    Flow injection analysis coupled with carbon electrodes as the tool for analysis of naphthoquinones with respect to their content and functions in biological samples

    Naphthoquinones are one of the groups of secondary metabolites widespread in nature, where they mostly appear as chromatic pigments. They embody broad-range of biological actions from phytotoxic to fungicidal. An anticancer effect of naphthoquinones stimulates an interest in determination and characterization of single derivatives of 1,2- and 1,4-quinones in biological samples. The main aim of this work was to suggest a technique suitable to determine lawsone, juglone and/or plumbagin in biological samples and to study of their influence on BY-2 tobacco cells. The BY-2 tobacco cells were cultivated in the presence of the naphthoquinones of interest ( 500 mu g.l(-1)) for 24 h and then the morphological changes were observed. We found out that naphthoquinones triggered the programmed cell death at BY-2 cells, which can be confirmed by the apoptotic bodies in nucleus. After that we suggested and optimized different electrochemical techniques such differential pulse voltammetry ( DPV) coupled with hanging mercury drop ( HMDE) and carbon paste electrode, micro flow device coupled with carbon screen printed electrodes and flow injection analysis coupled with Coulochem III detector to determine them. The detection limits of naphthoquinones of interest were expressed as 3S/N and varied from units to hundreds of ng per millilitres according to methods used. Moreover, we utilized DPV coupled with HMDE and micro flow device to determine content of juglone in leaves Persian walnut ( Juglans regia). We determined that the leaves contained juglone tenths of g per 100 g of fresh weight. The results obtained show the convincing possibilities of using of these methods in analysis of plant secondary metabolites

    E-Shops have become considerable source of income for enterprisers and companies -- blowing the internet bubble before several years have shown, however, that not every E-Shop is flourishing and competitive. In a rough competition has enforced just such shop that fit information, technologies and business together; effectively capitalized resources and accommodated customer's needs. The goal of this work is to try to describe important steps, activities, procedures and decisions dutiny foundation and operating of the profitable E-Shop. The work is classified into the theoretical and the practical part -- in separated chapters it describes economic analysis, lawful obligations and the general presentation of the E-Shop on a website as well as the specific presentation on example of the E-Shop MojePekárna.cz.Internetové obchody se staly nemalým zdrojem příjmů pro podnikatele a firmy ? prasknutí internetové bubliny před několika lety však ukázalo, že ne každý internetový obchod je prosperující a konkurenceschopný. V tvrdé konkurenci se prosadil jen takový obchod, který skloubil informační technologie a business, efektivně využíval zdroje a uspokojoval potřeby zákazníků. Cílem této práce je pokusit se popsat důležité kroky, činnosti, postupy a rozhodnutí při zakládání a provozování ziskového internetového obchodu. Práce je rozdělena na teoretickou a praktickou část ? v jednotlivých kapitolách popisuje ekonomickou analýzu, zákonné povinnosti a prezentaci obchodu na webu obecně i na konkrétním příkladu internetového obchodu MojePekárna.cz

    Chemokine and Cytokine Profiles in Patients with Hand Osteoarthritis

    Background: The development of hand osteoarthritis (HOA) and its progression into the erosive subset are unclear, but inflammation is suspected to be the main source. To verify the involvement of inflammation in HOA pathogenesis, we evaluate serum inflammatory mediators and their association with HOA-related clinical features in patients. Methods: 153 participants (50 non-erosive HOA patients, 54 erosive HOA patients, and 49 healthy control subjects) were included in this study. All patients underwent clinical examination, which included assessment of tender and swollen small hand joints, ultrasound (US) examination, and self-reported measures (e.g., AUSCAN or algofunctional indexes). Serum inflammatory mediators were quantified using human cytokine 27-plex immunoassay. We employed linear modelling, correlation analysis, and resampling statistics to evaluate the association of these mediators to HOA. Results: We identified increased levels of nine inflammatory mediators (e.g., eotaxin, monocyte chemoattractant protein 1, interleukin-8, and tumour necrosis factor) in HOA patients compared to healthy controls. Increased mediators correlated with ultrasound findings as well as with clinically tender and swollen joint counts in patients with erosive HOA. However, none of the mediators distinguished between erosive and non-erosive HOA subtypes. Conclusion: Our findings support the hypothesis on the involvement of inflammation in HOA

    Extrakce toponym z historických map pro efektivní vyhledávání informací

    Článek se zabývá detekcí, klasifikací a rozpoznáváním toponym v ručně kreslených historických katastrálních mapách. Toponyma jsou místní jména měst, vesnic a dalších míst, jako jsou řeky, lesy atd. Extrahovaná toponyma se používají jako klíčová slova v systému vyhledávání informací, které umožňuje inteligentní a efektivní vyhledávání v historických mapových sbírkách. Vytvořili jsme novou datovou sadu, která je volně dostupná pro výzkumné účely. Poté navrhujeme nový přístup ke klasifikaci toponym založený na deskriptoru KAZE. Dále porovnáváme a vyhodnocujeme několik nejmodernějších metod pro detekci na naší úloze detekce toponym. Na závěr prezentujeme výsledky rozpoznávání textu toponym pomocí populárního enginu Tesseract.The paper deals with detection, classification and recognition of toponyms in hand-drawn historical cadastral maps. Toponyms are local names of towns, villages and landscape features such as rivers, forests etc. The detected and recognized toponyms are utilized as keywords in an information retrieval system that allows intelligent and efficient searching in historical map collections. We create a novel annotated dataset that is freely available for research and educational purposes. Then, we propose a novel approach for toponym classification based on KAZE descriptor. Next we compare and evaluate several state-of-the-art methods for text and object detection on our toponym detection task. We further show the results of toponym text recognition using popular Tesseract engine
