    Symbiotic and phenotypic diversity of Rhizobium leguminosarum bv. viciae from Northern Gondar, Ethiopia

    Rhizobia that nodulate cool season legumes are widely spread in the Mediterranean and highland agro ecosystems. Faba bean is one of these important crops that represent the major protein source of human nutrition, cash crop for farmers and component of the farming systems. This study is aimed at identifying rhizobia isolates capable of guarantying efficient nitrogen derived from biological nitrogen fixation. Due to this, a total of twenty-one isolates of Rhizobium were isolated from as many sampling sites of northern Gondar using plant infection method. The isolates were characterized morphologically and physiologically and tested on sand and soils to evaluate their symbiotic effectiveness. Studies on symbiotic effectiveness on sand culture indicated that, the strains showed shoot dry matter ranging from 0.4 (AUFR127) to 2.3 g/plant (AUFR124), with negative control (0.2 g/plant) and positive control of 2.4 g/plant. Eighty percent of the isolates were found to be effective and have very effective nitrogen fixers based on their shoot dry matter accumulation (50 to 100%) in relation to the nitrogen fertilized control plants. Culturally, almost all of them displayed large colonies with diameters of 2 to 4.5 mm, generation time of 1.9 to 4.3 h and showed characteristics of fast growing rhizobia. With a few exceptions, isolates grew at temperatures of 15 and 35°C and were found to be sensitive to salt, except AUFR118 that grew up to 5% NaCl. The isolates also grew on a wide range of moderate acidity and alkalinity (pH5.5-9).With the exception of gluconate, citrate and tartarate, almost all isolates grew on all carbohydrates. The pattern of intrinsic antibiotic resistance showed that, almost all of them were tolerant to chloramphenicol, nalidixic acid and erythromycin. Isolates AUFR118, AUFR128 and AUFR132 were found to be resistant to almost all tested antibiotics. These isolates were also included in the effective and very effective symbiotic groups. The trial of the 5 selected effective isolates on two soil types showed that, they performed well without any significant difference on their respective soils. Interestingly, the isolates were found to nodulate the host on the highly acidic soil (pH 4.8) that failed to be nodulated by the indigenous rhizobia. Generally, the present work shows the physiological and symbiotic diversity of the isolates in the traditional agricultural areas of the region and the potential of these rhizobia to be used as effective commercial inoculants in areas where the indigenous rhizobia fail to do so.     Building Climate Resilience in the Blue Nile/Abay Highlands: A Framework for Action

    Ethiopia has become warmer over the past century and human induced climate change will bring further warming over the next century at unprecedented rates. On the average, climate models show a tendency for higher mean annual rainfall and for wetter conditions, in particular during October, November and December, but there is much uncertainty about the future amount, distribution, timing and intensity of rainfall. Ethiopia’s low level of economic development, combined with its heavy dependence on agriculture and high population growth rate make the country particularly susceptible to the adverse effects of climate change. Nearly 90% of Ethiopia’s population lives in the Highlands, which include the critical Blue Nile (Abay) Highlands—a region that holds special importance due to its role in domestic agricultural production and international water resources. A five year study of climate vulnerability and adaptation strategies in communities of Choke Mountain, located in the center of the Abay Highlands, has informed a proposed framework for enhancing climate resilience in communities across the region. The framework is motivated by the critical need to enhance capacity to cope with climate change and, subsequently, to advance a carbon neutral and climate resilient economy in Ethiopia. The implicit hypothesis in applying a research framework for this effort is that science-based information, generated through improved understanding of impacts and vulnerabilities of local communities, can contribute to enhanced resilience strategies. We view adaptation to climate change in a wider context of changes, including, among others, market conditions, the political-institutional framework, and population dynamics. From a livelihood perspective, culture, historical settings, the diversity of income generation strategies, knowledge, and education are important factors that contribute to adaptive capacities. This paper reviews key findings of the Choke Mountain study, describes the principles of the climate resilience framework, and proposes an implementation strategy for climate resilient development to be applied in the Abay Highlands, with potential expansion to agricultural communities across the region and beyond

    Solar photodegradation of irinotecan in water: optimization and robustness studies by experimental design

    Irinotecan, a widely prescribed anticancer drug, is an emerging contaminant of concern that has been detected in various aquatic environments due to ineffective removal by traditional wastewater treatment systems. Solar photodegradation is a viable approach that can effectively eradicate the drug from aqueous systems. In this study, we used the design of experiment (DOE) approach to explore the robustness of irinotecan photodegradation under simulated solar irradiation. A full factorial design, including a star design, was applied to study the effects of three parameters: initial concentration of irinotecan (1.0-9.0 mg/L), pH (5.0-9.0), and irradiance (450-750 W/m(2)). A high-performance liquid chromatography coupled with a high-resolution mass spectrometry (HPLC-HRMS) system was used to determine irinotecan and identify transformation products. The photodegradation of irinotecan followed a pseudo-first order kinetics. In the best-fitted linear model determined by the stepwise model fitting approach, pH was found to have about 100-fold greater effect than either irinotecan concentration or solar irradiance. Under optimal conditions (irradiance of 750 W/m(2), 1.0 mg/L irinotecan concentration, and pH 9.0), more than 98% of irinotecan was degraded in 60 min. With respect to irradiance and irinotecan concentration, the degradation process was robust in the studied range, implying that it may be effectively applied in locations and/or seasons with solar irradiance as low as 450 W/m(2). However, pH needs to be strictly controlled and kept between 7.0 and 9.0 to maintain the degradation process robust. Considerations about the behavior of degradation products were also drawn

The report assesses the relationship between urban form and function and the spread of the COVID-19 pandemic, in Addis Ababa. It explores what is meant by a healthy city, and why planning for and investing in a healthy city, matters to Addis Ababa. It goes on to investigate the state of health, urban infrastructure and social services in the city. The socio-economic and health impacts of the pandemic are also explored further, together with the institutional response to the public health emergency. The findings provide insights on the role of urban form and infrastructure to urban health and urban resilience. Finally, the authors highlight a post-pandemic agenda for a healthier, more resilient city

    Human Prion Diseases in the United States

    BACKGROUND: Prion diseases are a family of rare, progressive, neurodegenerative disorders that affect humans and animals. The most common form of human prion disease, Creutzfeldt-Jakob disease (CJD), occurs worldwide. Variant CJD (vCJD), a recently emerged human prion disease, is a zoonotic foodborne disorder that occurs almost exclusively in countries with outbreaks of bovine spongiform encephalopathy. This study describes the occurrence and epidemiology of CJD and vCJD in the United States. METHODOLOGY/PRINCIPAL FINDINGS: Analysis of CJD and vCJD deaths using death certificates of US residents for 1979-2006, and those identified through other surveillance mechanisms during 1996-2008. Since CJD is invariably fatal and illness duration is usually less than one year, the CJD incidence is estimated as the death rate. During 1979 through 2006, an estimated 6,917 deaths with CJD as a cause of death were reported in the United States, an annual average of approximately 247 deaths (range 172-304 deaths). The average annual age-adjusted incidence for CJD was 0.97 per 1,000,000 persons. Most (61.8%) of the CJD deaths occurred among persons >or=65 years of age for an average annual incidence of 4.8 per 1,000,000 persons in this population. Most deaths were among whites (94.6%); the age-adjusted incidence for whites was 2.7 times higher than that for blacks (1.04 and 0.40, respectively). Three patients who died since 2004 were reported with vCJD; epidemiologic evidence indicated that their infection was acquired outside of the United States. CONCLUSION/SIGNIFICANCE: Surveillance continues to show an annual CJD incidence rate of about 1 case per 1,000,000 persons and marked differences in CJD rates by age and race in the United States. Ongoing surveillance remains important for monitoring the stability of the CJD incidence rates, and detecting occurrences of vCJD and possibly other novel prion diseases in the United States

    Changing Patterns of Tree Cover in a Tropical Highland Region and Implications for Food, Energy, and Water Resources

    The Blue Nile Highlands of Ethiopia are a densely populated, predominantly rural region dominated by smallholder crop-livestock mixed farming systems. Population growth, coupled with low productivity, have long posed a threat to natural forest ecosystems in the region, as trees have been removed for fuelwood and to clear area for grazing or crop production. In recent years, however, there has been a trend to replace cropland with eucalyptus plantations. This change has major implications for the hydrology, soils, and agricultural economy of the region. This study examines changes in tree cover for a highland area at the center of the Blue Nile Highlands. Landsat imagery from 1986 to 2017 is applied to characterize changing tree cover patterns over space and time. We find that total tree cover in this highland region has shifted dramatically over the past 30 years. Between 1987 and 1999 there was dramatic loss of tree cover, particularly in areas of natural vegetation at high and low elevation. This period coincided with the fall of the Derg government and the transition to the current political system. In the period since 1999 there has been an increase in tree cover, with rapid gains in recent years. This increase has taken two distinct forms: regrowth in previously forested areas, due in part to active conservation measures, and the establishment of eucalyptus plantations in mid-elevation zones. The ecological and economic implications of these two types of tree cover—protected forest vs. woodlot plantations—are quite distinct, with plantation forestry providing biomass energy at a cost to food production and water resources. Mapping cropland conversion to eucalyptus in recent years makes it possible to quantify the net impacts that this trend has had on local production of energy and food, and to estimate implications for water consumption. Effective monitoring of these changes is important for the ongoing development and implementation of effective land use policy in the region

    Cellular and Cytokine Responses in the Granulomas of Asymptomatic Cattle naturally infected with Mycobacterium bovis in Ethiopia

    Cells (CD3+ T cell and CD68+ macrophages), cytokines (IFN-γ+ and TNF-α+) and effector molecule (iNOS+) responses were evaluated in the lymph nodes and tissue of cattle naturally infected with Detailed post mortem and immunohistochemical examinations of lesions were performed on 16 cows positive for single intradermal cervical comparative tuberculin (SICCT) test which were identified from dairy farms located around the Addis Ababa City. The severity of the gross lesion was significantly higher (p=0.003) in culture positive (n=12) cows than in culture negative (n=4). Immunohistochemical techniques showed that in culture positive cows, the mean immunolabeling fraction of CD3+ T cells decreased as the stage of granuloma increased from stage I to stage IV (p<0.001). In contrast, the immunolabelling fraction of CD68+ macrophages, IFN-γ+, TNF-α+ and iNOS+ increased from stage I to stage IV (p< 0.001). In culture negative cows, early stages showed a significantly higher fraction of CD68+ macrophages (p=0.03) and iNOS+ (p=0.007) when compared to culture positive cows. Similarly, at advanced granuloma stages, culture negative cows demonstrated significantly higher mean proportions of CD3+ T cells (p< 0.001) compared to culture positive cows. Thus, this study demonstrates that following natural infection of cows with , as the stage of granuloma increases from stage I to stage IV, the immunolabelling fraction of CD3+ cells decreases while the immunolabeling fraction of CD68+ macrophages, IFN-γ+, TNF-α+ and iNOS+ increases.     Synthesis and Characterization of Tetracycline Loaded Methionine-Coated NiFe2O4 Nanoparticles for Anticancer and Antibacterial Applications

    In the present study, nickel ferrite (NiFe(2)O(4))-based smart magnetic nanoparticles were fabricated and coated with methionine. Physiochemical characterization of the obtained Met-NiFe(2)O(4) nanoparticles revealed the presence of methionine coating over the nanoparticle surface. Drug release study indicated that Tet-Met-NiFe(2)O(4) nanoparticles possess pH-responsive controlled drug release behavior for tetracycline (Tet). The drug loading content for Tet was found to be 0.27 mg/L of nanoparticles. In vitro cytotoxicity test showed that the Met-NiFe(2)O(4) nanoparticles is biocompatible. Moreover, this magnetic nanostructured material shown strong anticancer property as these nanomaterials significantly reduced the viability of A375 cells when compared to free Tet solution. In addition, Tet-Met-NiFe(2)O(4) nanoparticles also showed strong antibacterial activity against different bacterial pathogens

    Chronic wasting disease prions are not transmissible to transgenic mice overexpressing human prion protein

    Chronic wasting disease (CWD) is a prion disease that affects free-ranging and captive cervids, including mule deer, white-tailed deer, Rocky Mountain elk and moose. CWD-infected cervids have been reported in 14 USA states, two Canadian provinces and in South Korea. The possibility of a zoonotic transmission of CWD prions via diet is of particular concern in North America where hunting of cervids is a popular sport. To investigate the potential public health risks posed by CWD prions, we have investigated whether intracerebral inoculation of brain and spinal cord from CWD-infected mule deer transmits prion infection to transgenic mice overexpressing human prion protein with methionine or valine at polymorphic residue 129. These transgenic mice have been utilized in extensive transmission studies of human and animal prion disease and are susceptible to BSE and vCJD prions, allowing comparison with CWD. Here, we show that these mice proved entirely resistant to infection with mule deer CWD prions arguing that the transmission barrier associated with this prion strain/host combination is greater than that observed with classical BSE prions. However, it is possible that CWD may be caused by multiple prion strains. Further studies will be required to evaluate the transmission properties of distinct cervid prion strains as they are characterized
