    Characterization of cellulase production by carbon sources in two Bacillus species

    The induction of cellulase production in two Bacillus spp. was studied by means of measuring cellulase activities under the condition of different carbon sources. The results indicate that cellulase could not be induced by cellulose material as a sole carbon source. Instead, they could be induced by monosaccharide or disaccharide with reducing group. Moreover, the expression of cellulase components was synergistic. When cell wall/envelope enzyme and endoenzyme from two Bacillus spp. acted on these inducers, analysis of reaction products by high performance liquid chromatography (HPLC) revealed that cell wall/envelope enzyme and endoenzyme from two Bacillus spp. were inactive on these inducers. It also indicated that these inducers entered cells directly and served function of induction.Keywords: Bacillus, cellulase, induction, carbon source

    Lnc RNA HOTAIR functions as a competing endogenous RNA to regulate HER2 expression by sponging miR-331-3p in gastric cancer

    BACKGROUND: Accumulating evidence indicates that the long non-coding RNA HOTAIR plays a critical role in cancer progression and metastasis. However, the overall biological role and clinical significance of HOTAIR in gastric carcinogenesis remains largely unknown. METHODS: HOTAIR expression was measured in 78 paired cancerous and noncancerous tissue samples by real-time PCR. The effects of HOTAIR on gastric cancer cells were studied by overexpression and RNA interference approaches in vitro and in vivo. Insights of the mechanism of competitive endogenous RNAs (ceRNAs) were gained from bioinformatic analysis, luciferase assays and RNA binding protein immunoprecipitation (RIP). The positive HOTAIR/HER2 interaction was identified and verified by immunohistochemistry assay and bivariate correlation analysis. RESULTS: HOTAIR upregulation was associated with larger tumor size, advanced pathological stage and extensive metastasis, and also correlated with shorter overall survival of gastric cancer patients. Furthermore, HOTAIR overexpression promoted the proliferation, migration and invasion of gastric carcinoma cells, while HOTAIR depletion inhibited both cell invasion and cell viability, and induced growth arrest in vitro and in vivo. In particular, HOTAIR may act as a ceRNA, effectively becoming a sink for miR-331-3p, thereby modulating the derepression of HER2 and imposing an additional level of post-transcriptional regulation. Finally, the positive HOTAIR/HER2 correlation was significantly associated with advanced gastric cancers. CONCLUSIONS: HOTAIR overexpression represents a biomarker of poor prognosis in gastric cancer, and may confer malignant phenotype to tumor cells. The ceRNA regulatory network involving HOTAIR and the positive interaction between HOTAIR and HER2 may contribute to a better understanding of gastric cancer pathogenesis and facilitate the development of lncRNA-directed diagnostics and therapeutics against this disease

    Two novel species and three new records of Torulaceae from Yunnan Province, China

    While investigating the diversity of lignicolous fungi in Yunnan Province, China, six fresh collections of Torulaceae were collected and identified based on morphological examination and phylogenetic analyses of combined LSU, ITS, SSU, tef1-α, and rpb2 sequence data. Two new species, viz. Neopodoconis yunnanensis and Torula suae, and three new records, viz. T. canangae (new freshwater habitat record), T. masonii (new host record), and T. sundara (new freshwater habitat record) are reported. Detailed descriptions, illustrations, and a phylogenetic tree to show the placement of these species are provided

    Spatial Pattern Analysis of Heavy Metals in Beijing Agricultural Soils Based on Spatial Autocorrelation Statistics

    This study explored the spatial pattern of heavy metals in Beijing agricultural soils using Moran’s I statistic of spatial autocorrelation. The global Moran’s I result showed that the spatial dependence of Cr, Ni, Zn, and Hg changed with different spatial weight matrixes, and they had significant and positive global spatial correlations based on distance weight. The spatial dependence of the four metals was scale-dependent on distance, but these scale effects existed within a threshold distance of 13 km, 32 km, 50 km, and 29 km, respectively for Cr, Ni, Zn, and Hg. The maximal spatial positive correlation range was 57 km, 70 km, 57 km, and 55 km for Cr, Ni, Zn, and Hg, respectively and these were not affected by sampling density. Local spatial autocorrelation analysis detected the locations of spatial clusters and spatial outliers and revealed that the pollution of these four metals occurred in significant High-high spatial clusters, Low-high, or even High-low spatial outliers. Thus, three major areas were identified and should be receiving more attention: the first was the northeast region of Beijing, where Cr, Zn, Ni, and Hg had significant increases. The second was the southeast region of Beijing where wastewater irrigation had strongly changed the content of metals, particularly of Cr and Zn, in soils. The third area was the urban fringe around city, where Hg showed a significant increase

    Comparison of heavy-ion transport simulations: Collision integral with pions and Δ resonances in a box

    We compare ten transport codes for a system confined in a box, aiming at improved handling of the production of Δ\Delta resonances and pions, which is indispensable for constraining high-density symmetry energy from observables such as the π/π+\pi^-/\pi^+ yield ratio in heavy-ion collisions. The system in a box is initialized with nucleons at saturation density and at 60 MeV temperature. The reactions NNNΔNN\leftrightarrow N\Delta and ΔNπ\Delta\leftrightarrow N\pi are implemented, but the Pauli blocking and the mean-field potential are deactivated in the present comparison. Results are compared to those from the two reference cases of a chemically equilibrated ideal gas mixture and of the rate equation. In the results of the numbers of Δ\Delta and π\pi, deviations from the reference values are observed in many codes, and they depend significantly on the size of the time step. These deviations are tied to different ways in ordering the sequence of collisions and decays, that take place in the same time step. Better agreements are seen in the reaction rates and the number ratios among the isospin species of Δ\Delta and π\pi. These are, however, affected by the correlations, which are absent in the Boltzmann equation, but are induced by the way particle scatterings are treated in transport calculations. The uncertainty in the transport-code predictions of the π/π+\pi^-/\pi^+ ratio for the system initialized at n/p = 1.5, after letting the existing Δ\Delta resonances decay, is found to be within a few percent, which is sufficiently small so that it does not strongly impact constraining the high-density symmetry energy from heavy-ion collisions. Most of the sources of uncertainties have been understood, and individual codes may be further improved.     Comparison of heavy-ion transport simulations: Collision integral in a box

    Simulations by transport codes are indispensable to extract valuable physical information from heavy-ion collisions. In order to understand the origins of discrepancies among different widely used transport codes, we compare 15 such codes under controlled conditions of a system confined to a box with periodic boundary, initialized with Fermi-Dirac distributions at saturation density and temperatures of either 0 or 5 MeV. In such calculations, one is able to check separately the different ingredients of a transport code. In this second publication of the code evaluation project, we only consider the two-body collision term; i.e., we perform cascade calculations. When the Pauli blocking is artificially suppressed, the collision rates are found to be consistent for most codes (to within 1 % or better) with analytical results, or completely controlled results of a basic cascade code. In orderto reach that goal, it was necessary to eliminate correlations within the same pair of colliding particles that can be present depending on the adopted collision prescription. In calculations with active Pauli blocking, the blocking probability was found to deviate from the expected reference values. The reason is found in substantial phase-space fluctuations and smearing tied to numerical algorithms and model assumptions in the representation of phase space. This results in the reduction of the blocking probability in most transport codes, so that the simulated system gradually evolves away from the Fermi-Dirac toward a Boltzmann distribution. Since the numerical fluctuations are weaker in the Boltzmann-Uehling-Uhlenbeck codes, the Fermi-Dirac statistics is maintained there for a longer time than in the quantum molecular dynamics codes. As a result of this investigation, we are able to make judgements about the most effective strategies in transport simulations for determining the collision probabilities and the Pauli blocking. Investigation in a similar vein of other ingredients in transport calculations, like the mean-field propagation or the production of nucleon resonances and mesons, will be discussed in the future publications

    Tools to Support Policy Decisions Related to Treatment Strategies and Surveillance of Schistosomiasis Japonica towards Elimination

    Immunodiagnostic assays are widely applied in the field to control schistosomiasis in P.R. China as the prevalence and infection intensity of schistosome infections decrease. Field evaluations are urgently needed before they can be adopted to support policy decisions of the national programme for the control and elimination of schistosomiasis in P.R. China. We carried out a large scale cross-sectional survey in field settings with different transmission situations to validate immunodiagnostic tools that can be used to formulate new schistosomiasis elimination strategy in P.R. China. Regarding stool examination as gold reference, the validity and screening efficacy of each immunodiagnostic kit were calculated and compared with each other. The association of the prevalence of schistosomiasis and antibody positive rates determined by immunoassays were analyzed using Pearson's correlation coefficient values. The study indicates that which test to use with the elimination strategy is dependent on the purpose of testing, the endemic status of community and the resources available. And more sensitive methods need to be explored and used to target infected individuals for treatment or to eliminate schistosomiasis

    The Genome Sequence of the Leaf-Cutter Ant Atta cephalotes Reveals Insights into Its Obligate Symbiotic Lifestyle

    Leaf-cutter ants are one of the most important herbivorous insects in the Neotropics, harvesting vast quantities of fresh leaf material. The ants use leaves to cultivate a fungus that serves as the colony's primary food source. This obligate ant-fungus mutualism is one of the few occurrences of farming by non-humans and likely facilitated the formation of their massive colonies. Mature leaf-cutter ant colonies contain millions of workers ranging in size from small garden tenders to large soldiers, resulting in one of the most complex polymorphic caste systems within ants. To begin uncovering the genomic underpinnings of this system, we sequenced the genome of Atta cephalotes using 454 pyrosequencing. One prediction from this ant's lifestyle is that it has undergone genetic modifications that reflect its obligate dependence on the fungus for nutrients. Analysis of this genome sequence is consistent with this hypothesis, as we find evidence for reductions in genes related to nutrient acquisition. These include extensive reductions in serine proteases (which are likely unnecessary because proteolysis is not a primary mechanism used to process nutrients obtained from the fungus), a loss of genes involved in arginine biosynthesis (suggesting that this amino acid is obtained from the fungus), and the absence of a hexamerin (which sequesters amino acids during larval development in other insects). Following recent reports of genome sequences from other insects that engage in symbioses with beneficial microbes, the A. cephalotes genome provides new insights into the symbiotic lifestyle of this ant and advances our understanding of host–microbe symbioses
