    Penicillin-binding proteins of protoplast and sporoplast membranes of Streptomyces griseus strains

    Membrane-bound penicillin-binding proteins (PBPs) of two Streptomyces griseus strains that sporulate well in liquid and solid medium have been investigated during the course of their life-cycle. The PBP patterns were analyzed by sodium dodecylsulphate polyacrylamide-gel electrophoresis and fluorography. One strain (No. 45 H) has only a single band (mol wt: 27,000) in early log phase, and two additional PBPs of higher mol wt (69,000 and 80,000) in the late log phase. The other strain (No. 2682) possessed two bands with mol wts 27,000 and 38,000 which did not change during its vegetative phase. In strain No. 2682, a new PBP with a mol wt of 58,000 appeared in spore membranes while one of those (mol wt 38,000) present in mycelial membranes disappeared. Our results suggest that appearance of the new PBP in the spore may be associated with the sporulation process. The major PBP band (mol wt: 27,000) present in all stages of the life cycle of these strains, may be characteristic of S. griseus while the other PBPs reflect certain stages of the life cycle. A new method was developed for the production of spore protoplasts by consecutive enzymatic treatments.

    Though synthesized with a cleavable signal peptide and devoid of membrane anchors, the 262-amino-acid-residue Streptomyces K15 DD-transpeptidase/penicillin-binding protein is membrane-bound. Overexpression in Streptomyces lividans resulted in the export of an appreciable amount of the synthesized protein (4 mg/litre of culture supernatant). The water-soluble enzyme was purified close to protein homogeneity with a yield of 75%. It requires the presence of 0.5 M-NaCl to remain soluble. It is indistinguishable from the detergent-extract wild-type enzyme with respect to molecular mass, thermostability, transpeptidase activity and penicillin-binding capacity

    Street-connectedness and education in Kenya: Experiences of formal schooling as rationale for inclusive pedagogies of practice

    This paper contributes to the literature on street-connectedness and inclusive education, presenting original research findings from two Kenyan studies aiming to understand street-connected young people’s experiences of education. The first focused on transitioning from the street into education or training to explore the challenges of making that transition. The second, on young people who had lived on the street for extended periods of time and were still there at the time of data generation. From these studies, significant understandings emerged concerning: a) education as motivating initial migrations to the street; b) the role of fear, embarrassment and shame in preventing young people going (back) into formal education; and c) how acceptance and support are key to overcoming feelings of not belonging and challenges faced when transitioning from the street into schools. The paper provides empirical evidence that should be considered when planning inclusive education provision for street-connected young people globally

    National audienceThe present research introduces the strengths of conceptual metaphors (Lakoff & Johnson, 1980) to promote responsible trade products and services as well as their stakeholders and underpinning fundamental principles. We suggest that the use of conceptual metaphors tends to foster the consumer appropriation of its various related societal issues. We then report the results of an experiment testing the impact of conceptual metaphors on consumers’ attitudes towards fair trade. The results show that some of these metaphors positively influence the perceived societal importance of fair trade and the willingness to pay for a fairly traded product. The findings of this study on metaphors are discussed from theoretical and managerial standpoints, and more specifically for socially conscious stakeholders involved in various linguistic and marketing contexts.Cette recherche suggère que les métaphores conceptuelles aident à la valorisation du commerce responsable. Les résultats d’une expérimentation testant l’impact de métaphores conceptuelles sur les attitudes des consommateurs vis-à-vis du commerce équitable sont rapportés. Ils montrent que certaines métaphores exercent une influence sur l’importance sociétale perçue du commerce équitable et la disposition à payer. Ces enseignements sont discutés aux plans théorique et managérial pour les acteurs des filières responsables opérant dans des contextes marketing variés

    Assessing feasibility and maternal acceptability of a biomechanically-optimized supine birth position: A pilot study.

    In order to manage a protracted second stage of labor, "eminence-based" birth positions have been suggested by some healthcare professionals. Recent biomechanical studies have promoted the use of an optimized supine birthing position in this setting. However, uncertainty exists regarding the feasibility of this posture, and its acceptability by women. This pilot study primarily aimed to assess these characteristics. In this monocentric prospective study, 20 women with a protracted second stage of labor were asked to maintain a biomechanically-optimized position for at least 20 minutes at full dilatation. This posture is similar to the McRoberts' maneuver. Maintaining the position for 20 minutes or more was considered clinically relevant and indicative of feasibility and acceptability. Satisfaction with the position was assessed using a Visual Analogue Scale (VAS). A sub-group analysis was performed to assess eventual differences between more and less satisfied patients, according to the median of patients' satisfaction scores. Seventeen patients (85%) maintained the optimized position for at least 20 minutes. The median satisfaction score of these participants was 8 (interquartile range: 1) out of 10. No significant differences were found between the two sub-groups (satisfaction score <8 vs satisfaction score ≥8) regarding general and obstetric characteristics, as well as obstetrical and fetal outcomes. The optimized position is acceptable and feasible for women experiencing a protracted second stage of labor. Further clinical studies are needed to assess the efficiency of such positions when women undergo an obstructed labor
