    Pharmacokinetic/Pharmacodynamic Analysis of Tedizolid Phosphate Compared to Linezolid for the Treatment of Infections Caused by Gram-Positive Bacteria

    Tedizolid and linezolid have antibacterial activity against the most important acute bacterial skin and skin-structure infection (ABSSSIs) pathogens. The objective of this work was to apply PK/PD analysis to evaluate the probability of attaining the pharmacodynamic target of these antimicrobials based on the susceptibility patterns of different clinical isolates causing ABSSSI. Pharmacokinetic and microbiological data were obtained from the literature. PK/PD breakpoints, the probability of target attainment (PTA) and the cumulative fraction of response (CFR) were calculated by Monte Carlo simulation. PTA and CFR are indicative of treatment success. PK/PD breakpoints of tedizolid and linezolid were 0.5 and 1 mg/L, respectively. Probability of treatment success of tedizolid was very high (>90%) for most staphylococci strains, including MRSA and coagulase-negative staphylococci (CoNS). Only for methicillin- and linezolid-resistant S. aureus (MLRSA) and linezolid resistant (LR) CoNS strains was the CFR of tedizolid very low. Except for LR, daptomycin-non-susceptible (DNS), and vancomycin-resistant (VRE) E. faecium isolates, tedizolid also provided a high probability of treatment success for enterococci. The probability of treatment success of both antimicrobials for streptococci was always higher than 90%. In conclusion, for empiric treatment, PK/PD analysis has shown that tedizolid would be adequate for most staphylococci, enterococci, and streptococci, even those LR whose linezolid resistance is mediated by the cfr gene.This research was funded by the University of the Basque Country UPV/EHU (GIU17/032)

    El presente Trabajo Fin de Máster de Profesorado en Educación Secundaria y Bachillerato de la especialidad de Lengua castellana y Literatura pertenece a la modalidad A3 y presenta una propuesta didáctica de animación a la lectura de uno de nuestros clásicos literarios: el Quijote. Dado el rechazo que suelen producir las obras clásicas en los adolescentes y la falta de hábito lector de su generación, se presentan variadas actividades que tienen como objetivo fomentar ese hábito, desarrollar la competencia lectora y producción literaria y, ante todo, animar y entender la lectura del Quijote como fuente de placer a través de metodologías activas, lúdicas y recursos tecnológicos como base de dicha propuesta.<br /

    Copper(II) complexes of quinoline polyazamacrocyclic scorpiand-type ligands: X-ray, equilibrium and kinetic studies

    The formation of Cu(II) complexes with two isomeric quinoline-containing scorpiand-type ligands has been studied. The ligands have a tetraazapyridinophane core appended with an ethylamino tail including 2-quinoline (L1) or 4-quinoline (L2) functionalities. Potentiometric studies indicate the formation of stable CuL2+ species with both ligands, the L1 complex being 3–4 log units more stable than the L2 complex. The crystal structure of [Cu(L1)](ClO4)2·H2O shows that the coordination geometry around the Cu2+ ions is distorted octahedral with significant axial elongation; the four Cu–N distances in the equatorial plane vary from 1.976 to 2.183 Å, while the axial distances are of 2.276 and 2.309 Å. The lower stability of the CuL22+ complex and its capability of forming protonated and hydroxo complexes suggest a penta-dentate coordination of the ligand, in agreement with the type of substitution at the quinoline ring. Kinetic studies on complex formation can be interpreted by considering that initial coordination of L1 and L2 takes place through the nitrogen atom in the quinoline ring. This is followed by coordination of the remaining nitrogen atoms, in a process that is faster in the L1 complex probably because substitution at the quinoline ring facilitates the reorganization. Kinetic studies on complex decomposition provide clear evidence on the occurrence of the molecular motion typical of scorpiands in the case of the L2 complex, for which decomposition starts with a very fast process (sub-millisecond timescale) that involves a shift in the absorption band from 643 to 690 nm

    Abrogating mitochondrial ROS in neurons or astrocytes reveals cell-specific impact on mouse behaviour

    Article 101917, (2021)[EN]Cells naturally produce mitochondrial reactive oxygen species (mROS), but the in vivo pathophysiological significance has long remained controversial. Within the brain, astrocyte-derived mROS physiologically regulate behaviour and are produced at one order of magnitude faster than in neurons. However, whether neuronal mROS abundance differentially impacts on behaviour is unknown. To address this, we engineered genetically modified mice to down modulate mROS levels in neurons in vivo. Whilst no alterations in motor coordination were observed by down modulating mROS in neurons under healthy conditions, it prevented the motor discoordination caused by the pro-oxidant neurotoxin, 3-nitropropionic acid (3-NP). In contrast, abrogation of mROS in astrocytes showed no beneficial effect against the 3-NP insult. These data indicate that the impact of modifying mROS production on mouse behaviour critically depends on the specific cell-type where they are generated

    Estudio del yeísmo en el español de Barcelona a partir de materiales de PRESEEA

    En enero de 2006, el equipo de investigación sociolingüística de la Universidad de Barcelona, integrado por profesores y doctorandos del Departamento de Filología Hispánica, se incorporó al Proyecto para el Estudio Sociolingüístico del Español de España y de América (PRESEEA), vinculado a la Asociación de Lingüística y Filología de América Latina (ALFAL), y que tiene como coordinador general a Francisco Moreno Fernández, Catedrático de Lengua Española de la Universidad de Alcalá. Dicho proyecto se organiza en una red internacional de equipos de investigación cuyo objetivo central es el de reunir un ingente corpus oral, sociolingüísticamente representativo, de un gran número de ciudades pertenecientes al mundo hispánico. La primera fase del proyecto, que se presentó en 1996, abarca hasta el año 2010 (Moreno Fernández 2006).1 Para el estudio de Barcelona, en el seno del grupo PRESEEA – BARCELONA – ES diseñamos una plantilla que contempla una representación de 108 informantes (los cuales deben haber nacido en la ciudad, haber llegado a ella antes de cumplir los diez años o llevar viviendo en el lugar más de veinte), distribuidos en tres variables de preestratificación: sexo, edad y nivel de instrucción. A este perfil general añadimos la variable ‘lengua primera’ (catalán / español), vista la repercusión que alcanza la convivencia de las dos lenguas en la ciudad (Torres 2007). En las páginas que siguen, nos proponemos abordar el avance real del yeísmo en la Ciudad Condal a partir de unas muestras seleccionadas de los datos de PRESEEABARCELONA- ES. El trabajo, que básicamente es de carácter fonético y presenta un análisis acústico centrado en aspectos frecuenciales, de duración y de intensidad de las muestras mencionadas, pretende ser un primer acercamiento al estadio en el que se encuentra el fenómeno en esta área específica

    Angiotensin II Induces Leukocyte–Endothelial Cell Interactions In Vivo Via AT1 and AT2 Receptor–Mediated P-Selectin Upregulation

    Background—Angiotensin II (Ang II) plays a critical role in the development of vascular lesions in hypertension, atherosclerosis, and several renal diseases. Because Ang II may contribute to the leukocyte recruitment associated with these pathological states, the aim of the present study was to assess the role of Ang II in leukocyte–endothelial cell interactions in vivo. Methods and Results—Intravital microscopy of the rat mesenteric postcapillary venules was used. Sixty minutes of superfusion with 1 nmol/L Ang II induced a significant increase in leukocyte rolling flux (83.8±20.7 versus 16.4±3.1 cells/min), adhesion (11.4±1.0 versus 0.8±0.5 cells/100 µm), and emigration (4.0±0.7 versus 0.2±0.2 cells/field) without any vasoconstrictor activity. These effects were not mediated by mast cell activation. Intravenous pretreatment with AT1 (losartan) or AT2 (PD123,319) receptor antagonists significantly reduced Ang II–induced responses. A combination of both receptor antagonists inhibited the leukocyte rolling flux, adhesion, and extravasation elicited by Ang II at 60 minutes. Pretreatment of animals with fucoidin or an adhesion-blocking anti–rat P-selectin monoclonal antibody abolished Ang II–induced leukocyte responses. Furthermore, rat platelet P-selectin expression was not affected by Ang II stimulation.     Quantification of Ceftaroline in Human Plasma Using High-Performance Liquid Chromatography with Ultraviolet Detection: Application to Pharmacokinetic Studies

    This study was conducted to develop a rapid, simple and reproducible method for the quantification of ceftaroline in plasma samples by high-performance liquid chromatography with ultraviolet detection (HPLC-UV). Sample processing consisted of methanol precipitation and then, after centrifugation, the supernatant was injected into the HPLC system, working in isocratic mode. Ceftaroline was detected at 238 nm at a short acquisition time (less than 5 min). The calibration curve was linear over the concentration range from 0.25 to 40 µg/mL, and the method appeared to be selective, precise and accurate. Ceftaroline in plasma samples was stable at −80 °C for at least 3 months. The method was successfully applied to characterize the pharmacokinetic profile of ceftaroline in two critically ill patients and to evaluate whether the pharmacokinetic/pharmacodynamic (PK/PD) target was reached or not with the dose regimen administered.This research was funded by Department of Education of the Basque Government (PIBA 2019-57) and by the University of the Basque Country UPV/EHU (GIU 17/32)

    Pseudomonas aeruginosa Susceptibility in Spain: Antimicrobial Activity and Resistance Suppression Evaluation by PK/PD Analysis

    Pseudomonas aeruginosa remains one of the major causes of healthcare-associated infection in Europe; in 2019, 12.5% of invasive isolates of P. aeruginosa in Spain presented combined resistance to ≥3 antimicrobial groups. The Spanish nationwide survey on P. aeruginosa antimicrobial resistance mechanisms and molecular epidemiology was published in 2019. Based on the information from this survey, the objective of this work was to analyze the overall antimicrobial activity of the antipseudomonal antibiotics considering pharmacokinetic/pharmacodynamic (PK/PD) analysis. The role of PK/PD to prevent or minimize resistance emergence was also evaluated. A 10,000-subject Monte Carlo simulation was executed to calculate the probability of target attainment (PTA) and the cumulative fraction of response (CFR) considering the minimum inhibitory concentration (MIC) distribution of bacteria isolated in ICU or medical wards, and distinguishing between sample types (respiratory and non-respiratory). Ceftazidime/avibactam followed by ceftolozane/tazobactam and colistin, categorized as the Reserve by the Access, Watch, Reserve (AWaRe) classification of the World Health Organization, were the most active antimicrobials, with differences depending on the admission service, sample type, and dose regimen. Discrepancies between EUCAST-susceptibility breakpoints for P. aeruginosa and those estimated by PK/PD analysis were detected. Only standard doses of ceftazidime/avibactam and ceftolozane/tazobactam provided drug concentrations associated with resistance suppression.This research was funded by the UPV/EHU (GIU 20/048)
