    Preparing for Employment: On the Home Front

    Work-based learning during the school years leads to better postschool employment outcomes (Hughes, Moore, & Bailey, 1999). Volunteer experiences and unpaid internships, in addition to paid employment, can be steppingstones to future employment. Youth and their families need not rely solely on school programs to pursue such opportunities. They can do much on their own to launch the youth's career search. Recent studies demonstrate the effectiveness of using personal networks as a job search strategy (Timmons, Hamner, & Boes, 2003), and highlight the fact that families make key contributions to successful employment outcomes for individuals with disabilities (26th Institute on Rehabilitation Issues, 2000).There are creative ways to combine community relationships, a young person's interests, and family or personal networks to help a young person effectively explore work-based learning outside of school settings. Parents may seek opportunities through co-workers, relatives, and neighbors. Moreover, parents often know their children better than professionals do and can help their sons and daughters explore their unique abilities, strengths, and interests -- all of which may lead to an appropriate career path

    Replicating capacity and congestion in microscale agent-based simulations

    Disaster events cause detrimental impacts for communities across the globe, ranging from large numbers of fatalities and injuries, to the loss of homes and devastating financial impacts. Emergency professionals are facedwith the challenge of providing sustainable solutions to mitigate these consequences and require tools to aid the assessment of potential impacts. Current modelling tools have either focused on modelling either the microscale (e.g. individual confined spaces such as buildings or stadiums) or the macroscale (e.g. city scale). The aim of thisresearch is to create microscale agent-based modelling (ABM) tools, incorporating a realistic representation of human behaviours, which will help management professionals assess and improve their contingency plans for emergency scenarios. The focus has been on creating a microscale agent-based model of a pedestrian pavement and crossroads, to include overtaking and giving way, alongside the inclusion of varied population characteristics. This research has found that by improving pedestrian interactions (e.g. overtaking and giving way interactions) on pavements and at crossroads more robust travel time estimates can be achieved. To produce more realistic behaviour traits, microscale models should consider: (1) varied walking speed (2) population density, (3) patience level and (4) an exit split percentage for crossroads. Comparisons to 1.34 m/s (3mph) models without additional variables show the travel times may be misrepresentative by up to 78% in pavements and 305% in crossroads for some population types. This has the potential to cause cascading effects such as a significant increase in fatalities or injuries as communities cannot reach safety in the anticipated time

    Cardiac and Skeletal Muscle Lipotoxicity in a Rat Model of Pulmonary Arterial Hypertension

    poster abstractPatients with Pulmonary Arterial Hypertension (PAH)experience shift from aerobic to anaerobic respiration in cardiac and skeletal muscle myocytes. This shift can be identified histologically by an increased presence of the glucose transporter Glut 1 in the tissue, indicating increased reliance on cytoplasmic glycolysis. Previous studies have demonstrated that in patients with diabetes, an increase in Glut 1 is accompanied by an increase in fat storage in the cell. Excessive myocyte fat storage may contribute to tissue and systemic inflammation and has therefore been termed ‘lipotoxicity’. This study tested the hypothesis in a PAH rat model that an increase in cardiac and skeletal muscle Glut 1 abundance would be associated with an increase in fat storage in the cell. Oil Red O staining was performed to assay for lipotoxicity in cryosections of right ventricle and soleus muscle tissue, imaged using brightfield microscopy. The experiment was conducted using tissue from a moderately severe PAH phenotype produced by monocrotaline(60 mg/kg) injection, as well as from saline injected control animals. Lipids have been observed in the first few samples tested with Oil Red O staining, and results are still pending as a larger sample size is currently being collected

    Evaluating the Effect of Kaftrio on Perspectives of Health and Wellbeing in Individuals with Cystic Fibrosis

    Background: Modulator therapy represents a significant step forward in CF care and is expected to have a significant impact on the health and mortality of many individuals with CF. Studies have predominantly explored the physiological effects of modulator therapy on clinical outcomes, with little consideration of the individual lived experience of modulator therapy among adults with Cystic Fibrosis.Methods: To explore this, semi-structured interviews were conducted with 12 individuals currently taking Kaftrio, which were subsequently thematically analysed.Results: Three overarching themes were identified: (i) positive perception of Kaftrio, (ii) negative perception of Kaftrio, and (iii) the relationships with the clinical team. The experience of modulator therapy should be recognised as being unique to the individual, with perceptions of illness, self-identity, and outcomes strongly dictating the lived experience.Conclusions: There is a consensus that, while for many, the quality of life is evidently increased through the use of Kaftrio, this is not without its own challenges. This highlights the need for both individuals with CF and their clinical teams to learn to navigate this new disease landscape

    Expression of the Flp proteins by Haemophilus ducreyi is necessary for virulence in human volunteers

    <p>Abstract</p> <p>Background</p> <p><it>Haemophilus ducreyi</it>, the causative agent of the sexually transmitted disease chancroid, contains a <it>flp </it>(fimbria like protein) operon that encodes proteins predicted to contribute to adherence and pathogenesis. <it>H. ducreyi </it>mutants that lack expression of Flp1 and Flp2 or TadA, which has homology to NTPases of type IV secretion systems, have decreased abilities to attach to and form microcolonies on human foreskin fibroblasts (HFF). A <it>tadA </it>mutant is attenuated in its ability to cause disease in human volunteers and in the temperature dependent rabbit model, but a <it>flp1flp2 </it>mutant is virulent in rabbits. Whether a <it>flp </it>deletion mutant would cause disease in humans is not clear.</p> <p>Results</p> <p>We constructed 35000HPΔ<it>flp1-3</it>, a deletion mutant that lacks expression of all three Flp proteins but has an intact <it>tad </it>secretion system. 35000HPΔ<it>flp1-3 </it>was impaired in its ability to form microcolonies and to attach to HFF in vitro when compared to its parent (35000HP). Complementation of the mutant with <it>flp1-3 </it>in trans restored the parental phenotype. To test whether expression of Flp1-3 was necessary for virulence in humans, ten healthy adult volunteers were experimentally infected with a fixed dose of 35000HP (ranging from 54 to 67 CFU) on one arm and three doses of 35000HPΔ<it>flp1-3 </it>(ranging from 63 to 961 CFU) on the other arm. The overall papule formation rate for the parent was 80% (95% confidence interval, CI, 55.2%-99.9%) and for the mutant was 70.0% (95% CI, 50.5%-89.5%) (<it>P </it>= 0.52). Mutant papules were significantly smaller (mean, 11.2 mm<sup>2</sup>) than were parent papules (21.8 mm<sup>2</sup>) 24 h after inoculation (<it>P </it>= 0.018). The overall pustule formation rates were 46.7% (95% CI 23.7-69.7%) at 30 parent sites and 6.7% (95% CI, 0.1-19.1%) at 30 mutant sites (<it>P </it>= 0.001).</p> <p>Conclusion</p> <p>These data suggest that production and secretion of the Flp proteins contributes to microcolony formation and attachment to HFF cells in vitro. Expression of <it>flp1-3 </it>is also necessary for <it>H. ducreyi </it>to initiate disease and progress to pustule formation in humans. Future studies will focus on how Flp proteins contribute to microcolony formation and attachment in vivo.</p

    Real-time Atomistic Observation of Structural Phase Transformations in Individual Hafnia Nanorods

    High-temperature phases of hafnium dioxide have exceptionally high dielectric constants and large bandgaps, but quenching them to room temperature remains a challenge. Scaling the bulk form to nanocrystals, while successful in stabilizing the tetragonal phase of isomorphous ZrO2, has produced nanorods with a twinned version of the room temperature monoclinic phase in HfO2. Here we use in situ heating in a scanning transmission electron microscope to observe the transformation of an HfO2 nanorod from monoclinic to tetragonal, with a transformation temperature suppressed by over 1000°C from bulk. When the nanorod is annealed, we observe with atomic-scale resolution the transformation from twinned-monoclinic to tetragonal, starting at a twin boundary and propagating via coherent transformation dislocation; the nanorod is reduced to hafnium on cooling. Unlike the bulk displacive transition, nanoscale size-confinement enables us to manipulate the transformation mechanism, and we observe discrete nucleation events and sigmoidal nucleation and growth kinetics

    Timing, Duration, and Pathways of Harlequin Duck Migration to Pacific Molting and Wintering Areas

    The core breeding range for Harlequin Ducks (Histrionicus histrionicus) in western North America extends from Alaska, and south through the Yukon, Northwest Territories, and British Columbia. Smaller breeding populations exist in southwestern Alberta, Washington, Oregon, Idaho, Wyoming, and Montana. Each state and province in these areas has identified the Harlequin Duck as a species of conservation priority, given its small and isolated populations, its specific nesting requirements, and changes in abundance or distribution. Conservation objectives for all areas have identified the importance of mapping migration routes that connect breeding sites to Pacific coast molting and wintering locations, as well as determining migration timing, duration, habitat use, and stopover sites. In spring 2016, we captured Harlequin Duck pairs on breeding streams and surgically implanted satellite transmitters in the males and attached geolocators to the leg bands of females. We marked 18 harlequin pairs (Alberta = 10 (minus one female), Montana = 5, Wyoming = 2, Washington = 1). Migration initiation dates varied by breeding areas and occurred from June 3 to July 10. Male migration lasted between 1-17 days and stopovers occurred approximately half-way to the coast and included rivers, mountain streams, and lakes. They arrived at their molting areas between June 5-July 24 and these areas ranged from southeast Alaska to northwestern Washington. The majority (71%) of harlequins departed molt locations to differing winter locations. Efforts will be made to retrieve the geolocators from females in spring 2017 to compare locations between males and females from different devices
