Anti-inflammatory and antioxidant properties of HDLs are impaired in type 2 diabetes.
ObjectiveIn mice, 4F, an apolipoprotein A-I mimetic peptide that restores HDL function, prevents diabetes-induced atherosclerosis. We sought to determine whether HDL function is impaired in type 2 diabetic (T2D) patients and whether 4F treatment improves HDL function in T2D patient plasma in vitro.Research design and methodsHDL anti-inflammatory function was determined in 93 T2D patients and 31 control subjects as the ability of test HDLs to inhibit LDL-induced monocyte chemotactic activity in human aortic endothelial cell monolayers. The HDL antioxidant properties were measured using a cell-free assay that uses dichlorofluorescein diacetate. Oxidized fatty acids in HDLs were measured by liquid chromatography-tandem mass spectrometry. In subgroups of patients and control subjects, the HDL inflammatory index was repeated after incubation with L-4F.ResultsThe HDL inflammatory index was 1.42 ± 0.29 in T2D patients and 0.70 ± 0.19 in control subjects (P < 0.001). The cell-free assay was impaired in T2D patients compared with control subjects (2.03 ± 1.35 vs. 1.60 ± 0.80, P < 0.05), and also HDL intrinsic oxidation (cell-free assay without LDL) was higher in T2D patients (1,708 ± 739 vs. 1,233 ± 601 relative fluorescence units, P < 0.001). All measured oxidized fatty acids were significantly higher in the HDLs of T2D patients. There was a significant correlation between the cell-free assay values and the content of oxidized fatty acids in HDL fractions. L-4F treatment restored the HDL inflammatory index in diabetic plasma samples (from 1.26 ± 0.17 to 0.71 ± 0.11, P < 0.001) and marginally affected it in healthy subjects (from 0.81 ± 0.16 to 0.66 ± 0.10, P < 0.05).ConclusionsIn patients with T2D, the content of oxidized fatty acids is increased and the anti-inflammatory and antioxidant activities of HDLs are impaired
EmotiBlog: towards a finer-grained sentiment analysis and its application to opinion mining

Preliminary studies demonstrated its relevance as a Machine Learning resource for detecting subjective information. In this paper we explore additional features by a detailed analysis. In addition, we compare EmotiBlog with other well-known Sentiment Analysis resource such as the JRC corpus. Finally, as a result of our research, we developed an Opinion Mining application, which takes into account user opinions when rating the results of a search engine specialized in mobile phones
In vitro culture with gemcitabine augments death receptor and NKG2D ligand expression on tumour cells
Much effort has been made to try to understand the relationship between chemotherapeutic treatment of cancer and the immune system. Whereas much of that focus has been on the direct effect of chemotherapy drugs on immune cells and the release of antigens and danger signals by malignant cells killed by chemotherapy, the effect of chemotherapy on cells surviving treatment has often been overlooked. In the present study, tumour cell lines: A549 (lung), HCT116 (colon) and MCF-7 (breast), were treated with various concentrations of the chemotherapeutic drugs cyclophosphamide, gemcitabine (GEM) and oxaliplatin (OXP) for 24 hours in vitro. In line with other reports, GEM and OXP upregulated expression of the death receptor CD95 (fas) on live cells even at sub-cytotoxic concentrations. Further investigation revealed that the increase in CD95 in response to GEM sensitised the cells to fas ligand treatment, was associated with increased phosphorylation of stress activated protein kinase/c-Jun N-terminal kinase and that other death receptors and activatory immune receptors were co-ordinately upregulated with CD95 in certain cell lines. The upregulation of death receptors and NKG2D ligands together on cells after chemotherapy suggest that although the cells have survived preliminary treatment with chemotherapy they may now be more susceptible to immune cell-mediated challenge. This re-enforces the idea that chemotherapy-immunotherapy combinations may be useful clinically and has implications for the make-up and scheduling of such treatments
2n Gamete formation in the genus Brachiaria (Poaceae: Paniceae).
Microsporogenesis of several Brachiaria species of the Brazilian collection at Embrapa Beef Cattle has been analyzed in detail. This paper reports abnormal cytokinesis in three accessions of three different species (Brachiaria humidicola, 2n = 4x = 36, Brachiaria decumbens, 2n = 4x = 36, and Brachiaria dura, 2n = 6x = 54). Chromosomes paired in bi-, tri-, and quadrivalents in these accessions, whereas chromosome segregation at meiosis I was characterized by exclusion of laggards as micronuclei. In a high number of meiocytes, the first sign of cytokinesis appeared only in metaphase II and did not divide the meiocyte into a dyad. Total absence of cytokinesis was also detected among meiocytes in the second division. Since in both cases the two metaphase plates were very close, they favored the rejoining of chromosome sets after anaphase II and formed a restitutional nucleus in telophase II. Second cytokinesis occurred after telophase II in most meiocytes. Monads, dyads, and triads with n or 2n nuclei were observed among meiotic products. The 2n gametes observed correspond to the first division restitution (FDR). The number of affected cells in each accession was variable, but the number of microspores with restitutional nucleus, including those scored in tetrads and the released ones, did not exceed 9%. Although polyploidy is common in the genus Brachiaria, its origin is still unclear. Current results suggest that 2n gametes may have contributed to the evolutionary history of the genus.CNPGC
Prenatal tobacco smoke exposure increases hospitalizations for bronchiolitis in infants
BACKGROUND: Tobacco smoke exposure (TSE) is a worldwide health problem and it is considered a risk factor for pregnant women's and children's health, particularly for respiratory morbidity during the first year of life. Few significant birth cohort studies on the effect of prenatal TSE via passive and active maternal smoking on the development of severe bronchiolitis in early childhood have been carried out worldwide. METHODS: From November 2009 to December 2012, newborns born at ≥ 33 weeks of gestational age (wGA) were recruited in a longitudinal multi-center cohort study in Italy to investigate the effects of prenatal and postnatal TSE, among other risk factors, on bronchiolitis hospitalization and/or death during the first year of life. RESULTS: Two thousand two hundred ten newborns enrolled at birth were followed-up during their first year of life. Of these, 120 (5.4%) were hospitalized for bronchiolitis. No enrolled infants died during the study period. Prenatal passive TSE and maternal active smoking of more than 15 cigarettes/daily are associated to a significant increase of the risk of offspring children hospitalization for bronchiolitis, with an adjHR of 3.5 (CI 1.5-8.1) and of 1.7 (CI 1.1-2.6) respectively. CONCLUSIONS: These results confirm the detrimental effects of passive TSE and active heavy smoke during pregnancy for infants' respiratory health, since the exposure significantly increases the risk of hospitalization for bronchiolitis in the first year of lif

Supramolecular interactions in clusters of polar and polarizable molecules
We present a model for molecular materials made up of polar and polarizable
molecular units. A simple two state model is adopted for each molecular site
and only classical intermolecular interactions are accounted for, neglecting
any intermolecular overlap. The complex and interesting physics driven by
interactions among polar and polarizable molecules becomes fairly transparent
in the adopted model. Collective effects are recognized in the large variation
of the molecular polarity with supramolecular interactions, and cooperative
behavior shows up with the appearance, in attractive lattices, of discontinuous
charge crossovers. The mean-field approximation proves fairly accurate in the
description of the gs properties of MM, including static linear and non-linear
optical susceptibilities, apart from the region in the close proximity of the
discontinuous charge crossover. Sizeable deviations from the excitonic
description are recognized both in the excitation spectrum and in linear and
non-linear optical responses. New and interesting phenomena are recognized near
the discontinuous charge crossover for non-centrosymmetric clusters, where the
primary photoexcitation event corresponds to a multielectron transfer.Comment: 14 pages, including 11 figure
Career models in the new tripolar order. Political profiles of the Italian MPs after 2022 general elections.
The 2022 general elections confirmed the instability of the Italian political system and in its shift from a bipolar to a tripolar competition dynamic. The article explores similarities and differences among MPs elected in 2022 during such electoral turmoil, aiming to understand their profiles and career paths. Specifically, the research investigates whether certain elements such as gender and affiliation with an organisational thick or thin party may influence the MPs’ career models. We run a multinomial logistic regression to test the effects on multilevel career patterns by relying on an original dataset which comprises the socio-economic and political characteristics of the current MPS and their career trajectories for a total of 588 observations. Our findings show that women are often newcomers, and they are more likely to display an alternative career model. They struggle to fully capitalize on the opportunities offered by multilevel democracy, both before and after attaining their position as MPs. Furthermore, MPs belonging to a party with a strong organisation are more likely to develop unidirectional and integrated models, indicating their ability to navigate the multilevel context with well-planned and structured career paths
The good, the bad and the implicit: a comprehensive approach to annotating explicit and implicit sentiment
We present a fine-grained scheme for the annotation of polar sentiment in text, that accounts for explicit sentiment (so-called private states), as well as implicit expressions of sentiment (polar facts). Polar expressions are annotated below sentence level and classified according to their subjectivity status. Additionally, they are linked to one or more targets with a specific polar orientation and intensity. Other components of the annotation scheme include source attribution and the identification and classification of expressions that modify polarity. In previous research, little attention has been given to implicit sentiment, which represents a substantial amount of the polar expressions encountered in our data. An English and Dutch corpus of financial newswire, consisting of over 45,000 words each, was annotated using our scheme. A subset of this corpus was used to conduct an inter-annotator agreement study, which demonstrated that the proposed scheme can be used to reliably annotate explicit and implicit sentiment in real-world textual data, making the created corpora a useful resource for sentiment analysis
