    The Oil Weapon: Myth of China's Vulnerability

    The geopolitical canvass on which China plots its strategy for energy security displays a ubiquitous presence of one country: the United States. Chinese energy security planners must reckon with America's ravenous consumption of imported oil, its strategic alliances with other heavy importers of oil in Asia, its overseas military operations in the heart of the world's leading oil producing region, its naval dominion over the world's oil transportation routes, and the global domination of U.S. oil companies or multinational oil companies heavily capitalized by American investment. This is the context in which China pursues its energy security, sometimes blandly described as 'conservation and diversification of supply', which masks the nation's real struggle to satisfy its rapidly growing energy needs without exposing its energy lifelines to external forces that may, intentionally or not, betray China's interests

    Protein synthesis in chloroplasts

    The function of chloroplast ribosomes was investigated by analysing the products of in vitro protein synthesis by isolated pea chloroplasts. Since previous attempts at identifying newly synthesised proteins in isolated chloroplasts had either been unsuccessful or inconclusive, careful attention was paid both to the choice of chloroplast preparation and to the analytical techniques used to identify labelled proteins. A rapid method of chloroplast isolation was used which gave a chloroplast preparation which contained about 50% intact chloroplasts and showed high rates of amino acid incorporation into protein. It was felt that the proteins synthesised by these chloroplasts would accurately reflect the nature and pattern of protein synthesis which occurs in chloroplasts in vivo. High rates of incorporation would also aid identification of newly-synthesised proteins. Amino acid incorporation was shown to be sensitive to selective inhibitors of 70S-type ribosomes, but was not affected by inhibitors of 80S-type ribosomes. In addition, incorporation was shown to by insensitive to ribonuclease, suggesting that protein synthesis was taking place in intact chloroplasts. When the labelled chloroplasts were fractionated by differential centrifugation, approximately 25% of this incorporation was present in a 150 000 x g chloroplast supernatant fraction. Further analysis was confined to this supernatant fraction since only released, and therefore completed, polypeptides should be present in this fraction, thus aiding identification. The 150 000x g supernatant fraction was analysed on polyacrylamide gels in the presence and absence of a denaturant, sodium dodecyl sulphate, and by gel chromatography on Sephadex G100 in a sodium dodecyl sulphate-containing buffer. Only one polypeptide was found to be labelled by all these procedures. This polypeptide was identified as the large subunit of Fraction 1 protein, a major protein constituent of the chloroplast. Identity of the in vitro product present in the soluble phase of the chloroplast with the large subunit of Fraction I protein was established by comparing a two-dimensional tryptic peptide map of its [S35] methionine-labelled peptides with a tryptic peptide map of the large subunit of Fraction I protein labelled in vivo with [S35] methionine. It may therefore be concluded that only one of the many proteins present in the soluble phase of the chloroplast, namely the large subunit of Fraction 1 protein, is synthesised on chloroplast ribosomes

    Identification of genes differentially expressed as result of adenovirus type 5- and adenovirus type 12-transformation

    Background: Cells transformed by human adenoviruses (Ad) exhibit differential capacities to induce tumours in immunocompetent rodents; for example, Ad12-transformed rodent cells are oncogenic whereas Ad5-transformed cells are not. The E1A gene determines oncogenic phenotype, is a transcriptional regulator and dysregulates host cell gene expression, a key factor in both cellular transformation and oncogenesis. To reveal differences in gene expression between cells transformed with oncogenic and non-oncogenic adenoviruses we have performed comparative analysis of transcript profiles with the aim of identifying candidate genes involved in the process of neoplastic transformation. Results: Analysis of microarray data revealed that a total of 232 genes were differentially expressed in Ad12 E1- or Ad5 E1-transformed BRK cells compared to untransformed baby rat kidney (BRK) cells. Gene information was available for 193 transcripts and using gene ontology (GO) classifications and literature searches it was possible to assign known or suggested functions to 166 of these identified genes. A subset of differentially-expressed genes from the microarray was further examined by real-time PCR and Western blotting using BRK cells immortalised by Ad12 E1A or Ad5 E1A in addition to Ad12 E1- or Ad5 E1-transformed BRK cells. Up-regulation of RelA and significant dysregulation of collagen type I mRNA transcripts and proteins were found in Ad-transformed cells. Conclusion: These results suggest that a complex web of cellular pathways become altered in Ad-transformed cells and that Ad E1A is sufficient for the observed dysregulation. Further work will focus on investigating which splice variant of Ad E1A is responsible for the observed dysregulation at the pathway level, and the mechanisms of E1A-mediated transcriptional regulation

    Salivary cortisol and cognitive development in infants from low-income communities

    Early stress exposure is proposed to have significant lasting effects on cognitive development. The glucocorticoid hormone cortisol, a product of the hypothalamic-pituitary-adrenal (HPA) axis, is a particular focus of research, however, the majority of past research has been based on studies of older children and adults. Evidence linking cortisol levels in infancy with cognitive development is lacking. In a large cohort sample of infants (N = 1,091) oversampled for psychosocial risk, we tested whether basal cortisol levels and cortisol reactivity to emotional stressors administered at 7 and 15 months of age were associated with cognitive development measured at 15 months. Cognitive development was measured using the Mental Development Index of the Bayley Scales of Infant Development. Multiple regression analyses indicated that basal cortisol levels at 15 months, and to a lesser extent at 7 months, were inversely associated with infant cognitive development after adjusting for psychosocial and obstetric risk. The findings provide some of the first evidence that HPA axis activity in infancy is associated with early cognitive development

    We have directly observed broadband thermal noise in silica/tantala coatings in a high-sensitivity Fabry-Perot interferometer.     Protein crystals in adenovirus type 5-infected cells: requirements for intranuclear crystallogenesis, structural and functional analysis

    Intranuclear crystalline inclusions have been observed in the nucleus of epithelial cells infected with Adenovirus serotype 5 (Ad5) at late steps of the virus life cycle. Using immuno-electron microscopy and confocal microscopy of cells infected with various Ad5 recombinants modified in their penton base or fiber domains, we found that these inclusions represented crystals of penton capsomers, the heteromeric capsid protein formed of penton base and fiber subunits. The occurrence of protein crystals within the nucleus of infected cells required the integrity of the fiber knob and part of the shaft domain. In the knob domain, the region overlapping residues 489–492 in the FG loop was found to be essential for crystal formation. In the shaft, a large deletion of repeats 4 to 16 had no detrimental effect on crystal inclusions, whereas deletion of repeats 8 to 21 abolished crystal formation without altering the level of fiber protein expression. This suggested a crucial role of the five penultimate repeats in the crystallisation process. Chimeric pentons made of Ad5 penton base and fiber domains from different serotypes were analyzed with respect to crystal formation. No crystal was found when fiber consisted of shaft (S) from Ad5 and knob (K) from Ad3 (heterotypic S5-K3 fiber), but occurred with homotypic S3K3 fiber. However, less regular crystals were observed with homotypic S35-K35 fiber. TB5, a monoclonal antibody directed against the Ad5 fiber knob was found by immunofluorescence microscopy to react with high efficiency with the intranuclear protein crystals in situ. Data obtained with Ad fiber mutants indicated that the absence of crystalline inclusions correlated with a lower infectivity and/or lower yields of virus progeny, suggesting that the protein crystals might be involved in virion assembly. Thus, we propose that TB5 staining of Ad-infected 293 cells can be used as a prognostic assay for the viability and productivity of fiber-modified Ad5 vectors

Published in this context, Ernst Freund's little-known The Legal Nature of Corporations (1897) was an original attempt to come to terms with a new legal and economic reality. But it can also be described, to paraphrase Oliver Wendell Holmes, as the earliest example of the rational study of corporate law. The paper shows that Freund had the intuitions of an institutional economist, and engaged in what today would be called comparative institutional analysis. Remarkably, his argument that the corporate form secures property against insider defection and against outsiders anticipated recent work on entity shielding and capital lock-in, and can be read as an early contribution to what today would be called the theory of the firm.Peer reviewe

    The cellular chloride channels CLIC1 and CLIC4 contribute to virus-mediated cell motility

    Ion channels regulate many aspects of cell physiology, including cell proliferation, motility, and migration, and aberrant expression and activity of ion channels is associated with various stages of tumor development, with K⁺ and Cl⁻ channels now being considered the most active during tumorigenesis. Accordingly, emerging in vitro and preclinical studies have revealed that pharmacological manipulation of ion channel activity offers protection against several cancers. Merkel cell polyomavirus (MCPyV) is a major cause of Merkel cell carcinoma (MCC), primarily because of the expression of two early regulatory proteins termed small and large tumor antigens (ST and LT, respectively). Several molecular mechanisms have been attributed to MCPyV-mediated cancer formation but, thus far, no studies have investigated any potential link to cellular ion channels. Here we demonstrate that Cl⁻ channel modulation can reduce MCPyV ST-induced cell motility and invasiveness. Proteomic analysis revealed that MCPyV ST up-regulates two Cl⁻ channels, CLIC1 and CLIC4, which when silenced, inhibit MCPyV ST-induced motility and invasiveness, implicating their function as critical to MCPyV-induced metastatic processes. Consistent with these data, we confirmed that CLIC1 and CLIC4 are up-regulated in primary MCPyV-positive MCC patient samples. We therefore, for the first time, implicate cellular ion channels as a key host cell factor contributing to virus-mediated cellular transformation. Given the intense interest in ion channel modulating drugs for human disease. This highlights CLIC1 and CLIC4 activity as potential targets for MCPyV-induced MCC

    Gravitational waves (GWs) are expected to play a crucial role in the development of multimessenger astrophysics. The combination of GW observations with other astrophysical triggers, such as from gamma-ray and X-ray satellites, optical/radio telescopes, and neutrino detectors allows us to decipher science that would otherwise be inaccessible. In this paper, we provide a broad review from the multimessenger perspective of the science reach offered by the third generation interferometric GW detectors and by the Einstein Telescope (ET) in particular.
