    Heterozygous mis-sense mutations in Prkcb as a critical determinant of anti-polysaccharide antibody formation

    To identify rate-limiting steps in T cell-independent type 2 (TI-2) antibody production against polysaccharide antigens, we performed a genome-wide screen by immunizing several hundred pedigrees of C57BL/6 mice segregating ENU-induced mis-sense mutations. Two independent mutations, Tilcara and Untied, were isolated that semi-dominantly diminished antibody against polysaccharide but not protein antigens. Both mutations resulted from single amino acid substitutions within the kinase domain of Protein Kinase C Beta (PKCĪ²). In Tilcara, a Ser552>Pro mutation occurred in helix G, in close proximity to a docking site for the inhibitory N-terminal pseudosubstrate domain of the enzyme, resulting in almost complete loss of active, autophosphorylated PKCĪ²I whereas the amount of alternatively spliced PKCĪ²II protein was not markedly reduced. Circulating B cell subsets were normal and acute responses to BCR-stimulation such as CD25 induction and initiation of DNA synthesis were only measurably diminished in Tilcara homozygotes, whereas the fraction of cells that had divided multiple times was decreased to an intermediate degree in heterozygotes. These results, coupled with evidence of numerous mis-sense PRKCB mutations in the human genome, identify Prkcb as a genetically sensitive step likely to contribute substantially to population variability in anti-polysaccharide antibody levels

    Spectral fingerprinting for specific algal groups on sediments in situ: a new sensor

    Currently it is still extremely difficult to adequately sample populations of microalgae on sediments for large-scale biomass determination. We have now devised a prototype of a new benthic sensor (BenthoFluor) for the quantitative and qualitative assessment of microphytobenthos populations in situ. This sensor enables a high spatial and temporal resolution and a rapid evaluation of the community structure and distribution. These determinations are based on the concept that five spectral excitation ranges can be used to differentiate groups of microalgae, in situ, within a few seconds. In addition, because sediments contain a lot of yellow substances, which can affect the fluorescence and optical differentiation of the algae, the device was equipped with a UV-LED for yellow substances correction. The device was calibrated against HPLC with cultures and tested in the field. Our real-time approach can be used to monitor algal assemblage composition on sediments and is an ideal tool for investigations on the large-scale spatial and temporal variation of algal populations in sediments. Apart from the differentiation of algal populations, the BenthoFluor allows instantaneous monitoring of the chlorophyll concentrations and determination of which algae are responsible for this on the uppermost surface of sediments in the field and in experimental set-ups

    Validating ocean tide loading models using GPS

    Abstract.: Ocean tides cause periodic deformations of the Earth's surface, also referred to as ocean tide loading (OTL). Tide-induced displacements of the Earth's crust relying on OTL models are usually taken into account in GPS (Global Positioning System) data analyses. On the other hand, it is also possible to validate OTL models using GPS analyses. The following simple approach is used to validate OTL models. Based on a particular model, instantaneous corrections of the site coordinates due to OTL are computed. Site-specific scale factors, f, for these corrections are estimated in a standard least-squares adjustment process of GPS observations together with other relevant parameters. A resulting value of f close to unity indicates a good agreement of the model with the actual site displacements. Such scale factors are computed for about 140 globally distributed IGS (International GPS Service) tracking sites. Three OTL models derived from the ocean tide models FES95.2.1, FES99, and GOT00.2 are analyzed. As expected, the most reliable factors are estimated for sites with a large loading effect. In general, the scaling factors have a value close to unity and no significant differences between the three ocean tide models could be observed. It is found that the validation approach is easy to apply. Without requiring much additional effort for a global and self-consistent GPS data analysis, it allows detection of general model misfits on the basis of a large number of globally distributed sites. For detailed validation studies on OTL models, the simultaneous estimation of amplitudes and phases for the main contributing partial tides within a GPS parameter adjustment process would provide more detailed answer

    Evaluation of HCMM data for assessing soil moisture and water table depth

    Soil moisture in the 0-cm to 4-cm layer could be estimated with 1-mm soil temperatures throughout the growing season of a rainfed barley crop in eastern South Dakota. Empirical equations were developed to reduce the effect of canopy cover when radiometrically estimating the soil temperature. Corrective equations were applied to an aircraft simulation of HCMM data for a diversity of crop types and land cover conditions to estimate the soil moisture. The average difference between observed and measured soil moisture was 1.6% of field capacity. Shallow alluvial aquifers were located with HCMM predawn data. After correcting the data for vegetation differences, equations were developed for predicting water table depths within the aquifer. A finite difference code simulating soil moisture and soil temperature shows that soils with different moisture profiles differed in soil temperatures in a well defined functional manner. A significant surface thermal anomaly was found to be associated with shallow water tables

Soil moisture in the 0-4 cm layer could be estimated with 1-mm soil temperatures throughout the growing season of a rainfed barley crop (% cover ranging from 30% to 90%) with an r squared = 0.81. Empirical equations were developed to reduce the effect of canopy cover when radiometrically estimating the 1-mm soil temperature, r squared = 0.88. The corrective equations were applied to an aircraft simulation of HCMM data for a diversity of crop types and land cover conditions to estimate the 0-4 cm soil moisture. The average difference between observed and measured soil moisture was 1.6% of field capacity. HCMM data were used to estimate the soil moisture for four dates with an r squared = 0.55 after correction for crop conditions. Location of shallow alluvial aquifers could be accomplished with HCMM predawn data. After correction of HCMM day data for vegetation differences, equations were developed for predicting water table depths within the aquifer (r=0.8)

    Variation of Area-to-Mass-Ratio of HAMR Space Debris Objects

    An unexpected space debris population has been detected in 2004 Schildknecht et al. (2003, 2004) with the unique properties of a very high area-to-mass ratio (HAMR) Schildknecht et al. (2005a). Ever since it has been tried to investigate the dynamical properties of those objects further. The orbits of those objects are heavily perturbed by the effect of direct radiation pressure. Unknown attitude motion complicates orbit prediction. The area-to-mass ratio of the objects seems to be not stable over time. Only sparse optical data is available for those objects in drift orbits. The current work uses optical observations of five HAMR objects, observed over several years and investigates the variation of their area-to-mass ratio and orbital parameters.
