    Influence of thermal hydrolysis pretreatment on physicochemical properties and anaerobic biodegradability of waste activated sludge with different solids content

    © 2018 Elsevier Ltd The influence of thermal hydrolysis pretreatment (THP) on physicochemical properties (pH, total solids, volatile solids, chemical oxygen demand, total nitrogen, ammonium nitrogen, volatile fatty acids, viscosity, and cell morphology) and anaerobic biodegradability of highly concentrated waste activated sludge (WAS) with TS content ranging from 1 to 7% was evaluated at different temperatures ranging from 100 to 220 °C. The biomethane potential (BMP) of the WAS was systematically analyzed and evaluated. Images of its cellular structure were also analyzed. The results indicated that THP is a useful method for solubilizing volatile solids and enhancing CH 4 production regardless of the TS content of the WAS feed. The ultimate CH 4 production determined from the BMP analysis was 313–348 L CH 4 /kg VS (72.6–74.1% CH 4 ) at the optimum THP temperature of 180 °C. The results showed that THP could improve both the capacity and efficiency of anaerobic digestion, even at a high TS content, and could achieve the dual purpose of sludge reduction and higher energy recovery

    A new approach for concurrently improving performance of South Korean food waste valorization and renewable energy recovery via dry anaerobic digestion under mesophilic and thermophilic conditions

    © 2017 Elsevier Ltd Dry semicontinuous anaerobic digestion (AD) of South Korean food waste (FW) under four solid loading rates (SLRs) (2.30–9.21 kg total solids (TS)/m3 day) and at a fixed TS content was compared between two digesters, one each under mesophilic and thermophilic conditions. Biogas production and organic matter reduction in both digesters followed similar trends, increasing with rising SLR. Inhibitor (intermediate products of the anaerobic fermentation process) effects on the digesters’ performance were not observed under the studied conditions. In all cases tested, the digesters’ best performance was achieved at the SLR of 9.21 kg TS/m3 day, with 74.02% and 80.98% reduction of volatile solids (VS), 0.87 and 0.90 m3 biogas/kg VSremoved, and 0.65 (65% CH4) and 0.73 (60.02% CH4) m3 biogas/kg VSfed, under mesophilic and thermophilic conditions, respectively. Thermophilic dry AD is recommended for FW treatment in South Korea because it is more efficient and has higher energy recovery potential when compared to mesophilic dry AD

    Thermophilic anaerobic digestion of model organic wastes: Evaluation of biomethane production and multiple kinetic models analysis

    © 2019 Elsevier Ltd The main aim of this work was to test various organic wastes, i.e. from a livestock farm, a cattle slaughterhouse and agricultural waste streams, for its ability to produce methane under thermophilic anaerobic digestion (AD) conditions. The stability of the digestion, potential biomethane production and biomethane production rate for each waste were assessed. The highest methane yield (110.83 mL CH4/g VSadded day) was found in the AD of crushed animal carcasses on day 4. The experimental results were analyzed using four kinetic models and it was observed that the Cone model described the biomethane yield as well as the methane production rate of each substrate. The results from this study showed the good potential of model organic wastes to produce biomethane

    For genetic counselling of a woman on familial breast cancer, an accurate evaluation of the probability that she carries a germ-line mutation is needed to assist in making decisions about genetic-testing. We used data from eight collaborating centres comprising 618 families (346 breast cancer only, 239 breast or ovarian cancer) recruited as research families or counselled for familial breast cancer, representing a broad range of family structures. Screening was performed in affected women from 618 families for germ-line mutations in BRCA1 and in 176 families for BRCA2 mutations, using different methods including SSCP, CSGE, DGGE, FAMA and PTT analysis followed by direct sequencing. Germ-line BRCA1 mutations were detected in 132 families and BRCA2 mutations in 16 families. The probability of being a carrier of a dominant breast cancer gene was calculated for the screened individual under the established genetic model for breast cancer susceptibility, first, with parameters for age-specific penetrances for breast cancer only [7] and, second, with age-specific penetrances for ovarian cancer in addition [20]. Our results indicate that the estimated probability of carrying a dominant breast cancer gene gives a direct measure of the likelihood of detecting mutations in BRCA1 and BRCA2. For breast/ovarian cancer families, the genetic model according to Narod et al. [20] is preferable for calculating the proband's genetic risk, and gives detection rates that indicate a 50% sensitivity of the gene test. Due to the incomplete BRCA2 screening of the families, we cannot yet draw any conclusions with respect to the breast cancer only families

    Eosinophil and T Cell Markers Predict Functional Decline in COPD Patients

    BACKGROUND. The major marker utilized to monitor COPD patients is forced expiratory volume in one second (FEV1). However, asingle measurement of FEV1 cannot reliably predict subsequent decline. Recent studies indicate that T lymphocytes and eosinophils are important determinants of disease stability in COPD. We therefore measured cytokine levels in the lung lavage fluid and plasma of COPD patients in order to determine if the levels of T cell or eosinophil related cytokines were predictive of the future course of the disease. METHODS. Baseline lung lavage and plasma samples were collected from COPD subjects with moderately severe airway obstruction and emphysematous changes on chest CT. The study participants were former smokers who had not had a disease exacerbation within the past six months or used steroids within the past two months. Those subjects who demonstrated stable disease over the following six months (ΔFEV1 % predicted = 4.7 ± 7.2; N = 34) were retrospectively compared with study participants who experienced a rapid decline in lung function (ΔFEV1 % predicted = -16.0 ± 6.0; N = 16) during the same time period and with normal controls (N = 11). Plasma and lung lavage cytokines were measured from clinical samples using the Luminex multiplex kit which enabled the simultaneous measurement of several T cell and eosinophil related cytokines. RESULTS AND DISCUSSION. Stable COPD participants had significantly higher plasma IL-2 levels compared to participants with rapidly progressive COPD (p = 0.04). In contrast, plasma eotaxin-1 levels were significantly lower in stable COPD subjects compared to normal controls (p < 0.03). In addition, lung lavage eotaxin-1 levels were significantly higher in rapidly progressive COPD participants compared to both normal controls (p < 0.02) and stable COPD participants (p < 0.05). CONCLUSION. These findings indicate that IL-2 and eotaxin-1 levels may be important markers of disease stability in advanced emphysema patients.     The ansamycin antibiotic, rifamycin SV, inhibits BCL6 transcriptional repression and forms a complex with the BCL6-BTB/POZ domain

    BCL6 is a transcriptional repressor that is over-expressed due to chromosomal translocations, or other abnormalities, in ~40% of diffuse large B-cell lymphoma. BCL6 interacts with co-repressor, SMRT, and this is essential for its role in lymphomas. Peptide or small molecule inhibitors, which prevent the association of SMRT with BCL6, inhibit transcriptional repression and cause apoptosis of lymphoma cells in vitro and in vivo. In order to discover compounds, which have the potential to be developed into BCL6 inhibitors, we screened a natural product library. The ansamycin antibiotic, rifamycin SV, inhibited BCL6 transcriptional repression and NMR spectroscopy confirmed a direct interaction between rifamycin SV and BCL6. To further determine the characteristics of compounds binding to BCL6-POZ we analyzed four other members of this family and showed that rifabutin, bound most strongly. An X-ray crystal structure of the rifabutin-BCL6 complex revealed that rifabutin occupies a partly non-polar pocket making interactions with tyrosine58, asparagine21 and arginine24 of the BCL6-POZ domain. Importantly these residues are also important for the interaction of BLC6 with SMRT. This work demonstrates a unique approach to developing a structure activity relationship for a compound that will form the basis of a therapeutically useful BCL6 inhibitor

    Intra-operative real time intracranial subarachnoid haemorrhage during glial tumour resection: A case report

    Glial tumours associated with subarachnoid haemorrhage are very rare. A 64-year-old woman admitted with a history of 3 weeks seizures and a left sided hemiparesis and dysphasia. The magnetic resonance disclosed heterogeneously enhancing a right temporal mass. During surgery, suddenly an abrupt and extensive swelling had occurred both in tumour and the brain tissue. The surgery was completed with a gross total tumour resection together with a partial temporal lobectomy. Postoperative computerized tomography demonstrated a massive subarachnoid hemorrhage (SAH). A cerebral Magnetic Resonance (MR) angiography showed neither an aneurysm nor arteriovenous malformation. Coincidence of an intracerebral tumour and subarachnoid haemorrhage would be devastating
