    Reduced expression of multiple gap junction proteins is a feature of cervical dysplasia

    Cervical dysplasia is a premalignant lesion associated with human papillomavirus (HPV) infection which, over time, can turn cancerous. Previous studies have indicated that loss of gap junctions may be a feature of cervical cancer and premalignant dysplasia. Loss of the gap junction protein connexin43 has been demonstrated in dysplastic cervix, but other connexins have not been investigated. In contrast we previously showed that HPV-associated cutaneous warts – and other hyperproliferative skin conditions – display a dramatic upregulation of certain connexins, in particular connexin26. By performing immunofluorescence staining after antigen retrieval of paraffin-embedded cervical tissue samples, this study reports for the first time that connexin26 and connexin30, in addition to connexin43, are expressed in differentiating cells of normal human cervical epithelia. Moreover, in dysplastic ectocervix, all connexins studied display a dramatic loss of expression compared to adjacent normal epithelia.     Enacting participatory, gender-sensitive slum redevelopment? Urban governance, power and participation in Trivandrum, Kerala

    This paper looks at two governance challenges that sit behind global commitments to deliver ‘cities without slums’: under what conditions can participatory ideals be successfully transferred to housing redevelopment programmes, and under what conditions can participatory slum redevelopment trigger wider shifts towards inclusive urban governance? It does so by examining Indian national slum redevelopment policy and its implementation in Kerala’s capital city, Trivandrum (Thiruvananthapuram). Kerala’s track-record of participatory governance and the lead role given to its women-focused poverty alleviation mission, Kudumbashree, in implementing housing projects make it an ideal place in which to examine these questions, and their gender implications. Primary data focusing on two housing projects are used to contrast intended governance changes featuring female-centred community participation with their actual operation on the ground. Despite moves to foreground women’s engagement, both projects suffered from shortfalls in institutional design, the inevitable administrative complexity of housing delivery, and resistance from local power brokers. Given Kerala’s favourable starting conditions, these outcomes highlight the need for slum redevelopment to be based around a deeper analysis of power dynamics and the explicit articulation of an agenda for inclusion at a city-level if participation is to realise its transformative potential

    Processing quantum information with relativistic motion of atoms

    We show that particle detectors, such as two-level atoms, in noninertial motion (or in gravitational fields) could be used to build quantum gates for the processing of quantum information. Concretely, we show that through suitably chosen noninertial trajectories of the detectors the interaction Hamiltonian's time dependence can be modulated to yield arbitrary rotations in the Bloch sphere due to relativistic quantum effects

    Global Wheat Head Detection (GWHD) dataset: a large and diverse dataset of high resolution RGB labelled images to develop and benchmark wheat head detection methods

    Detection of wheat heads is an important task allowing to estimate pertinent traits including head population density and head characteristics such as sanitary state, size, maturity stage and the presence of awns. Several studies developed methods for wheat head detection from high-resolution RGB imagery. They are based on computer vision and machine learning and are generally calibrated and validated on limited datasets. However, variability in observational conditions, genotypic differences, development stages, head orientation represents a challenge in computer vision. Further, possible blurring due to motion or wind and overlap between heads for dense populations make this task even more complex. Through a joint international collaborative effort, we have built a large, diverse and well-labelled dataset, the Global Wheat Head detection (GWHD) dataset. It contains 4,700 high-resolution RGB images and 190,000 labelled wheat heads collected from several countries around the world at different growth stages with a wide range of genotypes. Guidelines for image acquisition, associating minimum metadata to respect FAIR principles and consistent head labelling methods are proposed when developing new head detection datasets.     When assessment defines the content—understanding goals in between teachers and policy

In this paper, teachers’ perceptions about their work are explored, as goals and assessment play a more prominent role driven by the introduction of a learning outcomes‐oriented system. This is investigated through interviews of Norwegian teachers and extensive policy analysis of Norwegian policy documents. The findings indicate that the teachers are finding ways to negotiate and adjust to the language in the policies investigated in this study. Furthermore, the findings show that the teachers have developed their professional language according to the policies. The teachers referred to their self‐made criteria and goal sheets as central tools in explicating what is to be learned. In many ways, the tools for assessment, thus determine the content of education as well as what is valued in the educational system.publishedVersio

    Human chondrogenic paraxial mesoderm, directed specification and prospective isolation from pluripotent stem cells

    Directed specification and prospective isolation of chondrogenic paraxial mesoderm progeny from human pluripotent stem (PS) cells have not yet been achieved. Here we report the successful generation of KDR−PDGFRα+ progeny expressing paraxial mesoderm genes and the mesendoderm reporter MIXL1-GFP in a chemically defined medium containing the canonical WNT signaling activator, BMP-inhibitor, and the Nodal/Activin/TGFβ signaling controller. Isolated (GFP+)KDR−PDGFRα+ mesoderm cells were sensitive to sequential addition of the three chondrogenic factors PDGF, TGFβ and BMP. Under these conditions, the cells showed robust chondrogenic activity in micromass culture, and generated a hyaline-like translucent cartilage particle in serum-free medium. In contrast, both STRO1+ mesenchymal stem/stromal cells from adult human marrow and mesenchymal cells spontaneously arising from hPS cells showed a relatively weaker chondrogenic response in vitro, and formed more of the fibrotic cartilage particles. Thus, hPS cell-derived KDR−PDGFRα+ paraxial mesoderm-like cells have potential in engineered cartilage formation and cartilage repair

    Holographic optical traps for atom-based topological Kondo devices

    The topological Kondo (TK) model has been proposed in solid-state quantum devices as a way to realize non-Fermi liquid behaviors in a controllable setting. Another motivation behind the TK model proposal is the demand to demonstrate the quantum dynamical properties of Majorana fermions, which are at the heart of their potential use in topological quantum computation. Here we consider a junction of crossed Tonks-Girardeau gases arranged in a star-geometry (forming a Y-junction), and we perform a theoretical analysis of this system showing that it provides a physical realization of the TK model in the realm of cold atom systems. Using computer-generated holography, we experimentally implement a Y-junction suitable for atom trapping, with controllable and independent parameters. The junction and the transverse size of the atom waveguides are of the order of 5 mu m, leading to favorable estimates for the Kondo temperature and for the coupling across the junction. Since our results show that all the required theoretical and experimental ingredients are available, this provides the demonstration of an ultracold atom device that may in principle exhibit the TK effect
