    Experiments and simulations demonstrating the rapid ultrasonic rewarming of frozen tissue cryovials

    The development of methods to safely rewarm large cryopreserved biological samples remains a barrier to the widespread adoption of cryopreservation. Here, experiments and simulations were performed to demonstrate that ultrasound can increase rewarming rates relative to thermal conduction alone. An ultrasonic rewarming setup based on a custom 444 kHz tubular piezoelectric transducer was designed, characterized, and tested with 2 ml cryovials filled with frozen ground beef. Rewarming rates were characterized in the -20 °C to 5 °C range. Thermal conduction-based rewarming was compared to thermal conduction plus ultrasonic rewarming, demonstrating a tenfold increase in rewarming rate when ultrasound was applied. The maximum recorded rewarming rate with ultrasound was 57° C/min, approximately 2.5 times faster than with thermal conduction alone. Coupled acoustic and thermal simulations were developed and showed good agreement with the heating rates demonstrated experimentally and were also used to demonstrate spatial heating distributions with small (<3° C) temperature differentials throughout the sample when the sample was below 0° C. The experiments and simulations demonstrate the potential for ultrasonic cryovial rewarming with a possible application to large volume rewarming, as faster rewarming rates may improve the viability of cryopreserved tissues and reduce the time needed for cells to regain normal function

    Single Pulse Illumination of Multi-Layer Photoacoustic Holograms for Patterned Ultrasound Field Generation

    A new method for the creation of patterned, focused, optically generated acoustic fields using a single optical pulse is introduced. This utilises multi-layer `holograms' composed of several spatially separate absorbing layers. Each layer is individually patterned so as to focus at a set of targeted points. To create the patterns, a ray-tracing model was implemented to calculate the impulse response of pixels within each absorbing layer to a set of targeted points. An optimisation approach was then used to find the optimal pattern for each layer to create a field evenly focused at each of the target points. The method was validated using both numerical simulations and acoustic field measurements. It was demonstrated that a 3×3 array of acoustic foci could be generated from a 3-layer hologram using a single laser pulse

    Rapid Spatial Mapping of Focused Ultrasound Fields Using a Planar Fabry-Pérot Sensor

    Measurement of high acoustic pressures is necessary in order to fully characterise clinical high-intensity focused ultrasound (HIFU) fields, and for accurate validation of computational models of ultrasound propagation. However, many existing measurement devices are unable to withstand the extreme pressures generated in these fields, and those that can often exhibit low sensitivity. Here, a planar Fabry-Pérot interferometer with hard dielectric mirrors and spacer was designed, fabricated, and characterised and its suitability for measurement of nonlinear focused ultrasound fields was investigated. The noise equivalent pressure of the scanning system scaled with the adjustable pressure detection range between 49 kPa for pressures up to 8 MPa and 152 kPa for measurements up to 25 MPa, over a 125 MHz measurement bandwidth. Measurements of the frequency response of the sensor showed that it varied by less than 3 dB in the range 1 - 62 MHz. The effective element size of the sensor was 65 μm and waveforms were acquired at a rate of 200 Hz. The device was used to measure the acoustic pressure in the field of a 1.1 MHz single element spherically focused bowl transducer. Measurements of the acoustic field at low pressures compared well with measurements made using a PVDF needle hydrophone. At high pressures, the measured peak focal pressures agreed well with the focal pressure modelled using the Khokhlov-Zabolotskaya-Kuznetsov equation. Maximum peak positive pressures of 25 MPa, and peak negative pressures of 12 MPa were measured, and planar field scans were acquired in scan times on the order of 1 minute. The properties of the sensor and scanning system are well suited to measurement of nonlinear focused ultrasound fields, in both the focal region and the low pressure peripheral regions. The fast acquisition speed of the system and its low noise equivalent pressure are advantageous, and with further development of the sensor, it has potential in application to HIFU metrology

    Electrical detection of 31P spin quantum states

    In recent years, a variety of solid-state qubits has been realized, including quantum dots, superconducting tunnel junctions and point defects. Due to its potential compatibility with existing microelectronics, the proposal by Kane based on phosphorus donors in Si has also been pursued intensively. A key issue of this concept is the readout of the P quantum state. While electrical measurements of magnetic resonance have been performed on single spins, the statistical nature of these experiments based on random telegraph noise measurements has impeded the readout of single spin states. In this letter, we demonstrate the measurement of the spin state of P donor electrons in silicon and the observation of Rabi flops by purely electric means, accomplished by coherent manipulation of spin-dependent charge carrier recombination between the P donor and paramagnetic localized states at the Si/SiO2 interface via pulsed electrically detected magnetic resonance. The electron spin information is shown to be coupled through the hyperfine interaction with the P nucleus, which demonstrates the feasibility of a recombination-based readout of nuclear spins

    In the present study we have asked the question of whether F1 carrier-primed T cells can serve as helper cells for either or both parental B cells when (a) the carrier molecule employed is under genetic control such that one parental strain is a responder and the other is a nonresponder, and (b) the determinant specificity of the parental B cells being assessed is not under genetic control and bears no relationship to the specificity of the carrier molecule. Utilizing the system of immune response gene control of responses to the terpolymer L-glutamic acid-L-lysine-L-tyrosine (GLT) to which A strain mice (H-2a) are nonresponders, whereas BALB/c (H-2d) and (BALB/c x A)F1 hybrids (CAF1) are responders, these studies demonstrate that GLT-primed T cells of CAF1 donors can provide for responder BALB/c, but not for nonresponder A/J, the required stimulus for the anti-DNP responses of DNP-specific B cells of these respective parental strains to the DNP conjugate of GLT. The implications of these findings for Ir gene function in physiologic T-B cell interactions are discussed in detail

    Drivers and sustainability of bird hunting in Madagascar

    Bird conservation depends on robust data on the densities of and threats to each species, and an understanding of the choices and incentives of bird hunters. This first comprehensive study of bird hunting and its effects in Madagascar uses 8 years of data on 87 bird species to determine bird densities and hunting pressure, incentives, choices, methods, spatial variation, and sustainability on the Masoala Peninsula of Madagascar. We find that bird hunting is common, affecting human wellbeing and, for some species, long-term population viability. Hunters caught more abundant species of lower trophic levels and consumers preferred the flavor of abundant granivores and nectarivores, while they disliked carnivores, scavengers, and species with common cultural proscriptions. Wealth increased species selectivity among consumers, whereas food insecurity increased hunting pressure overall. Projected and documented declines in at least three species are concerning, qualifying at least two for increased IUCN threatened species categories. We provide novel, data-driven assessments of hunting's threat to Madagascar's birds, identify key species of concern, and suggest both species- and consumer-specific conservation actions

    Holocentric Chromosomes of Luzula elegans Are Characterized by a Longitudinal Centromere Groove, Chromosome Bending, and a Terminal Nucleolus Organizer Region

    The structure of holocentric chromosomes was analyzed in mitotic cells of Luzula elegans. Light and scanning electron microscopy observations provided evidence for the existence of a longitudinal groove along each sister chromatid. The centromere-specific histone H3 variant, CENH3, colocalized with this groove and with microtubule attachment sites. The terminal chromosomal regions were CENH3-negative. During metaphase to anaphase transition, L. elegans chromosomes typically curved to a sickle-like shape, a process that is likely to be influenced by the pulling forces of microtubules along the holocentric axis towards the corresponding microtubule organizing regions. A single pair of 45S rDNA sites, situated distal to Arabidopsis-telomere repeats, was observed at the terminal region of one chromosome pair. We suggest that the 45S rDNA position in distal centromere-free regions could be required to ensure chromosome stability. Copyright (C) 2011 S. Karger AG, Base

    Risk of prostate cancer after isolated high-grade prostatic intraepithelial neoplasia (HGPIN) detected on extended core needle biopsy : a UK hospital experience.

    Background High-grade prostatic intraepithelial neoplasia (HGPIN) is a precursor lesion to prostate cancer (CaP). UK-based studies examining the occurrence of isolated HGPIN and subsequent risk of CaP are lacking. Our aim was to assess the occurrence of HGPIN in a regional UK population and to determine whether in a retrievable cohort of such patients that had repeat extended core biopsies, there was an elevated risk of CaP. Methods A retrospective analysis of the pathology database was conducted at our institution (Lancashire Teaching Hospitals NHS Foundation Trust) for prostate biopsies recorded between January 2001 and December 2005 (all extended core biopsies). Those patients with isolated HGPIN on 1st set of biopsies were identified and, their clinical characteristics and pathological findings from subsequent biopsies (if any) were determined. The risk of CaP on subsequent biopsies based on presenting baseline PSA was stratified. Results Of 2,192 biopsied patients, there were 88 cases of isolated HGPIN of which 67 patients underwent one or more repeat biopsies. In this repeat-biopsy group, 28 CaP diagnoses were made. Age at first biopsy (P 20 ng/ml – 87.5%. Conclusion Based on our results, we recommend delaying the 1st repeat biopsy at low PSA range but to have a shorter interval to repeat biopsies at intermediate and higher PSA ranges
