    L1 and HERV-W retrotransposons are hypomethylated in human ovarian carcinomas

    Wide-spread hypomethylation of CpG dinucleotides is characteristic of many cancers. Retrotransposons have been identified as potential targets of hypomethylation during cellular transformation. We report the results of an preliminary examination of the methylation status of CpG dinucleotides associated with the L1 and HERV-W retrotransposons in benign and malignant human ovarian tumors. We find a reduction in the methylation of CpG dinucleotides within the promoter regions of these retroelements in malignant relative to non-malignant ovarian tissues. Consistent with these results, we find that relative L1 and HERV-W expression levels are elevated in representative samples of malignant vs. non-malignant ovarian tissues

    Visual Cortical Traveling Waves: From Spontaneous Spiking Populations to Stimulus-Evoked Models of Short-Term Prediction

    Thanks to recent advances in neurotechnology, waves of activity sweeping across entire cortical regions are now routinely observed. Moreover, these waves have been found to impact neural responses as well as perception, and the responses themselves are found to be structured as traveling waves. How exactly do these waves arise? Do they confer any computational advantages? These traveling waves represent an opportunity for an expanded theory of neural computation, in which their dynamic local network activity may complement the moment-to-moment variability of our sensory experience. This thesis aims to help uncover the origin and role of traveling waves in the visual cortex through three Works. In Work 1, by simulating a network of conductance-based spiking neurons with realistically large network size and synaptic density, distance-dependent horizontal axonal time delays were found to be important for the widespread emergence of spontaneous traveling waves consistent with those in vivo. Furthermore, these waves were found to be a dynamic mechanism of gain modulation that may explain the in-vivo result of their modulation of perception. In Work 2, the Kuramoto oscillator model was formulated in the complex domain to study a network possessing distance-dependent time delays. Like in Work 1, these delays produced traveling waves, and the eigenspectrum of the complex-valued delayed matrix, containing a delay operator, provided an analytical explanation of them. In Work 3, the model from Work 2 was adapted into a recurrent neural network for the task of forecasting the frames of videos, with the question of how such a biologically constrained model may be useful in visual computation. We found that the wave activity emergent in this network was helpful, as they were tightly linked with high forecast performance, and shuffle controls revealed simultaneous abolishment of both the waves and performance. All together, these works shed light on the possible origins and uses of traveling waves in the visual cortex. In particular, time delays profoundly shape the spatiotemporal dynamics into traveling waves. This was confirmed numerically (Work 1) and analytically (Work 2).     A linear domain decomposition method for two-phase flow in porous media

    This article is a follow up of our submitted paper [11] in which a decomposition of the Richards equation along two soil layers was discussed. A decomposed problem was formulated and a decoupling and linearisation technique was presented to solve the problem in each time step in a fixed point type iteration. This article extends these ideas to the case of two-phase in porous media and the convergence of the proposed domain decomposition method is rigorously shown.Comment: 8 page

    The social impact of planting and harvesting the jatropha curca for the production of biodiesel in the province of Manabí is shown, where there are several municipalities involved in this process, mainly the populations that live in rural areas where the conditions are created for their improvement of quality and good living from the income they receive for performing these tasks. A social relationship model was designed that links the social development of rural areas in the province of Manabí with the sowing, harvesting and industrialization of jatropha curca in social development, demonstrating the benefit of using indigenous natural resources for the in the improvement of the quality of life of the populations directed to the economic, energetic, social and environmental sustainable development

    State-dependent activity dynamics of hypothalamic stress effector neurons

    The stress response necessitates an immediate boost in vital physiological functions from their homeostatic operation to an elevated emergency response. However, the neural mechanisms underlying this state-dependent change remain largely unknown. Using a combination of in vivo and ex vivo electrophysiology with computational modeling, we report that corticotropin releasing hormone (CRH) neurons in the paraventricular nucleus of the hypothalamus (PVN), the effector neurons of hormonal stress response, rapidly transition between distinct activity states through recurrent inhibition. Specifically, in vivo optrode recording shows that under non-stress conditions, CRHPVN neurons often fire with rhythmic brief bursts (RB), which, somewhat counterintuitively, constrains firing rate due to long (~2 s) interburst intervals. Stressful stimuli rapidly switch RB to continuous single spiking (SS), permitting a large increase in firing rate. A spiking network model shows that recurrent inhibition can control this activity-state switch, and more broadly the gain of spiking responses to excitatory inputs. In biological CRHPVN neurons ex vivo, the injection of whole-cell currents derived from our computational model recreates the in vivo-like switch between RB and SS, providing direct evidence that physiologically relevant network inputs enable state-dependent computation in single neurons. Together, we present a novel mechanism for state-dependent activity dynamics in CRHPVN neurons

    Uso de anti-helmínticos e bioestimulantes no desempenho de bovinos de corte suplementados a pasto no estado do Pará.

    O experimento avaliou o efeito da vermifugação e da utilização de bioestimulantes no ganho de peso e no escore de condição corporal (ECC) de bovinos de corte, criados em sistema de pastejo rotacionado com suplementação a pasto, no Estado do Pará, durante 160 dias. Foram utilizados 132 bovinos machos não castrados, com idade média de 24 meses, da raça Nelore (Bos taurus indicus). Os grupos experimentais compreenderam o grupo G1 (controle; n=33), G2 (moxidectina 1%; n=33), G3 (moxidectina 10%; n=33) e G4 (ivermectina 3,15%; n=33). Em todos os grupos foram estabelecidas três subparcelas, a fim de serem testados dois bioestimulantes de crescimento animal (bioestimulante 1 e bioestimulante 2). Não houve diferença estatística significativa no ganho de peso médio, no ECC e nas contagens de OPG entre animais do G1, G2, G3 e G4, independentemente dos anti-helmínticos e/ou bioestimulantes usados. Contudo, o tratamento baseado na associação de moxidectina 1% e o bioestimulante 2 apresentou maior receita líquida e incrementou a lucratividade da terminação em 1,24%. Os resultados sugerem que não há necessidade de um controle contra nematódeos durante a terminação, desde que os animais apresentem uma baixa carga parasitária, porém o uso de fármacos pode, sob certas condições, apresentar resultado econômico favorável

    "Textural analysis of multiparametric MRI detects transition zone prostate cancer"

    OBJECTIVES: To evaluate multiparametric-MRI (mpMRI) derived histogram textural-analysis parameters for detection of transition zone (TZ) prostatic tumour. METHODS: Sixty-seven consecutive men with suspected prostate cancer underwent 1.5T mpMRI prior to template-mapping-biopsy (TPM). Twenty-six men had 'significant' TZ tumour. Two radiologists in consensus matched TPM to the single axial slice best depicting tumour, or largest TZ diameter for those with benign histology, to define single-slice whole TZ-regions-of-interest (ROIs). Textural-parameter differences between single-slice whole TZ-ROI containing significant tumour versus benign/insignificant tumour were analysed using Mann Whitney U test. Diagnostic accuracy was assessed by receiver operating characteristic area under curve (ROC-AUC) analysis cross-validated with leave-one-out (LOO) analysis. RESULTS: ADC kurtosis was significantly lower (p < 0.001) in TZ containing significant tumour with ROC-AUC 0.80 (LOO-AUC 0.78); the difference became non-significant following exclusion of significant tumour from single-slice whole TZ-ROI (p = 0.23). T1-entropy was significantly lower (p = 0.004) in TZ containing significant tumour with ROC-AUC 0.70 (LOO-AUC 0.66) and was unaffected by excluding significant tumour from TZ-ROI (p = 0.004). Combining these parameters yielded ROC-AUC 0.86 (LOO-AUC 0.83). CONCLUSION: Textural features of the whole prostate TZ can discriminate significant prostatic cancer through reduced kurtosis of the ADC-histogram where significant tumour is included in TZ-ROI and reduced T1 entropy independent of tumour inclusion. KEY POINTS: MR textural features of prostate transition zone may discriminate significant prostatic cancer; Transition zone (TZ) containing significant tumour demonstrates a less peaked ADC histogram; TZ containing significant tumour reveals higher post-contrast T1-weighted homogeneity; The utility of MR texture analysis in prostate cancer merits further investigation
