    Detection of the 13CO(J=6-5) Transition in the Starburst Galaxy NGC 253

    We report the detection of 13CO(J=6-5) emission from the nucleus of the starburst galaxy NGC 253 with the redshift (z) and Early Universe Spectrometer (ZEUS), a new submillimeter grating spectrometer. This is the first extragalactic detection of the 13CO(J=6-5) transition, which traces warm, dense molecular gas. We employ a multi-line LVG analysis and find ~ 35% - 60% of the molecular ISM is both warm (T ~ 110 K) and dense (n(H2) ~ 10^4 cm^-3). We analyze the potential heat sources, and conclude that UV and X-ray photons are unlikely to be energetically important. Instead, the molecular gas is most likely heated by an elevated density of cosmic rays or by the decay of supersonic turbulence through shocks. If the cosmic rays and turbulence are created by stellar feedback within the starburst, then our analysis suggests the starburst may be self-limiting.Comment: 4 pages, 2 figures, accepted by ApJ Letter

Bronchiolitis obliterans (BO) is the most common pathologic finding on biopsy, characterized by fibrous granulation tissue, which obliterates the lumen of the bronchiole. Clinically, in the absence of tissue for pathology, BO syndrome refers to a progressive irreversible drop in the forced expiratory volume in 1 second. Recently, a broader definition of chronic rejection, termed "chronic lung allograft dysfunction", has been used to encompass a more inclusive definition of posttransplant dysfunction. Recently, the lung transplant community has come to realize that chronic rejection may be the final common result after repetitive epithelial insults. Acute rejection, infection, and alloreactivity to mismatched HLA antigens are a few of these insults that damage the surface of the bronchioles. Recent evidence of autoimmunity to the normally hidden structural proteins collagen V and K-α1 tubulin have been correlated with a BO phenotype as well, perhaps correlating the epithelial damage with a mechanism for developing BO lesions. Many immunomodulatory medications and treatments have been studied for effectiveness for the treatment of chronic lung allograft dysfunction. New drugs, which more precisely target the immune system, are being developed and tested. Further study is required, but recent advances have improved our understanding of the pathogenesis and potential intervention for this common and deadly complication of lung transplantation.Keywords: lung transplant, bronchiolitis obliterans syndrome, rejectio

    CstF-64 is necessary for endoderm differentiation resulting in cardiomyocyte defects

    AbstractAlthough adult cardiomyocytes have the capacity for cellular regeneration, they are unable to fully repair severely injured hearts. The use of embryonic stem cell (ESC)-derived cardiomyocytes as transplantable heart muscle cells has been proposed as a solution, but is limited by the lack of understanding of the developmental pathways leading to specification of cardiac progenitors. Identification of these pathways will enhance the ability to differentiate cardiomyocytes into a clinical source of transplantable cells. Here, we show that the mRNA 3′ end processing protein, CstF-64, is essential for cardiomyocyte differentiation in mouse ESCs. Loss of CstF-64 in mouse ESCs results in loss of differentiation potential toward the endodermal lineage. However, CstF-64 knockout (Cstf2E6) cells were able to differentiate into neuronal progenitors, demonstrating that some differentiation pathways were still intact. Markers for mesodermal differentiation were also present, although Cstf2E6 cells were defective in forming beating cardiomyocytes and expressing cardiac specific markers. Since the extraembryonic endoderm is needed for cardiomyocyte differentiation and endodermal markers were decreased, we hypothesized that endodermal factors were required for efficient cardiomyocyte formation in the Cstf2E6 cells. Using conditioned medium from the extraembryonic endodermal (XEN) stem cell line we were able to restore cardiomyocyte differentiation in Cstf2E6 cells, suggesting that CstF-64 has a role in regulating endoderm differentiation that is necessary for cardiac specification and that extraembryonic endoderm signaling is essential for cardiomyocyte development

    Similarity-based gene detection: using COGs to find evolutionarily-conserved ORFs

    BACKGROUND: Experimental verification of gene products has not kept pace with the rapid growth of microbial sequence information. However, existing annotations of gene locations contain sufficient information to screen for probable errors. Furthermore, comparisons among genomes become more informative as more genomes are examined. We studied all open reading frames (ORFs) of at least 30 codons from the genomes of 27 sequenced bacterial strains. We grouped the potential peptide sequences encoded from the ORFs by forming Clusters of Orthologous Groups (COGs). We used this grouping in order to find homologous relationships that would not be distinguishable from noise when using simple BLAST searches. Although COG analysis was initially developed to group annotated genes, we applied it to the task of grouping anonymous DNA sequences that may encode proteins. RESULTS: "Mixed COGs" of ORFs (clusters in which some sequences correspond to annotated genes and some do not) are attractive targets when seeking errors of gene predicion. Examination of mixed COGs reveals some situations in which genes appear to have been missed in current annotations and a smaller number of regions that appear to have been annotated as gene loci erroneously. This technique can also be used to detect potential pseudogenes or sequencing errors. Our method uses an adjustable parameter for degree of conservation among the studied genomes (stringency). We detail results for one level of stringency at which we found 83 potential genes which had not previously been identified, 60 potential pseudogenes, and 7 sequences with existing gene annotations that are probably incorrect. CONCLUSION: Systematic study of sequence conservation offers a way to improve existing annotations by identifying potentially homologous regions where the annotation of the presence or absence of a gene is inconsistent among genomes

    Observation of H_2O in a strongly lensed Herschel-ATLAS source at z = 2.3

    The Herschel survey, H-ATLAS, with its large areal coverage, has recently discovered a number of bright, strongly lensed high-z submillimeter galaxies. The strong magnification makes it possible to study molecular species other than CO, which are otherwise difficult to observe in high-z galaxies. Among the lensed galaxies already identified by H-ATLAS, the source J090302.9-014127B (SDP.17b) at z = 2.305 is remarkable because of its excitation conditions and a tentative detection of the H_2O 2_(02)-1_(11) emission line (Lupu et al. 2010, ApJ, submitted). We report observations of this line in SDP.17b using the IRAM interferometer equipped with its new 277–371 GHz receivers. The H_2O line is detected at a redshift of z = 2.3049 ± 0.0006, with a flux of 7.8 ± 0.5 Jy km s^(-1) and a FWHM of 250 ± 60   km   s^(-1). The new flux is 2.4 times weaker than the previous tentative detection, although both remain marginally consistent within 1.6σ. The intrinsic line luminosity and ratio of H_2O(2_(02) − 1_(11))/CO(8 − 7) are comparable with those of the nearby starburst/enshrouded-AGN Mrk 231, and the ratio I(H_2O)/L_(FIR) is even higher, suggesting that SDP.17b could also host a luminous AGN. The detection of a strong H_2O 2_(02) − 1_(11) line in SDP.17b implies an efficient excitation mechanism of the water levels that must occur in very dense and warm interstellar gas probably similar to Mrk 231

    Effects of Dietary Fat Source on Performance of Lactating Dairy Cows Fed a Pre-mixed Concentrate

    Inclusion of a pre-blended concentrate (OneTrak®, Cargill Inc., Blair, NE) in the total mixed ration (TMR) for dairy cows can simplify the daily mixing of dietary ingredients. A cow’s response to fat supplementation can be affected by other dietary ingredients; however, little is known about production responses to dietary fat in diets with high concentrations of non-forage fiber. This study evaluated cow performance in response to fat sources when the ration contained a pre-blended concentrate composed largely of a non-forage fiber source. Six pens of mid-lactation cows were studied; the addition of saturated fat (Energy Booster 100) and rumen-protected unsaturated fat (Megalac) were compared to a control diet with no added fat. Milk yield tended to increase with both fat sources. Protein concentration decreased with fat supplementation but protein yield did not differ. Efficiency of conversion of feed to milk tended to increase for Megalac compared with Energy Booster. These responses to dietary fat are comparable to those for diets without OneTrak®, suggesting that no unusual dietary interactions with fat supplements are apparent with OneTrak® feeding programs
