    International audienceWe describe here the mature spermatozoa of three species of bucephalids, namely Bucephalus margar-itae, Rhipidocotyle khalili and Prosorhynchus longisaccatus. This study provides the first ultrastructural data on the genera Bucephalus and Rhipidocotyle and enabled us to confirm the model of the mature spermatozoon in the Bucephalinae. The spermatozoon exhibits two axonemes with the 9 + ''1'' pattern of the Trepaxonemata, one of which is very short, lateral expansion, external ornamentation of the plasma membrane located in the anterior extremity of the spermatozoon and associated with cortical microtubules, spine-like bodies, a mitochondrion, and a nucleus. The maximum number of cortical microtubules is located in the anterior part of the spermatozoon. However, more studies are needed to elucidate if spine-like bodies are present in all the Bucephalinae or not. In the Prosorhynchinae, the mature spermatozoon exhibits a similar ultrastructural pattern. Some differences are observed, particularly the ax-oneme lengths and the arrangement of the spine-like bodies. The posterior extremity of the spermatozoon in the Bucephalinae exhibits only the nucleus, but prosorhynchines have microtubules.Dans cette étude, nous décrivons les spermatozoïdes mûrs de trois espèces de bucephalidés, à savoir Bucephalus margaritae, Rhipidocotyle khalili et Prosorhynchus longisaccatus. Ces premières données ultrastructurales pour les genres Bucephalus et Rhipidocotyle nous ont permis de confirmer le modèle du spermatozoïde chez les Bucephalinae. Le spermatozoïde comprend deux axonèmes de type 9 + ‘‘1’’ des Trepaxonemata, l’un d’entre eux étant très court, une expansion latérale, une ornementation externe de la membrane plasmique située au niveau de l’extrémité antérieure du spermatozoïde et associée avec des microtubules corticaux, des corps en forme d’épines, une mitochondrie et un noyau. Le nombremaximum de microtubules corticaux est situé dans la partie antérieure du spermatozoïde. Toutefois, des études supplémentaires seraient nécessaires pour vérifier si les corps en forme d’épines sont présents chez tous les Bucephalinae. Chez les Prosorhynchinae, le spermatozoïde mûr présente une structure similaire. Quelques différences sont toutefois observées, notamment au niveau de la longueur des axonèmes et de la disposition des corps en forme d’épines.     Compensating for past deforestation:assessing the legal forest surplus and deficit of the state of Pará, eastern Amazonia

    Up to 80% of each private rural property in the Brazilian Amazon is protected by law through the Legal Reserve (LR) mechanism of the federal Forest Code, underlining the conservation importance of forests on private lands in one of the world́s most important biomes. However, our understanding of the discrepancies in levels of forest protection on private lands as obligated by the law versus what occurs in practice remains very poor. We assessed patterns of forest cover and legal compliance with the Forest Code in the 1.25 Mkm2 Brazilian state of Pará, which has the highest deforestation rate in the Amazon. We evaluate the LR deficit and surplus patterns for different sized properties and across 144 municipalities, and found that the total LR surplus (12.6 Mha) was more than five times the total area of deficit (2.3 Mha). Yet, from the total surplus, only 11% can be legally deforested while the remaining 89% is already protected by law but can be used (sold or rented) to compensate for areas that are under deficit. Medium and large-scale properties make up most of the total LR deficit area, while agrarian reform settlements had comparatively large amounts of both compensation-only surplus and deforestable surplus. Most of the municipalities (77%) in the state could compensate their total deficit with surplus areas of LR in the same municipality, while the remainder can be compensate their deficit in one or more neighbouring municipalities, indicating compensation can always take place close to the source of the deficit. Maximising the environmental benefits of achieving Forest Code compliance requires measures that go beyond the existing legal framework, including interventions to avoid further deforestation in places where it is still legal, compensate in close proximity to areas with legal reserve deficit and promote local restoration on degraded lands

    Radiopharmaceutical Validation for Clinical Use

    Radiopharmaceuticals are reemerging as attractive anticancer agents, but there are no universally adopted guidelines or standardized procedures for evaluating agent validity before early-phase trial implementation. To validate a radiopharmaceutical, it is desirous for the radiopharmaceutical to be specific, selective, and deliverable against tumors of a given, molecularly defined cancer for which it is intended to treat. In this article, we discuss four levels of evidence—target antigen immunohistochemistry, in vitro and in vivo preclinical experiments, animal biodistribution and dosimetry studies, and first-in-human microdose biodistribution studies—that might be used to justify oncology therapeutic radiopharmaceuticals in a drug-development sequence involving early-phase trials. We discuss common practices for validating radiopharmaceuticals for clinical use, everyday pitfalls, and commonplace operationalizing steps for radiopharmaceutical early-phase trials. We anticipate in the near-term that radiopharmaceutical trials will become a larger proportion of the National Cancer Institute Cancer Therapy Evaluation Program (CTEP) portfolio

    It is a well-known fact that laser-induced breakdown spectroscopy (LIBS) has emerged as one of the best analytical techniques for multi-elemental compositional analysis of samples. We report assembling and optimization of LIBS set up using high resolution and broad-range echelle spectrograph coupled to an intensified charge coupled device (ICCD) to detect and quantify trace elements in environmental and clinical samples. Effects of variations of experimental parameters on spectroscopy signals of copper and brass are reported. Preliminary results of some plasma diagnostic calculations using recorded time-resolved optical emission signals are also reported for brass samples

    First spermatological study in the Atractotrematidae (Digenea, Haploporoidea): the case of Atractotrema sigani, intestinal parasite of Siganus lineatus

    The ultrastructural organization of the mature spermatozoon of the digenean Atractotrema sigani(from Siganus lineatus off New Caledonia) was investigated by transmission electron microscopy. The male gamete of A. sigani exhibits the general morphology described in digeneans with the presence of two axonemes of different lengths showing the 9 + '1' pattern of the Trepaxonemata, a nucleus, two mitochondria, two bundles of parallel cortical microtubules, external ornamentation, spine-like bodies and granules of glycogen. However, the mature spermatozoon of A. sigani has some specific characters such as the morphology of its anterior region and the submembranous electron-dense material. Although similar structures have been reported in some digenean species, the presence of a submembranous electron-dense material describing a complete ring is reported here for the first time in the mature spermatozoon of A. sigani. In addition, sperm characteristics are compared between the Haploporoidea and their supposed close superfamilies, and possible phylogenetic implications of these findings for the Digenea are discussed

    Failure to Detect Xenotropic Murine Leukemia Virus-Related Virus in Blood of Individuals at High Risk of Blood-Borne Viral Infections

    A xenotropic murine leukemia virus-related virus (XMRV) has recently been reported in association with prostate cancer and chronic fatigue syndrome, with a prevalence of up to 3.7% in the healthy population. We looked for XMRV in 230 patients with human immunodeficiency virus type 1 or hepatitis C infection. XMRV was undetectable in plasma or peripheral blood mononuclear cells by polymerase chain reaction targeting XMRV gag or env. T cell responses to XMRV Gag were undetectable in peripheral blood mononuclear cells by ex vivo gamma interferon enzyme-linked immunospot assay. In our cohorts, XMRV was not enriched in patients with blood-borne or sexually transmitted infections fromthe United Kingdom and Western Europ

    One-year outcomes of coronary artery bypass graft surgery versus percutaneous coronary intervention with multiple stenting for multisystem disease: A meta-analysis of individual patient data from randomized clinical trials

    BackgroundWe aimed to provide a quantitative analysis of the 1-year clinical outcomes of patients with multisystem coronary artery disease who were included in recent randomized trials of percutaneous coronary intervention with multiple stenting versus coronary artery bypass graft surgery.MethodsAn individual patient database was composed of 4 trials (Arterial Revascularization Therapies Study, Stent or Surgery Trial, Argentine Randomized Trial of Percutaneous Transluminal Coronary Angioplasty Versus Coronary Artery Bypass Surgery in Multivessel Disease 2, and Medicine, Angioplasty, or Surgery Study 2) that compared percutaneous coronary intervention with multiple stenting (N = 1518) versus coronary artery bypass graft surgery (N = 1533). The primary clinical end point of this study was the combined incidence of death, myocardial infarction, and stroke at 1 year after randomization. Secondary combined end points included the incidence of repeat revascularization at 1 year. All analyses were based on the intention-to-treat principle.ResultsAfter 1 year of follow-up, 8.7% of patients randomized to percutaneous coronary intervention with multiple stenting versus 9.1% of patients randomized to coronary artery bypass graft surgery reached the primary clinical end point (hazard ratio 0.95 and 95% confidence interval 0.74’1.2). Repeat revascularization procedures occurred more frequently in patients allocated to percutaneous coronary intervention with multiple stenting compared with coronary artery bypass graft surgery (18% vs 4.4%; hazard ratio 4.4 and 95% confidence interval 3.3’5.9). The percentage of patients who were free from angina was slightly lower after percutaneous coronary intervention with multiple stenting than after coronary artery bypass graft surgery (77% vs 82%; P = .002).ConclusionsOne year after the initial procedure, percutaneous coronary intervention with multiple stenting and coronary artery bypass graft surgery provided a similar degree of protection against death, myocardial infarction, or stroke for patients with multisystem disease. Repeat revascularization procedures remain high after percutaneous coronary intervention, but the difference with coronary artery bypass graft surgery has narrowed in the era of stenting

Conference sessions and discussions reinforced the tangible negative effects of stigma on national legislation and policies. A strong theme throughout the conference was the need to replace prevention interventions that focus exclusively on individual behaviour change or biomedical prevention interventions with "combination prevention" approaches that address both individual and structural factors that increase vulnerability to HIV infection
