A XIII-as véralvadási faktor A és B alegységének kapcsolódásáért felelős struktúrális elemek azonosítása a B alegységben = Identification of the binding domain(s) involved in the binding with FXIII-A on FXIII-B

1. rFXIII-B. Máj cDNS könyvtárból klónoztam a teljes hosszúságú FXIII-B cDNS-t és előállítottam a 2, 4, 6, 8 Sushi domént tartalmazó mutánsokat. Az optimalizált expressziós rendszer a COS-7-DEAE-dextrán kombináció volt. A médiumból a rFXIII-t és mutánsait Ni-kromatográfiával részlegesen tisztítottam nem denaturáló körülmények között. A fehérjékben a diszulfid hidak kialakultak és a rekombinánsok molekulasúlya jól korrelált a várttal. A stabilitás vizsgálat során degradáció nem volt kimutatható. 2. rFXIII-A. A FXIII-A cDNS-t klónoztam és pET vektorba inzertáltam. A benne levő Shine-Dalgarno szekvenciát megszüntettem. Az expressziós kísérletek nem sikerültek. Ezután maltóz-kötő fehérjével fúzióban próbáltam meg a rFXIII-A-t expresszálni, de sem peri- sem citoplazmatikus esetben nem sikerült a rFXIII-A előállítása. Ezután a rFXIII-A esetében is pcDNA emslő rendszert használtam az expresszióra. Ni-oszlopon a sejtlizátumból történő tisztítás után a rFXIII-A kimutatható volt. 3. Az alegységek kötődésének vizsgálata. A mikrolemezre rögzített rFXIII-A-hoz hozzáadva a rFXIII-B-t és deléciós mutánsait, minden esetben kaptam a kötődésre utaló jelet, mely a lekötött rFXIII-A mennyiségének függvénye volt. Fordított esetben is hasonló eredményt kaptam, azaz a lerögzített rFXIII-B és deléciós mutánsai a rFXIII-A-t kötik. Ez a megfigyelés még további megerősítést igényel, de az eredmények arra utalnak, hogy a FXIII-B N-terminálisa vesz részt a FXIII-A kötésben. | 1. rFXIII-B. The cDNA of FXIII-B was cloned from liver cDNA library and the deletion mutants containing 2, 4, 6 and 8 Sushi domains were generated. The optimized expression system used COS-7 cells and DEAE-dextran for transfection. For partial purification of the recombinants Ni-chromatography was used under mild conditions. Disulphide bridges were formed and the molecular weight of the recombinants correlated well with the calculated ones. No degradation was observed after prolonged storage period at 4°C. 2. rFXIII-A. The cDNA of FXIII-A was cloned and inserted into pET vector. Although the Shine-Dalgarno sequence was abolished, no rFXIII-A could be detected. After subloning into maltose-binding fusion system, rFXIII-A production was unsuccessful both in the periplasm and in the cytoplasm. Final attempt was to clone cDNA into pcDNA vector and express it in mammalian cell line. rFXIII-A could be detected from the cell lysate after partial purification on Ni-column. 3. Testing the binding of the subunits. When rFXIII-A was coated into the microplate and the rFXIII-B and its mutants were added, rFXIII-B-specific signal could be detected, which was proportionate to the amount of coated rFXIII-A. The reversed setting, where rFXIII-B and its mutants were coated yielded similar results. These observations, though preliminary, suggest that the N-terminal region of FXIII-B is responsible for the binding of FXIII-A

A négy éves kutatási program eredetileg jelzett célkitűzését messze meghaladó elméleti eredményeket a 2005 novemberében elkészült és a L'Harmattan könyvkiadónak még ugyanabban a hónapban átadott - a tervezett tanulmánykötet műfajával ellentétben - szisztematikus monográfia foglalja össze. Az "Igazságosság és politika" címmel 2006 márciusában megjelenő, körülbelül 400 oldalas monográfia egymáshoz kapcsolódó főbb új eredményei az alábbiak: 1. A modern társadalomelmélet új szakaszának, a politikai társadalomelmélet kialakulásának, jellemzőinek, történeti feltételeinek bemutatása. 2. A kibontakozó politikai társadalomelmélet meghatározó vonásának, az erkölcsi-normatív probléma központi fontosságának a kiemelése. 3. Az igazságosságnak, mint a politikai legitimáció erkölcsi-normatív elveinek a meghatározása. 4. Az igazságosság elméleti tárgyalásának új - az eddigi igazoláselméleti megközelítéssel szemben - megalapozás-elméleti stratégiája részletes kidolgozása. 5. Az igazságosság elveinek eredetien új elméleti meghatározása, mely önbecsülési igazságosságként a kurrens elméletekkel szemben alternatív igénnyel lép fel. | The final outgrowths of the implementation of the research program goes far beyond its original aims. At the end of the year 2005 the theoretical achievements were summarized in a comprehensive monograph of 400 pages which was handed over to the L'Harmattan Publisher. The probable date of publication will be at the end of March, 2006. The main accomplishments of the monograph can be summarized as follows: 1. The description of the process, peculiarities and historical conditions which lead to the formation of a new period of the general social theory, to the develpoment of the political social theory. 2. Making clear, that the determinant elements of the political social theory are its moral and normative components 3. The definition of justice, as a set of moral and normative principles for political legitimation. 4. A new approach to the study of justice, which focuses to the theoretical and metodological problems of the theoretical foundation. 5. The definition of the genuinely new principles of justice, which can be called as the justice of self-respect. This could be considered, as a new concept in the recent theoretical diskussion

THE FORMATION OF SPHERICAL PARTICLES UNDER ABRASIVE CONDITIONS

The formation of spherical wear debris was investigated by an analytical ferrograph in different machines. It was stated that the abrasive contamination in gear boxes, I.C. engines and hydraulic systems is a contributor to the generation of spherical particles. According to examinations carried out so far the spherical third body formation can be created through joint plastic deformation of delaminated particles or as a result of forming to supercooled liquid due to the collective effects of state variables

Nowadays there are several grid connected converters in the grid system. These grid connected converters are generally the converters of renewable energy sources, industrial four quadrant drives and other converters with DC link. These converters are connected to the grid through a three phase bridge. The standards prescribe the maximal harmonic emission which could be easily limited with high switching frequency. The high switching losses can be reduced to the half with the utilization of the well-known Flat-top modulation. The suggested DC Link Floating (DLF) control method is an extension of the Flat-top modulation with which the switching losses could be further reduced

Elektroreológiai elasztomer korongok külső elektromos tér hatására történő deformációjának vizsgálata: Investigation of deformation of electrorheological elastomer discs by external electric fields

The aim of our research was to examine different concentrations of barium titanate based on electrorheological (ER) elastomeric discs and to investigate the extent of their deformation due to external electric field. A laser sensor measuring system was set up for more exact measurements. As a result, the discs contracted to the outer space, the extent of which depended on the concentration of ER elastomeric filler. Further examination of the samples revealed a relationship between the barium titanate content and the amount of dielectric permittivity. Kivonat Munkánk során különböző koncentrációjú bárium-titanát alapú elektroreológiai (ER) elasztomer korongokat készítettünk és vizsgáltuk ezek külső elektromos tér hatására bekövetkező deformációjának nagyságát. A megfelelő mérésekhez egy lézerszenzoros mérőrendszert állítottunk össze. Megállapítottuk, hogy a korongok a külső tér irányában kontrakciót szenvednek, amelynek mértéke függ az ER elasztomer töltőanyag koncentrációjától. A minták további vizsgálata során összefüggést állítottunk fel a bárium-titanát tartalom és a dielektromos permittivitás nagyságára vonatkozólag

3SC Control for high efficiency grid connected photovoltaic converters

Nowadays there are several grid connected converters in the grid system. These grid connected converters are generally the converters of renewable energy sources, industrial four quadrant drives and other converters with DC link. These converters are connected to the grid typically through a three phase IGBT bridge. The standards prescribe the permissible maximal harmonic emission and the maximal reactive current for the grid connected converters. For a converter working at nominal power these two prescriptions together mean almost unity power factor. The harmonic emission could be easily limited with high switching frequency and/or with large harmonic filters. Further financial viewpoints are the efficiency, the small size and weight. Comparing to traditional control these requirements can be simultaneously satisfied much better with the 3SC (three state control) method. At 3SC we utilize all the three allowed state of one arm of the IGBT bridge, i.e. upper part conducting, lower part conducting, none of the m conducting
