    AYURVEDA MANAGEMENT OF DYSFUNCTIONAL UTERINE BLEEDING - A CASE REPORT

    A 41yrs. female patient came to our clinic outpatient department Gune Ayurved College, Ahmednagar, Maharashtra with complaints of excessive vaginal bleeding. The case was diagnosed as Adhoga Raktapitta by Ayurvedic nidan, symptomatic relief by Stambhana dravyas was not ultimate solution to avoid hysterectomy. This case was successfully treated on basic principles of Ayurveda Panchbhautic chikitsa. A symptom free follow up in this case was noticed after completion of 8 months Ayurved therapy. The treatment options offered here primary aim at reducing Kshardharmiushnata in the body with Parthivdravya and increasing uterine tone (Dharana).Dysfunctional Uterine Bleeding is defined as a state of abnormal uterine bleeding without any clinically detectable organic, systemic & iatrogenic cause. Heavy Bleeding interferes with womens physical emotional, social & material quality & life. Currently DUB is defined as a state of abnormal uterine bleeding following anovulation due to dysfunction & hypothalamo-pituitary-ovarian axis endocrine origin. It is more prevalent in premenopausal & abortion. Overall 70%-80% results of endometrial ablation or resection amongst 10% of the patients need hysterectomy. In present case study we are presenting here with more than 2 yrs. history of Dysfunctional Uterine Bleeding without any response from modern medicine.

    Development and Validation of Stability Indicating RP-HPLC Method for Estimation of Rilpivirine Hydrochloride in Tablet Dosage Form

    A simple, sensitive, rapid and reproducible HPLC Method was developed and validated for estimation of Rilpivirine in the presence of degradation products generated from forced decomposition studies. The analysis was carried out on Hypersil BDS C18, 250 X 4.6mm, 5? column using a mixture of ammonium acetate Buffer (pH to 6.0 0.05) and Acetonitrile in the proportion 55:45 respectively as a mobile phase at a flow rate of 1.2 mL/minute. The wavelength selected for the analysis was 300 nm. The peak for Rilpivirine HCl was observed at 10.33 minute. A linear response was observed in the range of 12.5 - 62.5 ?g/mL with a correlation coefficient of 0.999. The method was validated for specificity, linearity, precision, accuracy and robustness. The obtained results were indicating that the method is selective in analysis of Rilpivirine in the presence of degradation products formed under various stress conditions

    DEVELOPMENT AND VALIDATION OF UV SPECTROPHOTOMETRIC METHOD FOR THE ESTIMATION OF ELVITEGRAVIR (EVG) IN BULK AND PHARMACEUTICAL FORMULATIONS

    ABSTRACT A simple, rapid, precise and economical spectrophotometric method has been developed for quantitative analysis of Elvitegravir (EVG) in manufactured tablet formulation. The stock solution and subsequent dilution of Elvitegravir was done in methanol. The standard solution of Elvitegravir in methanol showed absorption maxima at 313.00 nm. The drug obeyed Beers-Lamberts Law in the concentration range of 1-100 μg/mL with coefficient of correlation (R 2 ) was 0.999 . The method can be adopted in routine analysis of Elvitegravir in bulk and tablet dosage form and it involves relatively low cost solvents and no complex extraction technique

    In situ radiographic and ex situ tomographic analysis of pore interactions during multilayer builds in laser powder bed fusion

    Porosity and high surface roughness can be detrimental to the mechanical performance of laser powder bed fusion (LPBF) additive manufactured components, potentially resulting in reduced component life. However, the link between powder layer thickness on pore formation and surface undulations in the LPBF parts remains unclear. In this paper, the influence of processing parameters on Ti-6Al-4 V additive manufactured thin-wall components are investigated for multilayer builds, using a custom-built process replicator and in situ high-speed synchrotron X-ray imaging. In addition to the formation of initial keyhole pores, the results reveal three pore phenomena in multilayer builds resulting from keyhole melting: (i) healing of the previous layers' pores via liquid filling during remelting; (ii) insufficient laser penetration depth to remelt and heal pores; and (iii) pores formed by keyholing which merge with existing pores, increasing the pore size. The results also show that the variation of powder layer thickness influences which pore formation mechanisms take place in multilayer builds. High-resolution microcomputed tomography images reveal that clusters of pores form at the ends of tracks, and variations in the layer thickness and melt flow cause irregular remelting and track height undulations. Extreme variations in height were found to lead to lack of fusion pores in the trough regions. It is hypothesised that the end of track pores were augmented by soluble gas which is partitioned into the melt pool and swept to track ends, supersaturating during end of track solidification and diffusing into pores increasing their size

    Simple, sensitive, precise, and specific high-performance liquid chromategraphic (HPLC) and high-performance thin-layer chromatographic (HPTLC) methods for the determination of dolutegravir sodium in bulk drug and pharmaceutical dosage form were developed and validated. In the HPLC method, analysis of the drug was carried out on the ODS C18 column (150 × 4.6 mm, 5 μm particle size) using a mixture of acetonitrile: water (pH 7.5) in the ratio of 80:20 v/v as the mobile phase at the flow rate 1 mL/min at 260 nm. This method was found to be linear in the concentration range of 5-35 μg/mL. The peak for dolutegravir sodium was observed at 3.0 ± 0.1 minutes. In the HPTLC method, analysis was performed on aluminum-backed plates pre-coated with silica gel G60 F254 using methanol: chloroform: formic acid in the proportion of 8:2:0.5 v/v/v as the mobile phase. This solvent system was found to give compact spots for dolutegravir sodium with the Rf value 0.77 ± 0.01. Densitometric analysis of dolutegravir sodium was carried out in the absorbance mode at 265 nm. Linear regression analysis showed good linearity with respect to peak area in the concentration range of 200-900 ng/spot. The methods were validated for precision, limit of detection (LOD), limit of quantitation (LOQ), accuracy, and specificity. Statistical analysis showed that both of the methods are repeatable and specific for the estimation of the said drug. The methods can be used for routine quality control analysis of dolutegravir sodium

    A simple, accurate and precise HPTLC method has been developed for estimation of propranolol hydrochloride from bulk drug and tablet formulations. The separation was achieved on TLC plates using appropriate solvent system. The spots so developed were densitometrically scanned at 290 nm. The linearity of the method was found to be within the concentration range of 200–2000 ng/spot. The validation parameters, tested in accordance with the requirements of ICH guidelines, prove the suitability of this method. The method was successively applied for determination of drug in tablets, wherein, no interference from tablet excipients was observed
