    Deuteron yields from LHC: Continuum correlations and in-medium effects

    To explain the production of light nuclei in heavy-ion collisions at extreme energies, we focus on the deuteron case. A Gibbs ensemble at chemical freeze-out is a prerequisite to investigate the non-equilibrium evolution of the expanding fireball. Quantum statistical approaches allow to describe correlations including bound state formation in the strongly interacting and hot system. We consider the virial approach to evaluate proton-neutron correlations. In generalization of the treatment of protons in pionic matter (pion-proton puzzle), the influence of the pion environment on deuteron-like correlations is evaluated using data for the pion-deuteron scattering phase shifts. Calculated yields for deuteron production are compared with the ones observed at the LHC.Comment: 15 pages, 3 figures; accepted for publication by Phys. Rev.

    Unterschiede im Migrationsverhalten und Metabolismus zwischen weiblichen und mÀnnlichen Endothelzellen der Nabelschnur (HUVEC)

    Herz-Kreislauf-Erkrankungen stellen die Haupttodesursache fĂŒr MĂ€nner und Frauen dar, jedoch erkranken Frauen erst wesentlich spĂ€ter. Neben hormonellen EinflĂŒssen werden auch sexspezifische Unterschiede auf zellulĂ€rer Ebene als Grund dafĂŒr diskutiert. Das Endothel spielt eine entscheidende Rolle fĂŒr die GefĂ€ĂŸhomöostase. Eine Dysfunktion des Endothels korreliert mit der Entstehung der Atherosklerose und Herz-Kreislauf-Erkrankungen. In der vorliegenden Arbeit wurden Endothelzellen unter sexspezifischen Aspekten auf funktionelle und metabolische Unterschiede untersucht. DafĂŒr wurden weitgehend weibliche und mĂ€nnliche Endothelzellen der Nabelschnur (HUVECs) von dizygoten Zwillingen verwendet, um Hormon- und UmwelteinflĂŒsse auf sexspezifische Unterschiede zu minimieren. In unbehandelten Zellen ergaben sich keine wesentlichen sexspezifischen Unterschiede. Nach Behandlung mit VEGF jedoch wurde eine stĂ€rkere Migration in weiblichen HUVECs beobachtet. Die intrazellulĂ€re ATP-Konzentration war in weiblichen HUVECs nach VEGF signifikant höher. Serumreduktion fĂŒhrte in mĂ€nnlichen nicht jedoch in weiblichen HUVECs zu einer signifikanten Verminderung der ATP-Konzentration. Nach Behandlung mit VEGF war der Quotient aus Glykolyse und mitochondrialer Atmung in den meisten HUVEC-Zwillingspaaren höher bei weiblichen Zellen. Im Gegensatz dazu wurde nach Serumreduktion eine höhere basale mitochondriale Atmung in mĂ€nnlichen HUVECs gemessen. Ein wesentlicher sexspezifischer Unterschied in der PFKFB3-Expression (6-Phosphofructo-2- Kinase/Fructose-2,6-Biphosphatase-3), ein Hauptregulierungsenzym der Glykolyse, zeigte sich jedoch nicht. Insgesamt konnte in dieser Arbeit ein Sexualdimorphismus im Migrationsverhalten sowie im Energiemetabolismus festgestellt werden. Eine höhere ATP-Konzentration nach Serumreduktion und VEGF-Behandlung sowie eine stĂ€rkere VEGF-induzierte Migration deuten auf sexspezifische Unterschiede auf zellulĂ€rer Ebene hin. Eine bessere Energiebilanz sowie eine grĂ¶ĂŸere Glykolyse-AktivitĂ€t könnten im Zusammenhang mit einer besseren Migration weiblicher HUVECs stehen.Cardiovascular diseases (CVDs) are the main cause of death in men and women, however these develop later in women. In addition to hormonal influences, sex- specific differences at a cellular level could play a crucial role. The endothelium is responsible for maintaining vascular homeostasis and its dysfunction may lead to atherosclerosis und CVDs. The present study evaluated sex-specific functional and metabolic differences in endothelial cells. Human umbilical vein endothelial cells (HUVECs) from dizygotic twins were used to reduce hormonal and environmental influences on sex-specific differences. No sex-specific differences were observed in untreated cells. However, after VEGF stimulation, the migration in female HUVECs was significantly more pronounced after VEGF. Intracellular ATP levels were significantly higher in female cells after VEGF. Serum starvation resulted in a significant decrease of ATP levels in male but not in female HUVECs. After VEGF, the ratio of glycolysis/mitochondrial respiration was higher in female HUVECs in most twin pairs. In contrast, male HUVECs showed higher basal mitochondrial respiration after serum starvation. No sex-specific differences were observed in the expression of PFKFB3 (6- Phosphofructo-2-Kinase/Fructose-2,6-Biphosphatase-3), a rate-regulating enzyme of glycolysis. In conclusion, this study identified sex-specific differences in cellular migration and metabolism. Higher ATP levels after VEGF and serum starvation, as well as a more pronounced VEGF-induced migration in female HUVECs, indicate sex-specific differences at the cellular level. A better energy balance and higher glycolytic activity could contribute to a stronger migration capacity in female HUVECs

    Enterovirus D68 outbreak detection through a syndromic disease epidemiology network

    BACKGROUND: In 2014, enterovirus D68 (EV-D68) was responsible for an outbreak of severe respiratory illness in children, with 1,153 EV-D68 cases reported across 49 states. Despite this, there is no commercial assay for its detection in routine clinical care. BioFireÂź Syndromic Trends (Trend) is an epidemiological network that collects, in near real-time, deidentified. BioFire test results worldwide, including data from the BioFireÂź Respiratory Panel (RP). OBJECTIVES: Using the RP version 1.7 (which was not explicitly designed to differentiate EV-D68 from other picornaviruses), we formulate a model, Pathogen Extended Resolution (PER), to distinguish EV-D68 from other human rhinoviruses/enteroviruses (RV/EV) tested for in the panel. Using PER in conjunction with Trend, we survey for historical evidence of EVD68 positivity and demonstrate a method for prospective real-time outbreak monitoring within the network. STUDY DESIGN: PER incorporates real-time polymerase chain reaction metrics from the RPRV/EV assays. Six institutions in the United States and Europe contributed to the model creation, providing data from 1,619 samples spanning two years, confirmed by EV-D68 gold-standard molecular methods. We estimate outbreak periods by applying PER to over 600,000 historical Trend RP tests since 2014. Additionally, we used PER as a prospective monitoring tool during the 2018 outbreak. RESULTS: The final PER algorithm demonstrated an overall sensitivity and specificity of 87.1% and 86.1%, respectively, among the gold-standard dataset. During the 2018 outbreak monitoring period, PER alerted the research network of EV-D68 emergence in July. One of the first sites to experience a significant increase, Nationwide Children's Hospital, confirmed the outbreak and implemented EV-D68 testing at the institution in response. Applying PER to the historical Trend dataset to determine rates among RP tests, we find three potential outbreaks with predicted regional EV-D68 rates as high as 37% in 2014, 16% in 2016, and 29% in 2018. CONCLUSIONS: Using PER within the Trend network was shown to both accurately predict outbreaks of EV-D68 and to provide timely notifications of its circulation to participating clinical laboratories

    Modulation of Cell Adhesion and Migration by the Histone Methyltransferase Subunit mDpy-30 and Its Interacting Proteins

    We have previously shown that a subset of mDpy-30, an accessory subunit of the nuclear histone H3 lysine 4 methyltransferase (H3K4MT) complex, also localizes at the trans-Golgi network (TGN), where its recruitment is mediated by the TGN-localized ARF guanine nucleotide exchange factor (ArfGEF) BIG1. Depletion of mDpy-30 inhibits the endosome-to-TGN transport of internalized CIMPR receptors and concurrently promotes their accumulation at the cell protrusion. These observations suggest mDpy-30 may play a novel role at the crossroads of endosomal trafficking, nuclear transcription and adhesion/migration. Here we provide novel mechanistic and functional insight into this association. First, we demonstrate a direct interaction between mDpy-30 and BIG1 and locate the binding region in the N-terminus of BIG1. Second, we provide evidence that the depletion or overexpression of mDpy-30 enhances or inhibits cellular adhesion/migration of glioma cells in vitro, respectively. A similar increase in cell adhesion/migration is observed in cells with reduced levels of BIG1 or other H3K4MT subunits. Third, knockdown of mDpy-30, BIG1, or the RbBP5 H3K4MT subunit increases the targeting of ÎČ1 integrin to cell protrusions, and suppression of H3K4MT activity by depleting mDpy-30 or RbBP5 leads to increased protein and mRNA levels of ÎČ1 integrin. Moreover, stimulation of cell adhesion/migration via mDpy-30 knockdown is abolished after treating cells with a function-blocking antibody to ÎČ1 integrin. Taken together, these data indicate that mDpy-30 and its interacting proteins function as a novel class of cellular adhesion/migration modulators partially by affecting the subcellular distribution of endosomal compartments as well as the expression of key adhesion/migration proteins such as ÎČ1 integrin

    Visualizing variation within Global Pneumococcal Sequence Clusters (GPSCs) and country population snapshots to contextualize pneumococcal isolates.

    Knowledge of pneumococcal lineages, their geographic distribution and antibiotic resistance patterns, can give insights into global pneumococcal disease. We provide interactive bioinformatic outputs to explore such topics, aiming to increase dissemination of genomic insights to the wider community, without the need for specialist training. We prepared 12 country-specific phylogenetic snapshots, and international phylogenetic snapshots of 73 common Global Pneumococcal Sequence Clusters (GPSCs) previously defined using PopPUNK, and present them in Microreact. Gene presence and absence defined using Roary, and recombination profiles derived from Gubbins are presented in Phandango for each GPSC. Temporal phylogenetic signal was assessed for each GPSC using BactDating. We provide examples of how such resources can be used. In our example use of a country-specific phylogenetic snapshot we determined that serotype 14 was observed in nine unrelated genetic backgrounds in South Africa. The international phylogenetic snapshot of GPSC9, in which most serotype 14 isolates from South Africa were observed, highlights that there were three independent sub-clusters represented by South African serotype 14 isolates. We estimated from the GPSC9-dated tree that the sub-clusters were each established in South Africa during the 1980s. We show how recombination plots allowed the identification of a 20 kb recombination spanning the capsular polysaccharide locus within GPSC97. This was consistent with a switch from serotype 6A to 19A estimated to have occured in the 1990s from the GPSC97-dated tree. Plots of gene presence/absence of resistance genes (tet, erm, cat) across the GPSC23 phylogeny were consistent with acquisition of a composite transposon. We estimated from the GPSC23-dated tree that the acquisition occurred between 1953 and 1975. Finally, we demonstrate the assignment of GPSC31 to 17 externally generated pneumococcal serotype 1 assemblies from Utah via Pathogenwatch. Most of the Utah isolates clustered within GPSC31 in a USA-specific clade with the most recent common ancestor estimated between 1958 and 1981. The resources we have provided can be used to explore to data, test hypothesis and generate new hypotheses. The accessible assignment of GPSCs allows others to contextualize their own collections beyond the data presented here
