    Asymmetrical flow field flow fractionation - Multidetection system as a tool for studying metal-alginate interactions

    The present study explores the potential use of asymmetrical flow field flow fractionation (aFlFFF) with a multidetection system for the study of metal-alginate interactions. aFlFFF, coupled on-line to a differential refractive index and seven angle laser light scattering detectors was used to provide information on the alginate size distributions. In parallel, the metal distributions of metal-alginate complexes were probed by aFlFFF-high resolution inductively coupled plasma-mass spectrometry. Average values and continuous distributions of molar masses, radiuses of gyration and hydrodynamic radiuses, which are critical for understanding the role of alginates as carriers of metal pollutants, were evaluated in presence of Pb or Cd and compared with those in metal-free solutions of alginate. The values of number average and weight average molar mass, weight average radius of gyration and shape factor for alginate were 150 and 188 kg mol(-1), 53 nm and 1.7, respectively. Alginate molar mass and radius of gyration distributions were slightly shifted to higher values by the addition of micromolar concentrations of Pb or Cd. The alginate size distribution in the presence of Cd was similar to the alginate-alone control, whereas in the presence of Pb the size distribution was broader with a shift of the maximum toward higher molar masses

    Dissecting the Potential Roles of Nigella sativa and Its Constituent Thymoquinone on the Prevention and on the Progression of Alzheimer's Disease

    Several nutraceuticals have been investigated for preventing or retarding the progression of different neurodegenerative diseases, including Alzheimer's disease (AD). Because Nigella sativa (NS) and its isolated compound thymoquinone (TQ) have significant anti-oxidant and anti-inflammatory proprieties, they could represent effective neuroprotective agents. The purpose of this manuscript is to analyze and to recapitulate the results of in vitro and in vivo studies on the potential role of NS/TQ in AD's prevention and treatment. The level of evidence for each included animal study has been assessed by using a modified CAMARADES (Collaborative Approach to Meta-Analysis and Review of Animal Data from Experimental Studies) 10-item checklist. We used MEDLINE and EMBASE databases to screen relevant articles published up to July 2017. A manual search was also performed. The database search yielded 38 studies, of which 18 were included in this manuscript. Results from these approaches suggest that NS or TQ could represent an effective strategy against AD due to the balancing of oxidative processes and the binding to specific intracellular targets. The overall effects mainly regard the prevention of hippocampal pyramidal cell loss and the increased cognitive functions

    Variant of Rett Syndrome and CDKL5 gene: clinical and autonomic description of 10 cases

    Rett syndrome (RTT) is a severe neurodevelopmental disorder affecting almost exclusively females. The Hanefeld variant, or early-onset seizure variant, has been associated with mutations in CDKL5 gene. AIMS: In recent years more than 60 patients with mutations in the CDKL5 gene have been described in the literature, but the cardiorespiratory phenotype has not been reported. Our aim is to describe clinical and autonomic features of these girls. METHODS: 10 girls with CDKL5 mutations and a diagnosis of Hanefeld variant have been evaluated on axiological and clinical aspects. In all subjects an evaluation of the autonomic system was performed using the Neuroscope. RESULTS: Common features were gaze avoidance, repetitive head movements and hand stereotypies. The autonomic evaluation disclosed eight cases with the Forceful breather cardiorespiratory phenotype and two cases with the Apneustic breather phenotype. CONCLUSIONS: The clinical picture remains within the RTT spectrum but some symptoms are more pronounced in addition to the very early onset of seizures. The cardiorespiratory phenotype was dominated by Forceful breathers, while Feeble breathers were not found, differently from the general Rett population, suggesting a specific behavioral and cardiorespiratory phenotype of the RTT the Hanefeld variant
