simulation of microgravity. We hypothesized that LBNP(sub ex) would protect upright exercise capacity (VO2pk) and sprint performance in eight sets of identical male twins during a 30-d bed rest. Twins within each set were randomly assigned to either a control group (CON) who performed no exercise or to an exercise group (EX) who performed a 40-min interval (40-80% pre-BR VO2pk) LBNP(sub ex) (55+/-4 mmHg) exercise protocol, plus 5 min of resting LBNP, 6 d/wk. LBNP produced footward force equivalent to 1.0- 1.2 times body weight. Pre- and post-bed rest, subjects completed an upright graded exercise test to volitional fatigue and sprint test of 30.5 m. After bed rest, VO2pk was maintained in the EX subjects (-3+/-3%), but was significantly decreased in the CON subjects (-24+/-4%). Sprint time also was increased in the CON subjects (24+/-8%), but maintained in the EX group (8+/-2%). The performance of a supine, interval exercise protocol with LBNP maintains upright exercise capacity and sprint performance during 30 d of bed rest. This exercise countermeasure protocol may help prevent microgravity-induced deconditioning during long duration space flight

    Gemini Near Infrared Spectrograph - Distant Quasar Survey: Prescriptions for Calibrating UV-Based Estimates of Supermassive Black Hole Masses in High-Redshift Quasars

    The most reliable single-epoch supermassive black hole mass (MBHM_{\rm BH}) estimates in quasars are obtained by using the velocity widths of low-ionization emission lines, typically the Hβ\beta λ4861\lambda4861 line. Unfortunately, this line is redshifted out of the optical band at z≈1z\approx1, leaving MBHM_{\rm BH} estimates to rely on proxy rest-frame ultraviolet (UV) emission lines, such as C IV λ1549\lambda1549 or Mg II λ2800\lambda2800, which contain intrinsic challenges when measuring, resulting in uncertain MBHM_{\rm BH} estimates. In this work, we aim at correcting MBHM_{\rm BH} estimates derived from the C IV and Mg II emission lines based on estimates derived from the Hβ\beta emission line. We find that employing the equivalent width of C IV in deriving MBHM_{\rm BH} estimates based on Mg II and C IV provides values that are closest to those obtained from Hβ\beta. We also provide prescriptions to estimate MBHM_{\rm BH} values when only C IV, only Mg II, and both C IV and Mg II are measurable. We find that utilizing both emission lines, where available, reduces the scatter of UV-based MBHM_{\rm BH} estimates by ∼15%\sim15\% when compared to previous studies. Lastly, we discuss the potential of our prescriptions to provide more accurate and precise estimates of MBHM_{\rm BH} given a much larger sample of quasars at 3.20≲z≲3.503.20 \lesssim z \lesssim 3.50, where both Mg II and Hβ\beta can be measured in the same near-infrared spectrum.Comment: 19 pages (AASTeX 6.3.1), 9 figures, accepted for publication in Ap
