    Designing visual management in manufacturing from a user perspective

    Many organisations use daily meetings, whiteboards and an information system for employee intra-communication. While Operation Management research is often management centred, Human Centred Design, instead, takes a user’s perspective. This research aims to reflect upon and describe a method, applied in practice in a double case study within manufacturing, on how to (re-)design meetings and visual management boards, and what type of information and key performance indicators are most relevant for the personnel.     Single nucleotide polymorphisms in the apolipoprotein B and low density lipoprotein receptor genes affect response to antihypertensive treatment

    BACKGROUND: Dyslipidemia has been associated with hypertension. The present study explored if polymorphisms in genes encoding proteins in lipid metabolism could be used as predictors for the individual response to antihypertensive treatment. METHODS: Ten single nucleotide polymorphisms (SNP) in genes related to lipid metabolism were analysed by a microarray based minisequencing system in DNA samples from ninety-seven hypertensive subjects randomised to treatment with either 150 mg of the angiotensin II type 1 receptor blocker irbesartan or 50 mg of the β(1)-adrenergic receptor blocker atenolol for twelve weeks. RESULTS: The reduction in blood pressure was similar in both treatment groups. The SNP C711T in the apolipoprotein B gene was associated with the blood pressure response to irbesartan with an average reduction of 19 mmHg in the individuals carrying the C-allele, but not to atenolol. The C16730T polymorphism in the low density lipoprotein receptor gene predicted the change in systolic blood pressure in the atenolol group with an average reduction of 14 mmHg in the individuals carrying the C-allele. CONCLUSIONS: Polymorphisms in genes encoding proteins in the lipid metabolism are associated with the response to antihypertensive treatment in a drug specific pattern. These results highlight the potential use of pharmacogenetics as a guide for individualised antihypertensive treatment, and also the role of lipids in blood pressure control

    Thoracolumbar and Lumbopelvic Spinal Alignment During the Barbell Back Squat: A Comparison Between Men and Women

    # Background Maintaining neutral spinal alignment is considered important when performing the barbell back squat exercise. Since male and female lifters may differ in injury location it is important to examine whether they differ in spinal alignment during the back squat. # Objectives The study aimed to quantify the spinal alignment in the upper and lower lumbar spine during the barbell back squat exercise in male and female lifters. Secondary aims were to compare alignment during the back squat to standing habitual lumbar spine alignment and determine whether male and female lifters differ in these aspects. # Study Design Observational, Cross-sectional. # Methods Competitive power- and weightlifters were recruited and performed three repetitions of the barbell back squat exercise using a load equivalent to 70% of their one-repetition maximum. Spinal alignment and range of motion were measured using inertial measurement units placed on the thoracic, lumbar and sacral spine. Data was presented descriptively and comparisons between men and women as well as spinal alignment in four different positions were done with a factorial repeated measures analysis of variance. # Results Twenty-three (14 males, 9 females) were included. During execution of the squat, spinal alignment adjustments in the lumbar spine were made in all three planes of movement, compared to the start position, in both male and female lifters. Compared to their standing habitual posture, all lifters adjusted their upper lumbar spine to a less lordotic position when in the start position of the back squat (standing upright with the barbell on their back). Only male lifters assumed a less lordotic alignment in their lower lumbar spine in the start position compared their habitual posture. # Conclusions Adjustments of spinal alignment, predominantly in the sagittal plane, are made during execution of the back squat in both male and female lifters. Further, lifters adopt a less lordotic alignment with a heavy barbell on their upper back, more so in male than female lifters. In conclusion, it seems that spinal alignment changes noticeably during the barbell back squat. # Level of Evidence 3 ©The Author(s

    Factory Radio Design of a 5G Network in Offline Mode

    The manufacturing industry is connecting people and equipment with new digital technologies, enabling a more continuous stream of data to represent processes. With more things connected, the interest in a connectivity solution that can support communication with high reliability and availability will increase. The fifth generation of telecommunication, i.e., 5G has promising features to deliver this, but the factory environment introduces new challenges to ensure reliable radio coverage. This will require efficient ways to plan the Factory Radio Design prior to installation. 3D laser scanning is used at an ever-increasing rate for capturing the spatial geometry in a virtual representation to perform layout planning of factories. This paper presents how to combine 3D laser scanning and physical optics (PO) for planning the Factory Radio Design of a cellular Long-Term Evolution (LTE) network (5G) in a virtual environment. 3D laser scanning is applied to obtain the spatial data of the factory and the virtual representation serves as the environment where PO computation techniques can be performed. The simulation result is validated in this paper by comparison to measurements of the installed network and empirical propagation models. The results of the study show promising opportunities to simulate the radio coverage in a virtual representation of a factory environment

    Karonudib is a promising anticancer therapy in hepatocellular carcinoma

    Background: Hepatocellular carcinoma (HCC) is the most common form of liver cancer and is generally caused by viral infections or consumption of mutagens, such as alcohol. While liver transplantation and hepatectomy is curative for some patients, many relapse into disease with few treatment options such as tyrosine kinase inhibitors, for example, sorafenib or lenvatinib. The need for novel systemic treatment approaches is urgent. Methods: MTH1 expression profile was first analyzed in a HCC database and MTH1 mRNA/protein level was determined in resected HCC and paired paracancerous tissues with polymerase chain reaction (PCR) and immunohistochemistry. HCC cancer cell lines were exposed in vitro to MTH1 inhibitors or depleted of MTH1 by siRNA. 8-oxoG was measured by the modified comet assay. The effect of MTH1 inhibition on tumor growth was explored in HCC xenograft in vivo models. Results: MTH1 protein level is elevated in HCC tissue compared with paracancerous liver tissue and indicates poor prognosis. The MTH1 inhibitor Karonudib (TH1579) and siRNA effectively introduce toxic oxidized nucleotides into DNA, 8-oxoG, and kill HCC cell lines in vitro. Furthermore, we demonstrate that HCC growth in a xenograft mouse model in vivo is efficiently suppressed by Karonudib. Conclusion: Altogether, these data suggest HCC relies on MTH1 for survival, which can be targeted and may open up a novel treatment option for HCC in the future

    Technical Interoperability for Machine Connectivity on the Shop Floor

    This paper presents a generic technical solution that can increase Industry 4.0 maturity by collecting data from sensors and control systems on the shop floor. Within the research project “5G-Enabled Manufacturing”, an LTE (Long-Term Evolution) network with 5G technologies was deployed on the shop floor to enable fast and scalable connectivity. This network was used to connect a grinding machine to a remote private cloud where data was stored and streamed to a data analytics center. This enabled visibility and transparency of the production data, which is the basis for Industry 4.0 and smart manufacturing. The solution is described with a focus on high-level communication technologies above wireless communication standards. These technologies are discussed regarding technical interoperability, focusing on the system layout, communication standards, and open systems. From the discussion, it can be derived that generic solutions such as this are possible, but manufacturing end-users must expand and further internalize knowledge of future information and communication technologies to reduce their dependency on equipment and technology providers

    MTH1 inhibitor TH588 disturbs mitotic progression and induces mitosis-dependent accumulation of genomic 8-oxodG

    Reactive oxygen species (ROS) oxidize nucleotide triphosphate pools (e.g., 8-oxodGTP), which may kill cells if incorporated into DNA. Whether cancers avoid poisoning from oxidized nucleotides by preventing incorporation via the oxidized purine diphosphatase MTH1 remains under debate. Also, little is known about DNA polymerases incorporating oxidized nucleotides in cells or how oxidized nucleotides in DNA become toxic. Here we show that replacement of one of the main DNA replicases in human cells, DNA polymerase delta (Pol δ), with an error-prone variant allows increased 8-oxodG accumulation into DNA following treatment with TH588, a dual MTH1 inhibitor (MTH1i) and microtubule targeting agent. The resulting elevated genomic 8-oxodG correlated with increased cytotoxicity of TH588. Interestingly, no substantial perturbation of replication fork progression was observed, but rather mitotic progression was impaired and mitotic DNA synthesis triggered. Reducing mitotic arrest by reversin treatment prevented accumulation of genomic 8-oxodG and reduced cytotoxicity of TH588, in line with the notion that mitotic arrest is required for ROS buildup and oxidation of the nucleotide pool. Furthermore, delayed mitosis and increased mitotic cell death was observed following TH588 treatment in cells expressing the error-prone but not wild type Pol δ variant, which is not observed following treatments with anti-mitotic agents. Collectively, these results link accumulation of genomic oxidized nucleotides with disturbed mitotic progression
