The Impact of a Substance Abuse Treatment for Offenders in Relation to the Time Spent in Treatment
The objective of this research is to evaluate the outcomes of a treatment for addicts. 123 subjects were tested before treatment and at 5, 8 and 11 months follow-up periods with a French version of the Addiction Severity Index (ASI). Exposure to treatment was based on the number of clients’ contact-hours with a therapist. The sample was divided into three groups according to the number of hours spent in treatment. The data was analysed using MANOVA on the seven scales of the ASI for the three groups and the four time periods. Results showed that all groups improved significantly but that this improvement was not related to the number of hours spent in treatment
HMG-box domain stimulation of RAG1/2 cleavage activity is metal ion dependent
<p>Abstract</p> <p>Background</p> <p>RAG1 and RAG2 initiate V(D)J recombination by assembling a synaptic complex with a pair of antigen receptor gene segments through interactions with their flanking recombination signal sequence (RSS), and then introducing a DNA double-strand break at each RSS, separating it from the adjacent coding segment. While the RAG proteins are sufficient to mediate RSS binding and cleavage <it>in vitro</it>, these activities are stimulated by the architectural DNA binding and bending factors HMGB1 and HMGB2. Two previous studies (Bergeron <it>et al.</it>, 2005, and Dai <it>et al.</it>, 2005) came to different conclusions regarding whether only one of the two DNA binding domains of HMGB1 is sufficient to stimulate RAG-mediated binding and cleavage of naked DNA <it>in vitro</it>. Here we test whether this apparent discrepancy is attributed to the choice of divalent metal ion and the concentration of HMGB1 used in the cleavage reaction.</p> <p>Results</p> <p>We show here that single HMG-box domains of HMGB1 stimulate RAG-mediated RSS cleavage in a concentration-dependent manner in the presence of Mn2+, but not Mg2+. Interestingly, the inability of a single HMG-box domain to stimulate RAG-mediated RSS cleavage in Mg2+ is overcome by the addition of partner RSS to promote synapsis. Furthermore, we show that mutant forms of HMGB1 which otherwise fail to stimulate RAG-mediated RSS cleavage in Mg2+ can be substantially rescued when Mg2+ is replaced with Mn2+.</p> <p>Conclusion</p> <p>The conflicting data published previously in two different laboratories can be substantially explained by the choice of divalent metal ion and abundance of HMGB1 in the cleavage reaction. The observation that single HMG-box domains can promote RAG-mediated 23-RSS cleavage in Mg2+ in the presence, but not absence, of partner RSS suggests that synaptic complex assembly <it>in vitro </it>is associated with conformational changes that alter how the RAG and/or HMGB1 proteins bind and bend DNA in a manner that functionally replaces the role of one of the HMG-box domains in RAG-HMGB1 complexes assembled on a single RSS.</p

This article reports on six evaluative research projects led by a research team at the RISQ (Recherche et intervention sur les substances psychoactives – QuĂ©bec) on treatments offered in Quebec, mainly in public rehabilitation centres, between 1991 and 2005. Objectives: The goal of these studies was to evaluate: 1) whether the participants from drug addiction treatment centres in Quebec improved their situation during and after their treatment; 2) the spheres of life in which these improvements occurred; and 3) whether there was a relationship between the changes observed and the participants’ exposure to the treatment. Method: The method is referred to as a “naturalist” approach: participants were exposed to the regular treatment offered in the centres where they were followed. By means of the ASI-IGT, their situation concerning consumption of psychoactive substances and their bio-psychosocial condition were evaluated at three measurement times, on their arrival in treatment, then at intervals of approximately 6 and 12 months. Measurements also included the quantity (hours of exposure to treatment) and the duration of the intervention they received. Results: In the six groups under study, a constant improvement of the participants on the ASI-IGT “alcohol” and “drug” scales was observed as well as most of the dimensions evaluated by this tool in time 1 and time 2, and maintenance of this improvement in time 3. There was no significant difference between the participants who had little or no treatment and those who had a great deal. Interpretation: The meaning and the extent of these results are discussed by the authors, particularly in relation to the limitations of these studies. Research strategies developed thereafter to respond to the questions raised by these studies are also presented.
Risk factors associated with driving after cannabis use among Canadian young adults
This study identifid the most prominent risk factors associated with driving after cannabis use (DACU). 1,126 Canadian drivers (17–35 years old) who have used cannabis in the past 12 months completed an online questionnaire about sociodemographic information, substance use habits, cannabis effect expectancies, driving behaviours and peers’ behaviours and attitudes concerning DACU. A hierarchical logistic regression allowed identifying variables that were associated with DACU. Income (CA69,000), weekly-to-daily cannabis use, higher level of cannabis-related problems, expectation that cannabis facilitates social interactions, drunk driving, belief that DACU is safe, general risky driving behaviours, having a few friends who had DACU and injunctive norms predicted past 12-month DACU. Older age, holding negative expectations concerning cannabis, driving aggressively and perceived accessibility of public transportation decreased the probability of DACU. With restricted resources, programmes will be more efficient by targeting Canadian young adults most inclined to DACU by focussing on these risk factors
The aim of this study is to acquire a better understanding of the offenders admitted to an addiction treatment facility. The sample is Two hundred and sixteen client admitted to two public addiction treatment facilities of Province of Québec. These subjects answered to the French form of the Addiction Severity Index (ASI-IGT), the Millon Clinical Multiaxial Inventory (MCMI), the Jesness Inventory Personality Scales, and the University of Rhodes Island Change Assessment Scale (URICA). These psychometric tools are used to describe our sample. The analysis leads to two main discussion themes. First, the multiple problematics observed in our sample confront the clinicians with the difficulty to offer treatment services that will adequately answer to these various needs. Second, the low level of addiction of some clients referred by the justice system challenges the relevance to refer these clients to an addiction treatment facility
