    In Vitro Effect of Crude Extract from Traganum Nudatum on Glucose-Uptake in Liver Slices Isolated from Westar Rats

    Background: Diabetes mellitus is a metabolic disorder characterized by chronic hyperglycemia resulting from defects in insulin secretion, insulin action, or both. There are many classes of drugs used for treatment, and these include insulin sensitizers, insulin secretagogues, and agents that delay the absorption of carbohydrates from the bowel. This study intends to investigate the effect of crude extract from a plant from South Algeria Traganum nudatum (Chenopodiaceae) on glucose uptake in liver slices isolated from  Wistar rats. Methods: The liver slices were incubated for 90 min at 37° in normoglycaemic (1g/l of glucose)  and hyperglycaemic (3g/l of glucose) KRBA Krebs Ringer Bicarbonate Albumin 4% media using  24 well-polyethylene plates. In each, well different concentrations of insulin (10, 50 and 100µU/ml)  and hydromethanolic crude extract (100, 200 and 500µg/ml) were added. After every 30 minutes, aliquots of the culture media were assayed for the determination of glucose left. Results: Tests showed that the glucose left after 90 minutes in the media which contained insulin at 100µg/ml was the lowest (0.44 and 1.41 )g/l in the normo and hyperglycaemic media respectively, which reflect that insulin at this concentration was the most effective on the stimulation of glucose uptake. The extract had the highest effect at 500µg/ml, the concentrations of glucose left after 90 minutes of incubation were found to be (0.38 and 1.31)g/l in the normoglycaemic and hyperglycaemic media respectively. Conclusion: From the obtained results, it can be concluded that our extract seems to have an insulin-like effect on glucose uptake in liver slices isolated from Wistar rats

    A discrete numerical description of the mechanical response of soils subjected to degradation by suffusion

    Internal erosion is a major cause of the failure of hydraulic earthen structures. A particular case of such an erosion process is suffusion which constitutes a strongly coupled fluid-solid interaction problem. It is a selective erosion of fine particles from an unstable soil structure leaving behind the granular skeleton which possibly leads to deformations. Such a process may cause modification in the mechanical behavior of the soil. To study this problem numerically, a model is established based on the discrete element method implemented in Yade software (Smilauer et al. 2015). Periodic boundary conditions are adopted and the soil is represented by a 3D assembly of spherical discrete elements. Such an oversimplified particle’s shape leads to excessive rolling. To overcome this obstacle, rolling resistance was taken into consideration in the inter-particle contact law. Bearing in mind that numerical modeling of suffusion can constitute a difficult task requiring important computational resources due to the direct description of interactions between solid and liquid phases, a one-way coupling with a fluid phase is considered here. However, effects on the soil due to the loss of a fraction of fine particles is investigated either by modeling soils with different grains size distribution and different initial fines content to characterize its influence on the soil microstructure, or by mimicking the suffusion process by defining an extraction criterion of potentially erodible particles. This extraction criterion is based on the size of the particles, constriction sizes, and the velocity of particles under the effect of fluid forces. From these two approaches, we were able to specify the fines content from which their erosion may have a significant influence on the microstructure. Moreover, the defined extraction criterion was able to describe the effect of erosion on the stability of the soil structure

    Ce travail consiste à concevoir une antenne répondant aux exigences des systèmes Ultra Large Bande ULB. Ces derniers sont très présents dans différentes applications, à titre non exhaustif, nous citerons l’imagerie médicale microonde, les communications sans fil, les systèmes de positionnement et systèmes Radars. Pour cela, l’antenne doit présenter de bonnes performances sur la bande de fréquence 3.1- 10.6 GHz, spectre alloué à l’ULB par la commission FCC (Federal Communications Commission). Nous proposons une antenne imprimée miniature de forme rectangulaire qui satisfait les caractéristiques ULB en termes de bande passante et de coefficient de réflexion. Cette antenne est destinée à un système de détection de tumeurs malignes par imagerie microondes. Nous exploitons certaines techniques de miniaturisation et d’élargissement de la bande passante afin de réaliser notre objectif

    Internal states of model isotropic granular packings. I. Assembling process, geometry and contact networks

    This is the first paper of a series of three, reporting on numerical simulation studies of geometric and mechanical properties of static assemblies of spherical beads under an isotropic pressure. Frictionless systems assemble in the unique random close packing (RCP) state in the low pressure limit if the compression process is fast enough, slower processes inducing traces of crystallization, and exhibit specific properties directly related to isostaticity of the force-carrying structure. The different structures of frictional packings assembled by various methods cannot be classified by the sole density. While lubricated systems approach RCP densities and coordination number z^*~=6 on the backbone in the rigid limit, an idealized "vibration" procedure results in equally dense configurations with z^*~=4.5. Near neighbor correlations on various scales are computed and compared to available laboratory data, although z^* values remain experimentally inaccessible. Low coordination packings have many rattlers (more than 10% of the grains carry no force), which should be accounted for on studying position correlations, and a small proportion of harmless "floppy modes" associated with divalent grains. Frictional packings, however slowly assembled under low pressure, retain a finite level of force indeterminacy, except in the limit of infinite friction.Comment: 29 pages.     Micromechanical features of jet erosion - A numerical perspective

    The erosion mechanisms driving the particle detachment of a soil under an impinging jet are here analysed from a micromechanical perspective combining the numerical Discrete Element and Lattice Boltzmann methods (DEM-LBM). Firstly, the local hydrodynamic conditions of both free and impinging jets are examined and briefly discussed, particularly on the relevance of the superficial irregularities of the granular assembly, which can dominate the local distribution of hydrodynamic variables over the impingement area. Profiles of both macromechanic and discrete (micromechanic) variables will be put forward. Then, the onset of erosion is reviewed from a topological point of view (locality of first grain motion) on the basis of a parametric study. Finally, the variation of the critical inflow velocity for the initiation of erosion in dependence on the particle size is also examined and briefly discussed

    Study of FoxA Pioneer Factor at Silent Genes Reveals Rfx-Repressed Enhancer at Cdx2 and a Potential Indicator of Esophageal Adenocarcinoma Development

    Understanding how silent genes can be competent for activation provides insight into development as well as cellular reprogramming and pathogenesis. We performed genomic location analysis of the pioneer transcription factor FoxA in the adult mouse liver and found that about one-third of the FoxA bound sites are near silent genes, including genes without detectable RNA polymerase II. Virtually all of the FoxA-bound silent sites are within conserved sequences, suggesting possible function. Such sites are enriched in motifs for transcriptional repressors, including for Rfx1 and type II nuclear hormone receptors. We found one such target site at a cryptic “shadow” enhancer 7 kilobases (kb) downstream of the Cdx2 gene, where Rfx1 restricts transcriptional activation by FoxA. The Cdx2 shadow enhancer exhibits a subset of regulatory properties of the upstream Cdx2 promoter region. While Cdx2 is ectopically induced in the early metaplastic condition of Barrett's esophagus, its expression is not necessarily present in progressive Barrett's with dysplasia or adenocarcinoma. By contrast, we find that Rfx1 expression in the esophageal epithelium becomes gradually extinguished during progression to cancer, i.e, expression of Rfx1 decreased markedly in dysplasia and adenocarcinoma. We propose that this decreased expression of Rfx1 could be an indicator of progression from Barrett's esophagus to adenocarcinoma and that similar analyses of other transcription factors bound to silent genes can reveal unanticipated regulatory insights into oncogenic progression and cellular reprogramming

    SIRNA-Directed In Vivo Silencing of Androgen Receptor Inhibits the Growth of Castration-Resistant Prostate Carcinomas

    BACKGROUND: Prostate carcinomas are initially dependent on androgens, and castration or androgen antagonists inhibit their growth. After some time though, tumors become resistant and recur with a poor prognosis. The majority of resistant tumors still expresses a functional androgen receptor (AR), frequently amplified or mutated. METHODOLOGY/PRINCIPAL FINDINGS: To test the hypothesis that AR is not only expressed, but is still a key therapeutic target in advanced carcinomas, we injected siRNA targeting AR into mice bearing exponentially growing castration-resistant tumors. Quantification of siRNA into tumors and mouse tissues demonstrated their efficient uptake. This uptake silenced AR in the prostate, testes and tumors. AR silencing in tumors strongly inhibited their growth, and importantly, also markedly repressed the VEGF production and angiogenesis. CONCLUSIONS/SIGNIFICANCE: Our results demonstrate that carcinomas resistant to hormonal manipulations still depend on the expression of the androgen receptor for their development in vivo. The siRNA-directed silencing of AR, which allows targeting overexpressed as well as mutated isoforms, triggers a strong antitumoral and antiangiogenic effect. siRNA-directed silencing of this key gene in advanced and resistant prostate tumors opens promising new therapeutic perspectives and tools
