Mutation and genomic amplification of the PIK3CA proto-oncogene in pituitary adenomas
The tumorigenesis of pituitary adenomas is poorly understood. Mutations of the PIK3CA proto-oncogene, which encodes the p110-α catalytic subunit of PI3K, have been reported in various types of human cancers regarding the role of the gene in cell proliferation and survival through activation of the PI3K/Akt signaling pathway. Only one Chinese study described somatic mutations and amplification of the PIK3CA gene in a large series of pituitary adenomas. The aim of the present study was to determine genetic alterations of PIK3CA in a second series that consisted of 33 pituitary adenomas of different subtypes diagnosed by immunohistochemistry: 6 adrenocorticotropic hormone-secreting microadenomas, 5 growth hormone-secreting macroadenomas, 7 prolactin-secreting macroadenomas, and 15 nonfunctioning macroadenomas. Direct sequencing of exons 9 and 20 assessed by qPCR was employed to investigate the presence of mutations and genomic amplification defined as a copy number ≥4. Previously identified PIK3CA mutations (exon 20) were detected in four cases (12.1%). Interestingly, the Chinese study reported mutations only in invasive tumors, while we found a PIK3CA mutation in one noninvasive corticotroph microadenoma. PIK3CA amplification was observed in 21.2% (7/33) of the cases. This study demonstrates the presence of somatic mutations and amplifications of the PIK3CA gene in a second series of pituitary adenomas, corroborating the previously described involvement of the PI3K/Akt signaling pathway in the tumorigenic process of this gland
The use of barriers to limit the spread of aquatic invasive animal species: a global review
Aquatic invasive species (AIS) are one of the principal threats to freshwater biodiversity. Exclusion barriers are increasingly being used as a management strategy to control the spread of AIS. However, exclusion barriers can also impact native organisms and their effectiveness is likely to be context dependent. We conducted a quantitative literature review to evaluate the use of barriers to control animal AIS in freshwater ecosystems worldwide. The quantitative aspect of the review was supplemented by case studies that describe some of the challenges, successes, and opportunities for the use of the use of AIS exclusion barriers globally. Barriers have been used since the 1950s to control the spread of AIS, but effort has been increasing since 2005 (80% of studies) and an increasingly diverse range of AIS taxa are now targeted in a wide range of habitat types. The global use of AIS barriers has been concentrated in North America (74% of studies), Australasia (11%), and Europe (10%). Physical barriers (e.g., weirs, exclusion screens, and velocity barriers) have been most widely used (47%), followed by electric (27%) and chemical barriers (12%). Fish were the most targeted taxa (86%), followed by crustaceans (10%), molluscs (3%) and amphibians (1%). Most studies have been moderately successful in limiting the passage of AIS, with 86% of the barriers tested deterring >70% of individuals. However, only 25% of studies evaluated barrier impacts on native species, and development of selective passage is still in its infancy. Most studies have been too short (47% < 1 year, 87% < 5 years) to detect ecological impacts or have failed to use robust before-after-control-impact (BACI) study designs (only 5%). Hence, more effective monitoring is required to assess the long-term effectiveness of exclusion barriers as an AIS management tool. Our global case studies highlight the pressing need for AIS control in many ecoregions, and exclusion barriers have the potential to become an effective tool in some situations. However, the design and operation of exclusion barriers must be refined to deliver selective passage of native fauna, and exclusion barriers should only be used sparingly as part of a wider integrated management strategy
Vascular Flora Of The Legado Das Águas, Reserva Votorantim, Municipalities Of Tapiraí, Miracatú And Juquiá, São Paulo, Brazil
Fundação de Amparo à Pesquisa do Estado de São Paulo (FAPESP)Fundação de Amparo à Pesquisa do Estado de São Paulo (FAPESP)We present a list of vascular plants found in Legado das Águas, Reserva Votorantim, a private reserve having an area of approximately 35,000 ha that spans the municipalities of Tapiraí, Miracatú, and Juquiá, in the state of São Paulo, Brazil. The reserve is part of a complex of state-protected areas including Alto Ribeira Touristic State Park, Intervales State Park, Carlos Botelho State Park, Jurupará State Park, and Serra do Mar State Park. Together, these form an important and large area of continuous and well-preserved forest. Our study, which involved two main floristic surveys in March/April 2013 and April 2014, recorded 768 species, representing 131 families and 432 genera. The majority of species were angiosperms (619) representing 106 families and 370 genera. There were also two species of gymnosperms belonging to two genera in separate families; 147 species of ferns and lycophytes belonging to 23 families and 60 genera. Phosphate fertilization and phosphorus forms in an Oxisol under no-till
Under no-till phosphorus (P) accumulates in a few centimeters of the topsoil layer. Plant residues left on the soil surface release P and organic acids, which may improve P availability and fertilizer efficiency, including both soluble (such as triple super phosphate) and less soluble sources (such as reactive natural phosphates). In this study, soybean response to P fertilizer and P forms in the top 40 cm of an Oxisol were evaluated after surface application of different phosphates in a 5-year-old no-till system. Treatments consisted of 0 or 80 kg ha-1 of total P2O5 applied on the soil surface, both as natural reactive phosphate (NRP) or triple super phosphate (TSP). In addition, 80 kg ha-1 of P2O5 were applied to subplots, in furrows below and beside the soybean (Glycine max L.) seeds, in different combinations of NRP and TSP. Soil samples were taken before and after the soybean growth, down to 0.40 m and soil phosphorus was chemically fractionated. The responses to NRP were similar to TSP, with an increase in P reserves at greater depths, even in non-available forms, such as P-occluded. After the soybean harvest, P-occluded levels were lower at the surface layer, but an increase was observed in the soluble, organic and total P down to 40 cm. An improved P distribution in soil depth, especially regarding the soluble and organic forms, resulted in higher soybean yields, even when the phosphates were applied to the soil surface.Em semeadura direta o fósforo (P) acumula-se na camada mais superficial do solo, mas os resíduos deixados na superfície liberam P e ácidos orgânicos, que podem melhorar a disponibilidade e a eficiência de fertilizantes como o superfosfato triplo e fosfatos naturais reativos. Neste estudo, a resposta da soja à adubação com P e as formas de P até 40 cm de profundidade do solo foram avaliadas após a aplicação de fosfatos em um sistema conduzido em semeadura direta há cinco anos. Os tratamentos consistiram de 0 ou 80 kg ha-1 P2O5 total, aplicados na superfície do solo como fosfato natural reativo (FNR) ou superfosfato triplo (SFT). Nas subparcelas foram aplicados, no sulco de semeadura, 80 kg ha-1 de P2O5, em diferentes combinações de FNR e SFT. Amostras de solo foram coletadas até 0.4 m, antes e depois do cultivo da soja (Glycine max L.), para fracionamento do P. As respostas ao FNR foram semelhantes às do SFT, com aumento das reservas de P em profundidade, mesmo em formas não-disponíveis como P-ocluso. Após a colheita da soja, os teores de P-ocluso diminuíram na camada mais superficial, mas foi observado um aumento nas formas solúvel, orgânica e P - total em toda a espessura de solo estudada. A melhor distribuição do P no solo, principalmente em formas solúvel e orgânica, resultou em maior produtividade da soja, mesmo quando o fertilizante foi aplicado na superfície do solo
Ecological distribution and population structure of Acantholobulus schmitti (Rathbun, 1930) (Crustacea, Decapoda, Xanthoidea) on the southeastern Brazilian coast
This investigation analyzed the ecological distribution and population structure of A. schmitti on the southeastern coast of Brazil. Crabs were sampled monthly from January 1998 to December 1999 at the following bays: Ubatumirim (UBM), Ubatuba (UBA) and Mar Virado (MV). Water and sediment samples were also collected from all sampling sites for an analysis of environmental factors. Acantholobus schmitti was most abundant at UBM (224), followed by UBA (154) and MV (23) but its abundance showed no association with the environmental factors analyzed. The low abundance of these crabs in MV may be due to the high wave action that moved biodetritic material accumulated on the bottom and frequently removed small crabs from their sheltered positions among the shell fragments. The individuals captured included 269 males and 132 females, of which only 4 specimens were brooding females. Juvenile recruitment occurred throughout the year, but was less intense in the spring. The major abundance of individuals as well as of ovigerous females occurred during 1999, when the entrance of the South Atlantic Central Waters (SACW) was stronger than in previous year.
