    Spectral-Spatial Hyperspectral Image Classification Using Subspace-Based Support Vector Machines and Adaptive Markov Random Fields

    This paper introduces a new supervised classification method for hyperspectral images that combines spectral and spatial information. A support vector machine (SVM) classifier, integrated with a subspace projection method to address the problems of mixed pixels and noise, is first used to model the posterior distributions of the classes based on the spectral information. Then, the spatial information of the image pixels is modeled using an adaptive Markov random field (MRF) method. Finally, the maximum posterior probability classification is computed via the simulated annealing (SA) optimization algorithm. The combination of subspace-based SVMs and adaptive MRFs is the main contribution of this paper. The resulting methods, called SVMsub-eMRF and SVMsub-aMRF, were experimentally validated using two typical real hyperspectral data sets. The obtained results indicate that the proposed methods demonstrate superior performance compared with other classical hyperspectral image classification methods.Ritrýnt tímaritPeer Reviewe

    We present the 158 standard stars that define the u'g'r'i'z' photometric system. These stars form the basis for the photometric calibration of the Sloan Digital Sky Survey (SDSS). The defining instrument system and filters, the observing process, the reduction techniques, and the software used to create the stellar network are all described.     The Molecular Chaperone HSP90 Promotes Notch Signaling in the Germline of Caenorhabditis elegans

    In a genetic screen to identify genes that promote GLP-1/Notch signaling in Caenorhabditis elegans germline stem cells, we found a single mutation, om40, defining a gene called ego-3. ego-3(om40) causes several defects in the soma and the germline, including paralysis during larval development, sterility, delayed proliferation of germline stem cells, and ectopic germline stem cell proliferation. Whole genome sequencing identified om40 as an allele of hsp-90, previously known as daf-21, which encodes the C. elegans ortholog of the cytosolic form of HSP90. This protein is a molecular chaperone with a central position in the protein homeostasis network, which is responsible for proper folding, structural maintenance, and degradation of proteins. In addition to its essential role in cellular function, HSP90 plays an important role in stem cell maintenance and renewal. Complementation analysis using a deletion allele of hsp-90 confirmed that ego-3 is the same gene. hsp-90(om40) is an IN conservative missense mutation of a highly conserved residue in the middle domain of HSP-90. RNA interference-mediated knockdown of hsp-90 expression partially phenocopied hsp-90(om40), confirming the loss-of-function nature ofhsp-90(om40). Furthermore, reduced HSP-90 activity enhanced the effect of reduced function of both the GLP-1 receptor and thedownstream LAG-1 transcription factor. Taken together, our results provide the first experimental evidence of an essential role for HSP90 inNotch signaling in development

    Gradient microfluidics enables rapid bacterial growth inhibition testing

    Bacterial growth inhibition tests have become a standard measure of the adverse effects of inhibitors for a wide range of applications, such as toxicity testing in the medical and environmental sciences. However, conventional well-plate formats for these tests are laborious and provide limited information (often being restricted to an end-point assay). In this study, we have developed a microfluidic system that enables fast quantification of the effect of an inhibitor on bacteria growth and survival, within a single experiment. This format offers a unique combination of advantages, including long-term continuous flow culture, generation of concentration gradients, and single cell morphology tracking. Using Escherichia coli and the inhibitor amoxicillin as one model system, we show excellent agreement between an on-chip single cell-based assay and conventional methods to obtain quantitative measures of antibiotic inhibition (for example, minimum inhibition concentration). Furthermore, we show that our methods can provide additional information, over and above that of the standard well-plate assay, including kinetic information on growth inhibition and measurements of bacterial morphological dynamics over a wide range of inhibitor concentrations. Finally, using a second model system, we show that this chip-based systems does not require the bacteria to be labeled and is well suited for the study of naturally occurring species. We illustrate this using Nitrosomonas europaea, an environmentally important bacteria, and show that the chip system can lead to a significant reduction in the period required for growth and inhibition measurements (<4 days, compared to weeks in a culture flask)

    Hard X-ray nano-focusing with Montel mirror optics

    a b s t r a c t Kirkpatrick-Baez mirrors in the Montel (or nested) configuration were tested for hard X-ray nanoscale focusing at a third generation synchrotron beamline. In this scheme, two mirrors, mounted side-by-side and perpendicular to each other, provide for a more compact focusing system and a much higher demagnification and flux than the traditional sequential K-B mirror arrangement. They can accept up to a 120 mm  120 mm incident X-ray beam with a long working distance of 40 mm and broad-bandpass of energies up to $ 30 keV. Initial test demonstrated a focal spot of about 150 nm in both horizontal and vertical directions with either polychromatic or monochromatic beam. Montel mirror optics is important and very appealing for achromatic X-ray nanoscale focusing in conventional non-extra-long synchrotron beamlines
