    An evaluation of a constrained test method for obtaining free body responses

    A method for obtaining free body responses from dynamic tests on a constrained structure is investigated for practical feasibility. The method is based on the principle that a constrained structure can be considered to be a free body acted upon by multiple forces which include the forces of constraint. By measuring these forces and by exciting the structure so as to develop linearly independent sets of forces, the response of the free body to one force at a time can be computed. Techniques for producing these independent forces are discussed. The development of the theory, computer simulations of tests of representative aerospace vehicles (including experimental error), and a description and listing of the computer programs developed are included. The procedure appears to be a practical method for obtaining in-flight characteristics of such vehicles

    The development of B-1 aircraft rigid wind tunnel data for use in subsequent tasks of the Airloads Research Study is described. Data from the Rockwell International external structural loads data bank were used to generate coefficients of rigid airload shear, bending moment, and torsion at specific component reference stations or both symmetric and asymmetric loadings. Component stations include the movable wing, horizontal and vertical stabilizers, and forward and aft fuselages. The coefficient data cover a Mach number range from 0.7 to 2.2 for a wing sweep position of 67.5 degree

    The acquisition of B-1 aircraft flight loads data for use in subsequent tasks of the Airloads Research Study is described. The basic intent is to utilize data acquired during B-1 aircraft tests, analyze these data beyond the scope of Air Force requirements, and prepare research reports that will add to the technology base for future large flexible aircraft. Flight test data obtained during the airloads survey program included condition-describing parameters, surface pressures, strain gage outputs, and loads derived from pressure and strain gauges. Descriptions of the instrumentation, data processing, and flight load survey program are included.     Challenges of Profile Likelihood Evaluation in Multi-Dimensional SUSY Scans

    Statistical inference of the fundamental parameters of supersymmetric theories is a challenging and active endeavor. Several sophisticated algorithms have been employed to this end. While Markov-Chain Monte Carlo (MCMC) and nested sampling techniques are geared towards Bayesian inference, they have also been used to estimate frequentist confidence intervals based on the profile likelihood ratio. We investigate the performance and appropriate configuration of MultiNest, a nested sampling based algorithm, when used for profile likelihood-based analyses both on toy models and on the parameter space of the Constrained MSSM. We find that while the standard configuration is appropriate for an accurate reconstruction of the Bayesian posterior, the profile likelihood is poorly approximated. We identify a more appropriate MultiNest configuration for profile likelihood analyses, which gives an excellent exploration of the profile likelihood (albeit at a larger computational cost), including the identification of the global maximum likelihood value. We conclude that with the appropriate configuration MultiNest is a suitable tool for profile likelihood studies, indicating previous claims to the contrary are not well founded.Comment: 21 pages, 9 figures, 1 table; minor changes following referee report.     On a connection between factor analysis and multidimensional unfolding

    Given the preference ordering of each of a number of individuals over a set of stimuli, it is proposed that if the preference orderings are generated in a Euclidean space of r dimensions which can be recovered by unfolding the preference orderings, then a factor analysis of the correlations between individual's preference orderings will yield a space of r + 1 dimensions with the original r -space embedded in it, and the additional dimension will be one of social utility.     Photobacterium profundum under Pressure:A MS-Based Label-Free Quantitative Proteomics Study

    Photobacterium profundum SS9 is a Gram-negative bacterium, originally collected from the Sulu Sea. Its genome consists of two chromosomes and a 80 kb plasmid. Although it can grow under a wide range of pressures, P. profundum grows optimally at 28 MPa and 15°C. Its ability to grow at atmospheric pressure allows for both easy genetic manipulation and culture, making it a model organism to study piezophily. Here, we report a shotgun proteomic analysis of P. profundum grown at atmospheric compared to high pressure using label-free quantitation and mass spectrometry analysis. We have identified differentially expressed proteins involved in high pressure adaptation, which have been previously reported using other methods. Proteins involved in key metabolic pathways were also identified as being differentially expressed. Proteins involved in the glycolysis/gluconeogenesis pathway were up-regulated at high pressure. Conversely, several proteins involved in the oxidative phosphorylation pathway were up-regulated at atmospheric pressure. Some of the proteins that were differentially identified are regulated directly in response to the physical impact of pressure. The expression of some proteins involved in nutrient transport or assimilation, are likely to be directly regulated by pressure. In a natural environment, different hydrostatic pressures represent distinct ecosystems with their own particular nutrient limitations and abundances. However, the only variable considered in this study was atmospheric pressure

    Effect of lutein and antioxidant dietary supplementation on contrast sensitivity in age-related macular disease:A randomized controlled trial

    Objective: The aim of the study is to determine the effect of lutein combined with vitamin and mineral supplementation on contrast sensitivity in people with age-related macular disease (ARMD). Design: A prospective, 9-month, double-masked randomized controlled trial. Setting: Aston University, Birmingham, UK and a UK optometric clinical practice. Subjects: Age-related maculopathy (ARM) and atrophic age-related macular degeneration (AMD) participants were randomized (using a random number generator) to either placebo (n = 10) or active (n=15) groups. Three of the placebo group and two of the active group dropped out. Interventions: The active group supplemented daily with 6 mg lutein combined with vitamins and minerals. The outcome measure was contrast sensitivity (CS) measured using the Pelli-Robson chart, for which the study had 80% power at the 5% significance level to detect a change of 0.3log units. Results: The CS score increased by 0.07 ± 0.07 and decreased by 0.02 ± 0.18 log units for the placebo and active groups, respectively. The difference between these values is not statistically significant (z = 0.903, P = 0.376). Conclusion: The results suggest that 6 mg of lutein supplementation in combination with other antioxidants is not beneficial for this group. Further work is required to establish optimum dosage levels

    Conservative Fragments in Bacterial 16S rRNA Genes and Primer Design for 16S Ribosomal DNA Amplicons in Metagenomic Studies

    Bacterial 16S ribosomal DNA (rDNA) amplicons have been widely used in the classification of uncultured bacteria inhabiting environmental niches. Primers targeting conservative regions of the rDNAs are used to generate amplicons of variant regions that are informative in taxonomic assignment. One problem is that the percentage coverage and application scope of the primers used in previous studies are largely unknown. In this study, conservative fragments of available rDNA sequences were first mined and then used to search for candidate primers within the fragments by measuring the coverage rate defined as the percentage of bacterial sequences containing the target. Thirty predicted primers with a high coverage rate (>90%) were identified, which were basically located in the same conservative regions as known primers in previous reports, whereas 30% of the known primers were associated with a coverage rate of <90%. The application scope of the primers was also examined by calculating the percentages of failed detections in bacterial phyla. Primers A519–539, E969–983, E1063–1081, U515 and E517, are highly recommended because of their high coverage in almost all phyla. As expected, the three predominant phyla, Firmicutes, Gemmatimonadetes and Proteobacteria, are best covered by the predicted primers. The primers recommended in this report shall facilitate a comprehensive and reliable survey of bacterial diversity in metagenomic studies
