    Pre-steady state electrogenic events of Ca2+/H+ exchange and transport by the Ca2+-ATPase.

    Native or recombinant SERCA (sarco(endo)plasmic reticulum Ca(2+) ATPase) was adsorbed on a solid supported membrane and then activated with Ca(2+) and ATP concentration jumps through rapid solution exchange. The resulting electrogenic events were recorded as electrical currents flowing along the external circuit. Current transients were observed following Ca(2+) jumps in the absence of ATP and following ATP jumps in the presence of Ca(2+). The related charge movements are attributed to Ca(2+) reaching its binding sites in the ground state of the enzyme (E(1)) and to its vectorial release from the enzyme phosphorylated by ATP (E(2)P). The Ca(2+) concentration and pH dependence as well as the time frames of the observed current transients are consistent with equilibrium and pre-steady state biochemical measurements of sequential steps within a single enzymatic cycle. Numerical integration of the current transients recorded at various pH values reveal partial charge compensation by H(+) in exchange for Ca(2+) at acidic (but not at alkaline) pH. Most interestingly, charge movements induced by Ca(2+) and ATP vary over different pH ranges, as the protonation probability of residues involved in Ca(2+)/H(+) exchange is lower in the E(1) than in the E(2)P state. Our single cycle measurements demonstrate that this difference contributes directly to the reduction of Ca(2+) affinity produced by ATP utilization and results in the countertransport of two Ca(2+) and two H(+) within each ATPase cycle at pH 7.0. The effects of site-directed mutations indicate that Glu-771 and Asp-800, within the Ca(2+) binding domain, are involved in the observed Ca(2+)/H(+) exchange

    Maxillary arch development with Invisalign system: Analysis of expansion dental movements on digital dental casts

    Objectives: To evaluate tooth movements during maxillary arch expansion with clear aligner treatment.Materials and Methods: The study group included 28 subjects (16 females, 12 males, mean age 31.9 +/- 5.4 years) collected prospectively from January 2018 to May 2019. Inclusion criteria were European ancestry, posterior transverse discrepancy of 3-6 mm, permanent dentition stage, presence of second permanent molars, mild or moderate crowding, and good compliance with aligners. Treatment protocol included nonextraction strategies, application of Invisalign clear aligner system, and no auxiliaries other than Invisalign attachments. Linear and angular measurements were performed before treatment (T1), at the end of treatment (T2), and on final virtual models (T2 ClinCheck). A paired t-test was used to compare T2-T1 and T2-T2 ClinCheck changes. The level of significance was set at 5%.Results: Statistically significant differences were found for all measurements, except for ones at the upper second molars. The greatest increase in maxillary width was detected at the upper first and second premolars: 13.5 mm for the first premolar and 13.8 mm for the second premolar at T2. Comparison of T2-T1 angular outcomes showed statistically significant changes in the inclinations of all teeth except for the second permanent molars. T2-T2 ClinCheck showed significant differences for both linear and angular measurements for maxillary canines, resulting in poor predictability.Conclusions: Maxillary arch development revealed a progressive reduction of the expansion rate and buccal tipping in the anterior, lateral, and posterior regions, with the greatest net increase at the first and second premolars. Clinical attention should be paid to maxillary canine movements, and overcorrection should be planned for them during dentoalveolar expansion

    Clotrimazole (CLT) is an antimycotic imidazole derivative that is known to inhibit cytochrome P-450, ergosterol biosynthesis and proliferation of cells in culture, and to interfere with cellular Ca(2+) homeostasis. We found that CLT inhibits the Ca(2+)-ATPase of rabbit fast-twitch skeletal muscle (SERCA1), and we characterized in detail the effect of CLT on this calcium transport ATPase. We used biochemical methods for characterization of the ATPase and its partial reactions, and we also performed measurements of charge movements following adsorption of sarcoplasmic reticulum vesicles containing the ATPase onto a gold-supported biomimetic membrane. CLT inhibits Ca(2+)-ATPase and Ca(2+) transport with a K(I) of 35 mum. Ca(2+) binding in the absence of ATP and phosphoenzyme formation by the utilization of ATP in the presence of Ca(2+) are also inhibited within the same CLT concentration range. On the other hand, phosphoenzyme formation by utilization of P(i) in the absence of Ca(2+) is only minimally inhibited. It is concluded that CLT inhibits primarily Ca(2+) binding and, consequently, the Ca(2+)-dependent reactions of the SERCA cycle. It is suggested that CLT resides within the membrane-bound region of the transport ATPase, thereby interfering with binding and the conformational effects of the activating cation

    This work brings together the results of a series of researches on the importance of the visual communication for nocturnal birds, carried out using the Eagle Owl as a model species. We analysed the main characteristics of the white feathers of the adults and the offspring and we tested the function of the achromatic plumage in territorial behaviour and parental care. Furthermore, by means of radio-tracking, we monitored the hunting activity of the Eagle Owl and, by analysing the white tail of its main prey - the Rabbit - we highlighted the possible role of the visual communication for nocturnal birds also in the contest of the predator-prey interactions

    Spatial behaviour of yellow-necked wood mouse Apodemus flavicollis in two sub-Mediterranean oak coppice stands

    Strong mutual relationships exist between rodents and ecosystems. By modifying the structure and functioning of ecosystems, human activity can affect rodent behaviour and ecology. The yellow-necked mouse Apodemus flavicollis is widespread in Europe and its spatial ecology has been studied in various habitats, but studies are lacking for Mediterranean forests often altered by forest management practices. We investigated the spatial behaviour of A. flavicollis in a sub-Mediterranean deciduous oak forest of central Italy subject to forest management. We radio-tracked 27 individuals in two different coppice stands, i.e. a recently cut area and a high forest, differing in terms of species-specific habitat quality and rodent population density. We analysed the size of home ranges in relation to habitat type and sex. Our results revealed that home range and core area size did not differ between habitat types or sexes. The spatial behaviour of A. flavicollis thus did not appear to be influenced by population density and habitat quality. The lack of sex-related differences confirms the current knowledge on the species’ spatial ecology. Our findings provide the first useful information on the spatial behaviour of A. flavicollis in sub-Mediterranean deciduous oak forests, whose ecological processes may be markedly influenced by this key-species at several trophic levels.acceptedVersio

    Ziprasidone as Adjunctive Therapy in Severe Bipolar Patients Treated with Clozapine

    Aim. To confirm the efficacy and tolerability of ziprasidone as adjunctive therapy in bipolar patients partially responding to clozapine or with persisting negative symptoms, overweight, or with metabolic syndrome. Methods. Eight patients with psychotic bipolar disorder were tested with the BPRS, the HAM-D, and the CGI at T0 and retested after 2 weeks (T1). Plasma clozapine and norclozapine levels and BMI were tested at T0 and T1. Results. Ziprasidone was well tolerated by all the patients. BPRS and HAM-D scores were reduced in all patients. BMI was reduced in patients with a BMI at T0 higher than 25. Plasma levels of clozapine and norclozapine showed an irregular course

    A framework for prioritising present and potentially invasive mammal species for a national list

    The European Union (EU) has recently adopted a regulation on invasive alien species that foresees the possibility of developing lists of species of National Concern. We developed a prioritisation process for alien mammals already established in Italy, but not yet included in the EU list (n = 6 species) and a systematic horizon-scanning procedure to obtain ranked lists for those species that are already introduced worldwide or traded in Italy (n = 213). Experts were asked to score these species, by evaluating their likelihood of establishment and spread and the magnitude of their potential impacts on biodiversity, economy, human-health and society. The manageability of each species was also evaluated, both for the proritisation and the horizon-scanning processes. We produced five lists that ranked species according to their potential spread and impacts and their manageability. These will allow policy-makers to select outputs according to a balance between risk assessment and risk management, establishing priorities for alien species management at the national level
