    Investigations of a compartmental model for leucine kinetics using nonlinear mixed effects models with ordinary and stochastic differential equations

    Nonlinear mixed effects models represent a powerful tool to simultaneously analyze data from several individuals. In this study a compartmental model of leucine kinetics is examined and extended with a stochastic differential equation to model non-steady state concentrations of free leucine in the plasma. Data obtained from tracer/tracee experiments for a group of healthy control individuals and a group of individuals suffering from diabetes mellitus type 2 are analyzed. We find that the interindividual variation of the model parameters is much smaller for the nonlinear mixed effects models, compared to traditional estimates obtained from each individual separately. Using the mixed effects approach, the population parameters are estimated well also when only half of the data are used for each individual. For a typical individual the amount of free leucine is predicted to vary with a standard deviation of 8.9% around a mean value during the experiment. Moreover, leucine degradation and protein uptake of leucine is smaller, proteolysis larger, and the amount of free leucine in the body is much larger for the diabetic individuals than the control individuals. In conclusion nonlinear mixed effects models offers improved estimates for model parameters in complex models based on tracer/tracee data and may be a suitable tool to reduce data sampling in clinical studies

    Lærerens støtte av matematisk argumentasjon på småtrinnet og mellomtrinnet

    Hensikten med studien er å få innsikt i hvilke muligheter og støtte elever på barneskolen får til å argumentere for sine svar og resonnement i samtaler i matematikkundervisningen. Disse samtalene ledes av lærer i ulik grad og det er derfor interessant å se på hvordan læreren støtter og legger til rette for argumentasjon og resonnement i matematikksamtalene. I denne studien har vi svart på følgende forskningsspørsmål: Hvordan støtter læreren elevenes argumentasjon i de matematiske samtalene? Studien har en kvalitativ tilnærming og bygger på lyd- og videoopptak, samt observasjonsnotater i to ulike klasser ved en barneskole. Utvalget består av to ulike klasser med to ulike lærere – én 6. klasse og én 3. klasse. De to klassene har hatt forskjellig undervisningsopplegg, der temaet for økten er bestemt av den respektive læreren. I studien er det gjennomført en analyse av elevenes resonnering og argumentasjon ved bruk av rammeverket til Conner, Singletary, Smith, Wagner og Francisco (2014) som identifiserer lærerens grep i den kollektive klasseromssamtalen. Videoopptakene våre ble transkribert og systematisert i henhold til overnevnte rammeverk. I diskusjonskapittelet (Kapittel 5), vil vi trekke frem mønster vi har sett i klasseromssamtalen og sammenlikne funn fra analysen opp mot tidligere forskning og teori på området. Grad av åpenhet i undervisningen, klassemiljø, normer og regler i klassen og lærerens rolle er faktorer som kan påvirke elevenes resonnering og argumentasjon

    Improving the thermal stability of different types of xylan by acetylation

    The impact of various degrees of acetylation on improving the thermal stability of xylan isolated from different botanical source has been studied; methylglucuronoxylan from birch and eucalyptus, arabinoglucuronoxylan from spruce and glucuronoarabinoxylan from sugarcane bagasse and straw. The lower molecular weight of nonacetylated methylglucuronoxylan (17.7-23.7 kDa) and arabinoglucuronoxylan (16.8 kDa) meant that they were more soluble in water than glucuronoarabinoxylan (43.0-47.0 kDa). The temperature at the onset of degradation increased by 17-61 degrees C and by 75-145 degrees C for low and high acetylated xylans respectively, as a result of acetylation. A glass transition temperature in the range of 121-132 degrees C was observed for the samples non-acetylated and acetylated at low degree of acetylation (0.0-0.6). The acetylation to higher degrees (1.4-1.8) increased the glass transition temperature of the samples to 189-206 degrees C. Acetylation proved to be an efficient method for functionalization of the xylan to increase the thermal stability.Peer reviewe

    Impact of birch xylan composition and structure on film formation and properties

    Commercial birch xylan (CX) and alkali-soluble birch xylan (ASX) were subjected to controlled acetylation and used for film formation in the presence (20% and 40%) or absence of plasticizers (i.e. glycerol, sorbitol and xylitol). Although the content of Klason lignin was similar (1.2-1.4%), the acetylation process was favored by the high-purity CX (97% xylan) over the ASX (89% xylan). On the other hand, the presence of residual pectin heteropolysaccharides rather than xylan in the ASX sample was beneficial for film formation. These heteropolysaccharides seemed to act as natural plasticizers during film formation, allowing the formation of coherent films from ASX, even in the absence of an external plasticizer. The use of plasticizers favored the mechanical properties of films, especially in a dosage of 40%, when plastic behavior was created. Acetylation favored the film formation and slightly improved the mechanical properties of the films, and this improvement was in the same range as that achieved when using 20% plasticizer in non-acetylated ASX.Peer reviewe

    Tillstånd och trender för arter och deras livsmiljöer

    2015 års upplaga av den svenska rödlistan är den fjärde i ordningen. Den är baserad på IUCN:s rödlistningskriterier och revideras vart femte år. I rödlistan bedöms risken som enskilda arter av djur, växter och svampar löper att försvinna från Sverige. Bedömningen utförs av ArtDatabankens medarbetare i samverkan med över 100 externa experter, indelade i 14 expertkommittéer för olika organismgrupper. Under arbetet med 2015 års rödlista har tillstånd och trender bedömts för 21 600 arter och 1 318 lägre taxa (apomiktiska arter, underarter och varieteter), sammanlagt ca 22 900 taxa. Av de bedömda arterna klassificerades 2 029 som hotade (kategorierna CR, EN och VU) och 4 273 som rödlistade (inkluderar även kategorierna NT, RE och DD). Förhållandet mellan antalet rödlistade och antalet bedömda arter ar 19,8 %, vilket är ungefär samma värde som 2010 och 2005. I denna rapport jämförs antalet och andelen rödlistade arter mellan olika organismgrupper, biotoper, substrat och påverkansfaktorer. Texten ar indelad i en allmän del och åtta kapitel inriktade på olika landskapstyper. Landskapstyperna utgör en grov indelning av landets miljöer enligt följande kategorier: Skog, Jordbrukslandskap, Urbana miljöer, Fjäll, Våtmarker, Sötvatten, Havsstränder och Havsmiljöer. Skogen och jordbrukslandskapet är de artrikaste landskapstyperna med 1 800 respektive 1 400 arter som har en stark anknytning dit, och ytterligare flera hundra arter som förekommer där mer sporadiskt. De faktorer som påverkar flest rödlistade arter i Sverige är skogsavverkning och igenväxning, som båda utgör ett hot mot vardera ca 30 % av de rödlistade arterna. Avverkning minskar arealen av skog där naturliga strukturer och naturlig dynamik upprätthålls, och den orsakar därmed förlust av livsmiljöer. Igenväxning orsakas av ett antal faktorer, bland annat upphörande hävd (bete och slåtter), gödsling, trädplantering och brist på naturliga störningsregimer som t.ex. regelbundna översvämningar kring vattendrag och sjöar. Andra viktiga påverkansfaktorer är fiske, torrläggning av våtmarker, tillbakagång hos värdarter (främst alm och ask som drabbats av invasiva svampsjukdomar), klimatförändringar och konkurrens från invasiva arter. IUCN:s rödlisteindex beräknas för ett urval av de bedömda organismgrupperna. Rödlisteindex visar att skillnaderna mellan rödlistorna från 2000, 2005, 2010 och 2015 är små. Ett par undantag finns dock. Groddjur och stora däggdjur har fått en något förbättrad situation sedan 2000. Totalt förefaller det ändå som att trycket mot Sveriges artstock har förblivit relativt konstant under de senaste 15 åren

    Genetically controlled mtDNA deletions prevent ROS damage by arresting oxidative phosphorylation

    Deletion of mitochondrial DNA in eukaryotes is currently attributed to rare accidental events associated with mitochondrial replication or repair of double-strand breaks. We report the discovery that yeast cells arrest harmful intramitochondrial superoxide production by shutting down respiration through genetically controlled deletion of mitochondrial oxidative phosphorylation genes. We show that this process critically involves the antioxidant enzyme superoxide dismutase 2 and two-way mitochondrial-nuclear communication through Rtg2 and Rtg3. While mitochondrial DNA homeostasis is rapidly restored after cessation of a short-term superoxide stress, long-term stress causes maladaptive persistence of the deletion process, leading to complete annihilation of the cellular pool of intact mitochondrial genomes and irrevocable loss of respiratory ability. This shows that oxidative stress-induced mitochondrial impairment may be under strict regulatory control. If the results extend to human cells, the results may prove to be of etiological as well as therapeutic importance with regard to age-related mitochondrial impairment and disease

    Phthalate Diesters and Their Metabolites in Human Breast Milk, Blood or Serum, and Urine as Biomarkers of Exposure in Vulnerable Populations

    BACKGROUND: Phthalates may pose a risk for perinatal developmental effects. An important question relates to the choice of suitable biological matrices for assessing exposure during this period. OBJECTIVES: This study was designed to measure the concentrations of phthalate diesters or their metabolites in breast milk, blood or serum, and urine and to evaluate their suitability for assessing perinatal exposure to phthalates. METHODS: In 2001, 2-3 weeks after delivery, 42 Swedish primipara provided breast milk, blood, and urine samples at home. Special care was taken to minimize contamination with phthalates (e.g., use of a special breast milk pump, heat treatment of glassware and needles, addition of phosphoric acid). RESULTS: Phthalate diesters and metabolites in milk and blood or serum, if detected, were present at concentrations close to the limit of detection. By contrast, most phthalate metabolites were detectable in urine at concentrations comparable to those from the general population in the United States and in Germany. No correlations existed between urine concentrations and those found in milk or blood/serum for single phthalate metabolites. Our data are at odds with a previous study documenting frequent detection and comparatively high concentrations of phthalate metabolites in Finnish and Danish mothers' milk. CONCLUSIONS: Concentrations of phthalate metabolites in urine are more informative than those in milk or serum. Furthermore, collection of milk or blood may be associated with discomfort and potential technical problems such as contamination (unless oxidative metabolites are measured). Although urine is a suitable matrix for health-related phthalate monitoring, urinary concentrations in nursing mothers cannot be used to estimate exposure to phthalates through milk ingestion by breast-fed infants
