    We report on the design, synthesis, and properties of innovative, planar, π-conjugated compounds in which a thiophene ring is fused with the skeleton of the naturally occurring dye isatin. The synthesis is achieved in high yields making use of an intramolecular direct arylation reaction as the key step, making the overall process potentially scalable. The synthetic sequence has been demonstrated also for an isatin bearing fluorine substituents on the aromatic ring. NMR and X-ray studies demonstrate the crosstalk occurring between the fused, coplanar, and conjugated moieties, making these novel dyes with a donor–acceptor character. Cyclic voltammetry and UV–vis studies confirm very interesting HOMO–LUMO levels and energy gaps for the new compounds

    The three nitrophenyl–cyclam derivatives (nitrocyclams): 1-(4-nitrophenyl)-1,4,8,11-tetraazacyclotetradecane (2), 1-(2-nitrophenyl)-1,4,8,11-tetraazacyclotetradecane (3), and 1-(2,4-dinitrophenyl)-1,4,8,11-tetraazacyclotetradecane (4), in an MeCN solution, specifically incorporate the CuII ion according to an irreversible process signaled by disappearance of the yellow color for a concentration c < 1 × 10–4 M and by a yellow-to-red color change for c ≥ 1 × 10–3, and must be considered efficient and specific dosimeters of copper(II) salts. When present in the ortho position of the nitrophenyl substituent, the −NO2 group coordinates the CuII according to a scorpionate mode, while the metallocyclam system exhibits a trans-I configuration. In an MeCN solution the red trans-I-[CuII(3)]2+ and trans-I-[CuII(4)]2+ scorpionate complexes slowly convert into the violet trans-III scorpionate complexes. Kinetic aspects of the trans-I-to-trans-III configurational rearrangement were investigated in detail for the [CuII(4)]2+ system. In particular, the conversion is spectacularly accelerated by catalytic amounts of Cl–, NCO–, and F–. While for Cl– and NCO– the effect can be associated with the capability of the anion to stabilize through coordination a possible dissociative intermediate, the amazingly powerful effect of F– must be related to the preliminary deprotonation of one N–H fragment of the macrocycle, driven by the formation of the HF2– ion. Most of the metal complex species studied in solution were isolated in a crystalline form, and their molecular structures were elucidated through X-ray diffraction studies.     Prostatic stromal tumor with fatal outcome in a young man: histopathological and immunohistochemical case presentation

    Stromal tumors of the prostate are rare and only a few cases have been described in the literature, including exceptional cases of stromal tumors with unknown malignant potential (STUMP) and a fatal outcome in young patients. Morphologically distinguishing a STUMP from a stromal sarcoma of the prostate (PSS) is still a challenge. We describe the histopathological and immunohistochemical findings in a 34-year-old man with a malignant specialized cell stromal tumor of the prostate that was diagnosed initially as STUMP, and he developed lung metastases within a few months. The patient attended our hospital because of lower urinary tract symptoms, after having excreted tissue through the urethra a few months before. Ultrasonography and urethrocystoscopy examinations showed a mass arising from the verumontanum, and a transurethral resection (TUR) revealed a highgrade spindle cell sarcoma reminiscent of a phyllode tumor of the breast. The tumor cells were immunoreactive for vimentin, progesterone receptor and, focally, CD34. The preliminary histological findings were subsequently confirmed after radical prostatectomy. The patient developed bilateral lung metastases and died 25 months after the initial diagnosis. Although rare in young patients, the challenging differential diagnosis of STUMP and PSS means that a prostate STUMP diagnosis made on the basis of biopsy or TUR specimens also requires urethrocystoscopic monitoring for the early detection of any progression to PSS. Radical prostatectomy should also be carefully considered

    Thymidylate synthetase mRNA levels are increased in liver metastases of colorectal cancer patients resistant to fluoropyrimidine-based chemotherapy

    BACKGROUND: Fluoropyrimidines such as 5-fluorouracil (5-FU) and 5-fluoro-2'deoxyuridine (FUDR) are among the most effective chemotherapeutic agents for treatment of metastatic colorectal cancer (CRC). Increased expression of thymidylate synthetase (TS) in CRC metastases has been proposed to be an important mechanism of resistance to fluoropyrimidine-based chemotherapy. METHODS: The present study investigated whether TS mRNA levels in liver metastases of 20 CRC patients before treatment with FUDR by hepatic arterial infusion (HAI) correlated with frequency of clinical response or survival duration. RESULTS: Median survival duration of patients with TS mRNA levels above and below the median was 15 and 18 months, respectively (p > 0.05). Clinical response was achieved in 40% of patients with low TS mRNA levels, but in only 20% of patients with high TS mRNA levels (p = 0.01). TS mRNA levels were also measured for liver metastases of 7 of the patients that did not achieve a clinical response. A statistically significant increase in expression of TS mRNA was observed for liver metastases resistant to chemotherapy (21 ± 14) in comparison to liver metastases of the same patients before chemotherapy (8 ± 4) (p = 0.03). CONCLUSION: This is the first report to demonstrate increased TS expression in liver metastases from CRC patients resistant to fluoropyrimidine based chemotherapy. These findings are consistent with previous studies indicating that increased TS expression is associated with resistance to fluoropyrimidine-based chemotherapy

    The first single-crystal structure refinement of aschamalmite (Pb6Bi2S9) from Susa Valley (Piedmont, Italy) is reported. The mineral is monoclinic, C2/m, a = 13.719(1) Å, b = 4.132(1) Å, c = 31.419(3) Å, β = 90.94(1)°, V = 1 780.8(4) Å3, Z = 4. The Pb6Bi2S9 compound crystallizes also in an orthorhombic form as heyrovskyite (Cmcm) and our study is focused on understanding the reason leading to a change in symmetry. The aschamalmite structure forms because of ordering between Pb and Bi on the margins of the two octahedral layers that are symmetrically equivalent in heyrovskyite. The two alternate set of octahedral slabs are not related by a crystallographic mirror plane and the symmetry decreases to monoclinic. The cation ordering couples opposite sequences of Pb and Bi octahedra at the margins of slabs. In particular, the succession [Me4A]Bi-[Me5A]Pb-[Me4A]Bi-[Me5A]Pb faced to the series [Me4B]Pb-[Me5B]Bi-[Me4B]Pb-[Me5B]Bi occurs in about 70% of the unit-cells of the crystal, while the contrary sequence ([Me4A]Pb-[Me5A]Bi-[Me4A]Pb-[Me5A]Bi faced to [Me4B]Bi-[Me5B]Pb-[Me4B]Bi-[Me5B]Pb) occurs in the remaining unit-cells. The marginal octahedra have ideal populations (a.p.f.u.): [Me4A]1.40Bi+0.60Pb, [Me4B]1.40Pb+0.60Bi, [Me5A]1.40Pb+0.60Bi, [Me5B]1.40Bi+0.60Pb, in agreement with our structure-refinement results. The probable site populations for pure heyrovskyite have been proposed, as well as the reasons that prevent the formation of a completely ordered monoclinic phase
