    A novel role for thrombopoietin in regulating osteoclast development in humans and mice

    Emerging data suggest that megakaryocytes (MKs) play a significant role in skeletal homeostasis. Indeed, osteosclerosis observed in several MK-related disorders may be a result of increased numbers of MKs. In support of this idea, we have previously demonstrated that MKs increase osteoblast (OB) proliferation by a direct cell-cell contact mechanism and that MKs also inhibit osteoclast (OC) formation. As MKs and OCs are derived from the same hematopoietic precursor, in these osteoclastogenesis studies we examined the role of the main MK growth factor, thrombopoietin (TPO) on OC formation and bone resorption. Here we show that TPO directly increases OC formation and differentiation in vitro. Specifically, we demonstrate the TPO receptor (c-mpl or CD110) is expressed on cells of the OC lineage, c-mpl is required for TPO to enhance OC formation in vitro, and TPO activates the mitogen-activated protein kinases, Janus kinase/signal transducer and activator of transcription, and nuclear factor-kappaB signaling pathways, but does not activate the PI3K/AKT pathway. Further, we found TPO enhances OC resorption in CD14+CD110+ human OC progenitors derived from peripheral blood mononuclear cells, and further separating OC progenitors based on CD110 expression enriches for mature OC development. The regulation of OCs by TPO highlights a novel therapeutic target for bone loss diseases and may be important to consider in the numerous hematologic disorders associated with alterations in TPO/c-mpl signaling as well as in patients suffering from bone disorders

    Effect of a web-based chronic disease management system on asthma control and health-related quality of life: study protocol for a randomized controlled trial

    <p>Abstract</p> <p>Background</p> <p>Asthma is a prevalent and costly disease resulting in reduced quality of life for a large proportion of individuals. Effective patient self-management is critical for improving health outcomes. However, key aspects of self-management such as self-monitoring of behaviours and symptoms, coupled with regular feedback from the health care team, are rarely addressed or integrated into ongoing care. Health information technology (HIT) provides unique opportunities to facilitate this by providing a means for two way communication and exchange of information between the patient and care team, and access to their health information, presented in personalized ways that can alert them when there is a need for action. The objective of this study is to evaluate the acceptability and efficacy of using a web-based self-management system, My Asthma Portal (MAP), linked to a case-management system on asthma control, and asthma health-related quality of life.</p> <p>Methods</p> <p>The trial is a parallel multi-centered 2-arm pilot randomized controlled trial. Participants are randomly assigned to one of two conditions: a) MAP and usual care; or b) usual care alone. Individuals will be included if they are between 18 and 70, have a confirmed asthma diagnosis, and their asthma is classified as not well controlled by their physician. Asthma control will be evaluated by calculating the amount of fast acting beta agonists recorded as dispensed in the provincial drug database, and asthma quality of life using the Mini Asthma Related Quality of Life Questionnaire. Power calculations indicated a needed total sample size of 80 subjects. Data are collected at baseline, 3, 6, and 9 months post randomization. Recruitment started in March 2010 and the inclusion of patients in the trial in June 2010.</p> <p>Discussion</p> <p>Self-management support from the care team is critical for improving chronic disease outcomes. Given the high volume of patients and time constraints during clinical visits, primary care physicians have limited time to teach and reinforce use of proven self-management strategies. HIT has the potential to provide clinicians and a large number of patients with tools to support health behaviour change.</p> <p>Trial Registration</p> <p>Current Controlled Trials <a href="http://www.controlled-trials.com/ISRCTN34326236">ISRCTN34326236</a>.</p

    Abstract Nickel Metal-Hydride (NiMH) is an advanced high-power battery technology that is presently employed in Hybrid Electric Vehicles (HEVs) and is one of several technologies undergoing continuing research and development by FreedomCAR. Unlike some other HEV battery technologies, NiMH exhibits a strong hysteresis effect upon charge and discharge. This hysteresis has a profound impact on the ability to monitor state-of-charge and battery performance. Researchers at the Idaho National Engineering and Environmental Laboratory (INEEL) have been investigating the implications of NiMH hysteresis on HEV battery testing and performance.     La sistematización de experiencias del ejercicio profesional supervisado -EPS-, realizada por la estudiante Arelis Elizabeth Rodríguez Rodríguez, carné 200941543 de la carrera de Trabajo social del Centro Universitario de Sur Occidente –CUNSUROC–, de la Universidad de San Carlos de Guatemala –USAC– con sede en Mazatenango, Suchitepéquez. Como requisito previo a realizar el Examen General Público para optar al título de Trabajadora Social en grado de Licenciada. El informe de sistematización de experiencias se encuentra organizado en siete apartados. En el primero se abordan los temas de desarrollo, concepción del desarrollo, visiones, teorías o paradigmas, modelos, gestión, concepto, autogestión, cogestión y gestión del desarrollo. En el segundo apartado se da a conocer el análisis del contexto en aspectos como: historia, demografía, proyectos en ejecución, recursos naturales, medios de transporte y comunicación, infraestructura, costumbres, religión, condición de los servicios básicos, comercios y organización local. El tercer apartado detalla el análisis de lo social, proceso de correlación de fuerzas en un informe analítico, se describen actores sociales externos e internos y la relación de convivencia. En el cuarto apartado se describe el análisis de la cuestión social en tres dimensiones, lo social, se especifica la violencia manifestada en el barrio, así como el nivel de conocimiento acerca de derechos humanos; en la dimensión económica se da a conocer la manifestación de la pobreza y política social; en la dimensión política el nivel de corrupción, transparencia y auditoría social
