    Unusual red tide of the dinoflagellate Lingulodinium polyedra during an upwelling event off the Algarve coast (SW Iberia)

    Low biomass, but highly toxic algal blooms are frequent in the Ria Formosa coastal lagoon and adjacent coastal zone (southern Portugal), usually associated with specific oceanographic and nutrient conditions. In June 2019, a massive bloom of the dinoflagellate Lingulodinium polyedra developed along the coast, causing water discolouration, and the closure of beaches and of bivalve harvesting, with significant socioeconomic consequences. This study aims to evaluate the relationships between phytoplankton composition and environmental conditions before, during, and after the L. polyedra bloom. Water samples were collected in the Ria Formosa coastal lagoon throughout June and July 2019 for determination of nutrient and chlorophyll-a concentration using spectrophotometric methods. Phytoplankton composition and abundance were evaluated using inverted microscopy. Upwelling intensity was estimated considering a wind-based index, the cross-shore Ekman transport. Results show strong relationships between phytoplankton community composition and nutrients, particularly a positive relationship between centric diatoms and nitrogen, and a negative relationship between several dinoflagellate genera, and phosphorus and silicon. L. polyedra was not significantly associated with any nutrient variable, most likely due to its mixotrophic capabilities. A clear phytoplankton succession, typical of eastern boundary upwelling locations, was observed throughout the different bloom phases, from dominance of type II dinoflagellates (Heterocapsa, Prorocentrum) in the pre-bloom period, to dominance of L. polyedra (type V) during the bloom, and finally dominance of centric diatoms in the post-bloom period.     S-nitrosoglutathione reverts dietary sucrose-induced insulin resistance

    The liver is a fundamental organ to ensure whole-body homeostasis, allowing for a proper increase in insulin sensitivity from the fast to the postprandial status. Hepatic regulation of glucose metabolism is crucial and has been shown to be modulated by glutathione (GSH) and nitric oxide (NO). However, knowledge of the metabolic action of GSH and NO in glucose homeostasis remains incomplete. The current study was designed to test the hypothesis that treatment with S-nitrosoglutathione is sufficient to revert insulin resistance induced by a high-sucrose diet. Male Wistar rats were divided in a control or high-sucrose group. Insulin sensitivity was determined: (i) in the fast state; (ii) after a standardized test meal; (iii) after GSH + NO; and after (iv) S-nitrosoglutathione (GSNO) administration. The fasting glucose level was not different between the control and high-sucrose group. In the liver, the high-sucrose model shows increased NO and unchanged GSH levels. In control animals, insulin sensitivity increased after a meal or administration of GSH+NO/GSNO, but this was abrogated by sucrose feeding. GSNO was able to revert insulin resistance induced by sucrose feeding, in a dose-dependent manner, suggesting that they have an insulin-sensitizing effect in vivo. These effects are associated with an increased insulin receptor and Akt phosphorylation in muscle cells. Our findings demonstrate that GSNO promotes insulin sensitivity in a sucrose-induced insulin-resistant animal model and further implicates that this antioxidant molecule may act as a potential pharmacological tool for the treatment of insulin resistance in obesity and type 2 diabetes.publishersversionpublishe

    Nutritional value of selected macroalgae

Their protein and fiber content have been widely studied and differ according to the species, their geographic origin and their seasonal conditions. In addition to their value for human nutrition, seaweeds have multiple therapeutically applications (e.g., weight control, hypocholesterolemic, antioxidant and antitumor activities, others) and, in general, contribute and promote human health. In the archipelago of the Azores, the consumption of seaweeds is widespread and accepted as a common practice in some islands. This work is aimed at providing information on the protein and fiber content of the locally consumed species, to promote this regional food product that can be potentially profitable from the biotechnology and commercial perspective, and also benefit public health, particularly, taking into account the low level of marine pollution in the Azores archipelago. Protein and fiber content of eight seaweeds (Porphyra sp., Osmundea pinnatifida, Pterocladiella capillacea, Sphaerococcus coronopifolius, and Gelidium microdon, Rhodophyta; Cystoseira abies-marina and Fucus spiralis, Phaeophyta; Ulva compressa, Chlorophyta) were determined using the Kjeldahl method and the Weende method, respectively. The protein content ranged from 6.81 to 26.62 of dry weight for C. abies-marina and U. compressa, respectively. Fiber content was generally higher as compared with that in seaweeds from other origins and ranged from 33.82 to 63.88 for O. pinnatifida and F. spiralis, respectively

    Antioxidant activity of macroalgae from the Azores

    Free radical scavenging activity (FRSA) was studied to determine the antioxidant activity of 8 common macroalgae species found in the Azores. Species under study were Porphyra sp., Osmundea pinnatifida, Pterocladiella capillacea, Sphaerococcus coronopifolius and Gelidium microdon, from Rhodophyta; Ulva compressa and Chaetomorpha pachynema, from Chlorophyta and Fucus spiralis from Phaeophyta.The antioxidant activity was evaluated in methanolic extract by a decolourisation solution test of 2,2-diphenyl-1-picrylhydrazyl (DPPH), used as a stable radical. The methanolic extracts were obtained from dried biomass by sequential extractions, attaining a final concentration of 2 mg.mL-1. The FRSA values ranged from 19.54% for S. coronopifolius to 60.05% for F. spiralis with standard deviation (SD) varying between 1.41% and 6.80%. Results indicated that the studied seaweeds are a very promising source of biological active compounds with antioxidante properties. The seaweeds were collected in the Azorean islands where seawater pollution levels are low. Consequently, these seaweeds represent a valuable and good source of antioxidante material with superior beneficial effects on human health

    Adaptation and validation of the WHOQOL-OLD for the Portuguese population: Results of focus groups implementation

    No âmbito dos estudos de adaptação e validação do WHOQOL-OLD, foram implementados oito grupos focais, num total de 33 participantes, em amostra de conveniência: um com profissionais em formação (N=4), um com cuidadores (N=4) e seis grupos com adultos idosos (N=25; quatro com idosos da comunidade e dois com idosos institucionalizados). Os resultados corroboram a natureza multidimensional do constructo QdV. Foram evocados espontaneamente os seis domínios do WHOQOL-100 e todas as suas facetas (exceptuando Actividade sexual), bem como as seis facetas do WHOQOL-OLD. Com maior frequência, oadultos idosos destacam Qualidade de vida e saúde geral e dimensões de Relações sociais, Ambiente, Psicológico, Aspectos espirituais e Autonomia; os cuidadores referem dimensões de Autonomia, Saúde, Ambiente e Relações sociais; e os profissionais destacam Saúde, Nível de independência, Ambiente e Relações sociais. Por todos os grupos foi apontada a dimensão “Família/Vida familiar”, que poderá constituir-se como uma nova faceta do módulo WHOQOL-OLD português
