Generation of multipotent foregut stem cells from human pluripotent stem cells

Human pluripotent stem cells (hPSCs) could provide an infinite source of clinically relevant cells with potential applications in regenerative medicine. However, hPSC lines vary in their capacity to generate specialized cells, and the development of universal protocols for the production of tissue-specific cells remains a major challenge. Here, we have addressed this limitation for the endodermal lineage by developing a defined culture system to expand and differentiate human foregut stem cells (hFSCs) derived from hPSCs. hFSCs can self-renew while maintaining their capacity to differentiate into pancreatic and hepatic cells. Furthermore, near-homogenous populations of hFSCs can be obtained from hPSC lines which are normally refractory to endodermal differentiation. Therefore, hFSCs provide a unique approach to bypass variability between pluripotent lines in order to obtain a sustainable source of multipotent endoderm stem cells for basic studies and to produce a diversity of endodermal derivatives with a clinical value

The Universidad Politécnica of Madrid (UPM) includes schools and faculties that were for engineering degrees, architecture and computer science, that are now in a quick EEES Bolonia Plan metamorphosis getting into degrees, masters and doctorate structures. They are focused towards action in machines, constructions, enterprises, that are subjected to machines, human and environment created risks. These are present in actions such as use loads, wind, snow, waves, flows, earthquakes, forces and effects in machines, vehicles behavior, chemical effects, and other environmental factors including effects of crops, cattle and beasts, forests, and varied essential economic and social disturbances. Emphasis is for authors in this session more about risks of natural origin, such as for hail, winds, snow or waves that are not exactly known a priori, but that are often considered with statistical expected distributions giving extreme values for convenient return periods. These distributions are known from measures in time, statistic of extremes and models about hazard scenarios and about responses of man made constructions or devices. In each engineering field theories were built about hazards scenarios and how to cover for important risks. Engineers must get that the systems they handle, such as vehicles, machines, firms or agro lands or forests, obtain production with enough safety for persons and with decent economic results in spite of risks. For that risks must be considered in planning, in realization and in operation, and safety margins must be taken but at a reasonable cost. That is a small level of risks will often remain, due to limitations in costs or because of due to strange hazards, and maybe they will be covered by insurance in cases such as in transport with cars, ships or aircrafts, in agro for hail, or for fire in houses or in forests. These and other decisions about quality, security for men or about business financial risks are sometimes considered with Decision Theories models, using often tools from Statistics or operational Research. The authors have done and are following field surveys about risk consideration in the careers in UPM, making deep analysis of curricula taking into account the new structures of degrees in the EEES Bolonia Plan, and they have considered the risk structures offered by diverse schools of Decision theories. That gives an aspect of the needs and uses, and recommendations about improving in the teaching about risk, that may include special subjects especially oriented for each career, school or faculty, so as to be recommended to be included into the curricula, including an elaboration and presentation format using a multi-criteria decision model

Trimeric Antp-TBP complexes with TFIIEβ and Exd modulated the transcriptional activity of Antp

Homeoproteins are transcription factors that bind to DNA through a highly conserved binding domain known as the homeodomain (HD), which recognizes short regions rich in A-T. This DNA binding triggers the modulation of multiple target genes responsible for identity in various structures along the A-P axis of D. melanogaster. However, the high similarity of homeodomains and their affinity for widely distributed sites in the genome poses the Hox paradox, which seeks to explain how homeoproteins acquire functional specificity so precisely. Previous evidence shows that homeoproteins like Antennapedia (Antp) form dimers with cofactors and other transcription factors to modulate their activity and also can form trimeric complexes with TBP/TFIIEβ/Exd. For this reason, we have conducted transactivation analyses to determine the effect of Antp-TBP-TFIIEβ and Antp-TBP-Exd complexes on Antp\u27s transcriptional activity. The experimental strategy involved transfecting HEK-293 cells with different combinations of plasmids producing Antp, TBP, TFIIEβ, and Exd, along with two reporter plasmids: a luciferase reporter with 5 binding sites for Antp and the sequence of a D. melanogaster basal promoter, and a β-galactosidase reporter. Our results reveal that the trimeric interaction of Antp-TBP-TFIIEβ induces a highly significant increase of 173% in Antp transcriptional activity. On the other hand, Antp-TBP-Exd trimer shows opposite results by decreasing Antp transcriptional activity until 61%. These findings suggest that the trimeric complexes Antp-TBP/TFIIEβ/Exd play a role in modulating Antp\u27s transcriptional activity, raising new questions about the in vivo effects of these complexes on Antp interactome and target genes

[EN] In this paper, we investigated the monitoring and characterization of the pest Magnaporthe oryzae, known as rice blast, in the Bomba rice variety at the Albufera Natural Park, located in Valencia, Spain during the 2022 and 2023 seasons. Using reflectance data from different Sentinel-2 satellite bands, various vegetative indices were calculated for each year. Significant differences in reflectance in the visible (B4), infrared (B8), red-edge (B6 and B7), and SWIR (B11) bands were detected between healthy and unhealthy fields. Additionally, variations were observed in the vegetation indices, with RVI and IRECI standing out for their higher accuracy in identifying blast-affected plots compared to NDVI and NDRE. Early differences in band values, vegetative indices, and spectral signatures were observed between the unhealthy and healthy plots, allowing for the anticipation of control treatments, whose effectiveness relies on timely intervention. This research has been funded by the DETECTORYZA project INNEST/2022/227, INNEST/2022/319 and INNEST/2022/361 Regional Operational Programme, FEDER Comunitat Valenciana de la Innovacio, Generalitat Valenciana.Agenjos-Moreno, A.; Rubio Michavila, C.; Uris Martínez, A.; Simeon-Brocal, R.; Franch-Gras, B.; Domingo Carrasco, C.; San Bautista Primo, A. Get PDF
Objective To study socio-demographics, sexual practices, drug use behaviors, and prevalences of HIV, syphilis, hepatitis B and C, HTLV-1 and HTLV-2 in immigrant (foreigner) and non-immigrant (local/native) female sex workers (FSW). Design This was a cross-sectional study in immigrant and non-immigrant FSW living in Buenos Aires, Argentina. Participants were interviewed using a standardized questionnaire. Results A total of 625 FSW were enrolled, of whom 169 (27%) were immigrant FSW from Paraguay, the Dominican Republic, Brazil, Peru, and Uruguay. The prevalence of syphilis and hepatitis C was significantly higher among Argentinean FSW than among immigrant FSW. However, hepatitis B prevalence was higher among immigrant FSW. Adjusted risk factor analysis comparing immigrant FSW with Argentinean FSW indicated that marital status (single), occupation (none), fee per sex act (≤US$7), workplace (bar and cabaret), and anal sex with clients were significantly associated with immigrant FSW status. Conclusions Effective HIV/STI prevention and medical care programs need to be tailored to the specific needs of both FSW groups in Argentina.Fil: Bautista, Christian T.. Dengue virus NS1 protein interacts with the ribosomal protein RPL18: This interaction is required for viral translation and replication in Huh-7 cells

    Get PDF
AbstractGiven dengue virus (DENV) genome austerity, it uses cellular molecules and structures for virion entry, translation and replication of the genome. NS1 is a multifunctional protein key to viral replication and pathogenesis. Identification of cellular proteins that interact with NS1 may help in further understanding the functions of NS1. In this paper we isolated a total of 64 proteins from DENV infected human hepatic cells (Huh-7) that interact with NS1 by affinity chromatography and immunoprecipitation assays. The subcellular location and expression levels during infection of the ribosomal proteins RPS3a, RPL7, RPL18, RPL18a plus GAPDH were determined. None of these proteins changed their expression levels during infection; however, RPL-18 was redistributed to the perinuclear region after 48hpi. Silencing of the RPL-18 does not affect cell translation efficiency or viability, but it reduces significantly viral translation, replication and viral yield, suggesting that the RPL-18 is required during DENV replicative cycle
