    Structural Evidence of a Major Conformational Change Triggered by Substrate Binding in DapE Enzymes: Impact on the Catalytic Mechanism

    The X-ray crystal structure of the dapE-encoded N-succinyl-l,l-diaminopimelic acid desuccinylase from Haemophilus influenzae (HiDapE) bound by the products of hydrolysis, succinic acid and l,l-DAP, was determined at 1.95 Å. Surprisingly, the structure bound to the products revealed that HiDapE undergoes a significant conformational change in which the catalytic domain rotates ∼50° and shifts ∼10.1 Å (as measured at the position of the Zn atoms) relative to the dimerization domain. This heretofore unobserved closed conformation revealed significant movements within the catalytic domain compared to that of wild-type HiDapE, which results in effectively closing off access to the dinuclear Zn(II) active site with the succinate carboxylate moiety bridging the dinculear Zn(II) cluster in a μ-1,3 fashion forming a bis(μ-carboxylato)dizinc(II) core with a Zn–Zn distance of 3.8 Å. Surprisingly, His194.B, which is located on the dimerization domain of the opposing chain ∼10.1 Å from the dinuclear Zn(II) active site, forms a hydrogen bond (2.9 Å) with the oxygen atom of succinic acid bound to Zn2, forming an oxyanion hole. As the closed structure forms upon substrate binding, the movement of His194.B by more than ∼10 Å is critical, based on site-directed mutagenesis data, for activation of the scissile carbonyl carbon of the substrate for nucleophilic attack by a hydroxide nucleophile. Employing the HiDapE product-bound structure as the starting point, a reverse engineering approach called product-based transition-state modeling provided structural models for each major catalytic step. These data provide insight into the catalytic reaction mechanism and also the future design of new, potent inhibitors of DapE enzymes

    What Determines the Depth of BALs? Keck HIRES Observations of BALQSO 1603+300

    We find that the depth and shape of the broad absorption lines (BALs) in BALQSO 1603+3002 are determined largely by the fraction of the emitting source which is covered by the BAL flow. In addition, the observed depth of the BALs is poorly correlated with their real optical depth. The implication of this result is that abundance studies based on direct extraction of column densities from the depth of the absorption troughs are unreliable. Our conclusion is based on analysis of unblended absorption features of two lines from the same ion (in this case the Si IV doublet), which allows unambiguous separation of covering factor and optical depth effects. The complex morphology of the covering factor as a function of velocity suggests that the BALs are produced by several physically separated outflows. The covering factor is ion dependent in both depth and velocity width.     The Properties of Radio Galaxies and the Effect of Environment in Large Scale Structures at z1z\sim1

    In this study we investigate 89 radio galaxies that are spectroscopically-confirmed to be members of five large scale structures in the redshift range of 0.65z0.960.65 \le z \le 0.96. Based on a two-stage classification scheme, the radio galaxies are classified into three sub-classes: active galactic nucleus (AGN), hybrid, and star-forming galaxy (SFG). We study the properties of the three radio sub-classes and their global and local environmental preferences. We find AGN hosts are the most massive population and exhibit quiescence in their star-formation activity. The SFG population has a comparable stellar mass to those hosting a radio AGN but are unequivocally powered by star formation. Hybrids, though selected as an intermediate population in our classification scheme, were found in almost all analyses to be a unique type of radio galaxies rather than a mixture of AGN and SFGs. They are dominated by a high-excitation radio galaxy (HERG) population. We discuss environmental effects and scenarios for each sub-class. AGN tend to be preferentially located in locally dense environments and in the cores of clusters/groups, with these preferences persisting when comparing to galaxies of similar colour and stellar mass, suggesting that their activity may be ignited in the cluster/group virialized core regions. Conversely, SFGs exhibit a strong preference for intermediate-density global environments, suggesting that dusty starbursting activity in LSSs is largely driven by galaxy-galaxy interactions and merging.Comment: 28 pages, 10 figures, accepted to MNRA

    “Dermal Dendritic Cells” Comprise Two Distinct Populations: CD1+ Dendritic Cells and CD209+ Macrophages

    A key cell type of the resident skin immune system is the dendritic cell (DC), which in normal skin is located in two distinct microanatomical compartments: Langerhans cells (LCs), mainly in the epidermis, and dermal DCs (DDCs), in the dermis. Here, the lineage of DDCs was investigated using monoclonal antibodies and immunohistology. We provide evidence that “DDC” comprise at least two major phenotypic populations of dendritic-appearing cells, immature DC expressing CD1, CD11c and CD208; and macrophages expressing CD209, CD206, CD163, and CD68. These data suggest that dermal dendritic-appearing macrophages comprise a novel part of the innate immune response in the resident skin immune system

    Extracorporeal membrane oxygenation (ECMO) as salvage treatment for pulmonary Echinococcus granulosus infection with acute cyst rupture

    Extracorporeal membrane oxygenation (ECMO) has been used successfully for the treatment of patients with respiratory failure due to severe infections. Although rare, parasites can also cause severe pulmonary disease. Tapeworms of the genus Echinococcus give rise to the development of cystic structures in the liver, lungs, and other organs. Acute cyst rupture leads to potentially life-threatening infection, and affected patients may deteriorate rapidly. The case of a young woman from Bulgaria who was admitted to hospital with severe dyspnoea, progressive chest pain, and haemoptysis is described. Computed tomography of the chest was pathognomonic for cystic echinococcosis with acute cyst rupture. Following deterioration on mechanical ventilation, she was cannulated for veno-venous ECMO. The patient's condition improved considerably, and she was weaned successfully from ECMO and mechanical ventilation. Following lobectomy of the affected left lower lobe, the patient was discharged home in good condition. This appears to be the first report of the successful use of ECMO as salvage treatment for a severe manifestation of a helminthic disease. Due to recent migration to Western Europe, the number of patients presenting with respiratory failure due to pulmonary echinococcosis with cyst rupture is likely to increase

    Initial Development of a Patient-Reported Instrument Assessing Harm, Efficacy, and Misuse of Long-Term Opioid Therapy

    Guidelines on long-term opioid therapy recommend frequent reassessment of harm, efficacy, and misuse of these potentially harmful and sometimes ineffective medications. In primary care, there is a need for a brief, patient-reported instrument. This report details the initial steps in the development of such an instrument. An interdisciplinary team of clinician-scientists performed four discrete steps in this study: (1) conceptualization of the purpose and function of the instrument, (2) assembly of an item pool, (3) expert rating on which items were most important to include in the instrument, and (4) modification of expert-selected items based on a reading level check and cognitive interviews with patients. A diverse panel of 47 subject matter experts was presented with 69 items to rate on a 1–9 scale in terms of importance for inclusion in the instrument. The panel highly rated 37 items: 8 related to harm, 4 related to efficacy, and 25 related to misuse. These 37 items were then tested for patient comprehension and modified as needed. Next steps in development will include further item reduction, testing against a gold standard, and assessment of the instrument’s effect on clinical outcomes
