    A control volume based formulation of the discrete Kirchoff triangular thin plate bending element

    A control volume method is presented for predicting the displacement and rotation of thin transversely loaded flat plates. The new procedure uses discrete Kirchoff triangle (DKT) elements but introduces a dual mesh of interconnected control volumes (CVs) centred on the finite element (FE) vertices. Discrete equations for the unknown degrees of freedom are subsequently derived by enforcing equilibrium on these CVs; as such this implementation is a quadrature free routine. To allow a comparison, a quadrature free implementation of the DKT element, using the standard finite element procedure, was developed using symbolic methematics. The CV based procedure is validated by patch tests for a state of pure bending and twist. Convergence tests for various loading types show enhanced performance for coarse meshes over the equivalent FE method

    Genetic islands of Streptococcus agalactiae strains NEM316 and 2603VR and their presence in other Group B Streptococcal strains

    BACKGROUND: Streptococcus agalactiae (Group B Streptococcus; GBS) is a major contributor to obstetric and neonatal bacterial sepsis. Serotype III strains cause the majority of late-onset sepsis and meningitis in babies, and thus appear to have an enhanced invasive capacity compared with the other serotypes that cause disease predominantly in immunocompromised pregnant women. We compared the serotype III and V whole genome sequences, strains NEM316 and 2603VR respectively, in an attempt to identify genetic attributes of strain NEM316 that might explain the propensity of strain NEM316 to cause late-onset disease in babies. Fourteen putative pathogenicity islands were described in the strain NEM316 whole genome sequence. Using PCR- and targeted microarray- strategies, the presence of these islands were assessed in a diverse strain collection including 18 colonizing isolates from healthy pregnant women, and 13 and 8 invasive isolates from infants with early- and late-onset sepsis, respectively. RESULTS: Side-by-side comparison of the strain NEM316 and strain 2603VR genomes revealed that they are extremely similar, with the only major difference being the capsulation loci and mobile genetic elements. PCR and Comparative Genome Hybridization (CGH) were used to define the presence of each island in 39 GBS isolates. Only islands I, VI, XII, and possibly X, met criteria of a true pathogenicity island, but no significant correlation was found between the presence of any of the fourteen islands and whether the strains were invasive or colonizing. Possible associations were seen between the presence of island VI and late-onset sepsis, and island X and early-onset sepsis, which warrant further investigation. CONCLUSION: The NEM316 and 2603VR strains are remarkable in that their whole genome sequences are so similar, suggesting that the capsulation loci or other genetic differences, such as pathogenicity islands, are the main determinants of the propensity of serotype III strains to cause late-onset disease. This study supports the notion that GBS strain NEM316 has four putative pathogenicity islands, but none is absolutely necessary for disease causation, whether early- or late-onset sepsis. Mobile genetic elements are a common feature of GBS isolates, with each strain having its own peculiar burden of transposons, phages, integrases and integrated plasmids. The majority of these are unlikely to influence the disease capacity of an isolate. Serotype associated disease phenotypes may thus be solely related to differences in the capsulation loci

    Cryogenic and room temperature strength of sapphire jointed by hydroxide-catalysis bonding

    Hydroxide-catalysis bonding is a precision technique used for jointing components in opto-mechanical systems and has been implemented in the construction of quasi-monolithic silica suspensions in gravitational wave detectors. Future detectors are likely to operate at cryogenic temperatures which will lead to a change in test mass and suspension material. One candidate material is mono-crystalline sapphire. Here results are presented showing the influence of various bonding solutions on the strength of the hydroxide-catalysis bonds formed between sapphire samples, measured both at room temperature and at 77 K, and it is demonstrated that sodium silicate solution is the most promising in terms of strength, producing bonds with a mean strength of 63 MPa. In addition the results show that the strengths of bonds were undiminished when tested at cryogenic temperatures

    MHD Wave Propagation in the Neighbourhood of Two Null Points

    The nature of fast magnetoacoustic and Alfv\'en waves is investigated in a zero Ī²\beta plasma in the neighbourhood of a pair of two-dimensional null points. This gives an indication of wave propagation in the low Ī²\beta solar corona, for a more complicated magnetic configuration than that looked at by McLaughlin & Hood (2004). It is found that the fast wave is attracted to the null points and that the front of the wave slows down as it approaches the null point pair, with the wave splitting and part of the wave accumulating at one null and the rest at the other. Current density will then accumulate at these points and ohmic dissipation will then extract the energy in the wave at these points. This suggests locations where wave heating will occur in the corona. The Alfv\'en wave behaves in a different manner in that the wave accumulates along the separatrices. Hence, the current density will accumulate at this part of the topology and this is where wave heating will occur. However, the phenomenon of wave accumulation at a specific place is a feature of both wave types, and illustrates the importance of studying the topology of the corona when considering MHD wave propagation.Comment: 11 pages, 14 figure

    Safety and immunogenicity of a new tuberculosis vaccine, MVA85A, in mycobacterium tuberculosisā€“infected individuals

    Copyright Ā© 2009 by the American Thoracic Society.Rationale: An effective new tuberculosis (TB) vaccine regimen must be safe in individuals with latent TB infection (LTBI) and is a priority for global health care. Objectives: To evaluate the safety and immunogenicity of a leading new TB vaccine, recombinant Modified Vaccinia Ankara expressing Antigen 85A (MVA85A) in individuals with LTBI. Methods: An open-label, phase I trial of MVA85A was performed in 12 subjects with LTBI recruited from TB contact clinics in Oxford and London or by poster advertisements in Oxford hospitals. Patients were assessed clinically and had blood samples drawn for immunological analysis over a 52-week period after vaccination with MVA85A. Thoracic computed tomography scans were performed at baseline and at 10 weeks after vaccination. Safety of MVA85A was assessed by clinical, radiological, and inflammatory markers. The immunogenicity of MVA85A was assessed by IFNĪ³ and IL-2 ELISpot assays and FACS. Measurements and Main Results: MVA85A was safe in subjects with LTBI, with comparable adverse events to previous trials of MVA85A. There were no clinically significant changes in inflammatory markers or thoracic computed tomography scans after vaccination. MVA85A induced a strong antigen-specific IFN-Ī³ and IL-2 response that was durable for 52 weeks. The magnitude of IFN-Ī³ response was comparable to previous trials of MVA85A in bacillus Calmette-GuĆ©rinā€“vaccinated individuals. Antigen 85Aā€“specific polyfunctional CD4+ T cells were detectable prior to vaccination with statistically significant increases in cell numbers after vaccination. Conclusions: MVA85A is safe and highly immunogenic in individuals with LTBI.     Muon Catalyzed Fusion in 3 K Solid Deuterium

    Muon catalyzed fusion in deuterium has traditionally been studied in gaseous and liquid targets. The TRIUMF solid-hydrogen-layer target system has been used to study the fusion reaction rates in the solid phase of D_2 at a target temperature of 3 K. Products of two distinct branches of the reaction were observed; neutrons by a liquid organic scintillator, and protons by a silicon detector located inside the target system. The effective molecular formation rate from the upper hyperfine state of Ī¼d\mu d and the hyperfine transition rate have been measured: Ī»~(3/2)=2.71(7)stat.(32)syst.Ī¼/s\tilde{\lambda}_(3/2)=2.71(7)_{stat.}(32)_{syst.} \mu/s, and Ī»~(3/2)(1/2)=34.2(8)stat.(1)syst.Ī¼/s\tilde{\lambda}_{(3/2)(1/2)} =34.2(8)_{stat.}(1)_{syst.} \mu /s. The molecular formation rate is consistent with other recent measurements, but not with the theory for isolated molecules. The discrepancy may be due to incomplete thermalization, an effect which was investigated by Monte Carlo calculations.
