    Identification of sites phosphorylated by the vaccinia virus B1R kinase in viral protein H5R

    Background: Vaccinia virus gene B1R encodes a erine/threonine protein kinase. In vitro this protein kinase phosphorylates ribosomal proteins Sa and S2 and vaccinia virus protein H5R, proteins that become phosphorylated during infection. Nothing is known about the sites phosphorylated on these proteins or the general substrate specificity of the kinase. The work described is the first to address these questions. Results: Vaccinia virus protein H5R was phosphorylated by the B1R protein kinase in vitro, digested with V8 protease, and phosphopeptides separated by HPLC. The N-terminal sequence of one radioactively labelled phosphopeptide was determined and found to correspond to residues 81-87 of the protein, with Thr-84 and Thr-85 being phosphorylated. A synthetic peptide based on this region of the protein was shown to be a substrate for the B1R protein kinase, and the extent of phosphorylation was substantially decreased if either Thr residue was replaced by an Ala. Conclusions: We have identified the first phosphorylation site for the vaccinia virus B1R protein kinase. This gives important information about the substrate-specificity of the enzyme, which differs from that of other known protein kinases. It remains to be seen whether the same site is phosphorylated in vivo

    With resources at a premium, great attention is being paid to persuasive methods and convincing stakeholders to support and advocate for archival and library needs. Digital libraries, like others, are not immune to the seemingly perpetual budget cuts across our field. However, using stories and statistics to illustrate our impact can help advocate for our needs and preserve our digital history. This session will focus on compiling qualitative stories from our patrons with quantifiable statistics to advocate for digital preservation needs. Using Digital Collections at the University of Southern Mississippi as a case study, this session will show examples of patron stories and measurable statistics to advocate for a digital preservation system and larger data preservation needs at the university. While the example is academic, the overall framework is adaptable to all settings

    Digital Preservation Efforts at USM

    The University of Southern Mississippi (USM) has steadily been working to improve its digital preservation infrastructure over the past four years. In 2013, with funding from a NEH Preservation Assistance Grant, consultants Tom Clareson and Liz Bishoff conducted a digital preservation readiness assessment and jump started USM’s education on the topic. Since then, USM has added geographically distributed backups, manual fixity checks, manual metadata logs, and manual file format migrations to its arsenal with varying degrees of success. The influx in needed manpower and technical infrastructure precipitated a financial commitment from the university and the purchase of a robust digital preservation system. Using the National Digital Stewardship Alliance’s (NDSA) Levels of Digital Preservation as a guide, this session will follow the progression of USM’s digital preservation efforts over the past four years from education to implementation. Attendees will learn how to evaluate an institution’s digital preservation readiness using the NDSA’s Levels of Digital Preservation, and come away with ideas for lost cost, high impact ways to improve their own digital preservation efforts
