Slx5/Slx8-dependent ubiquitin hotspots on chromatin contribute to stress tolerance
Chromatin is a highly regulated environment, and protein association with chromatin is often controlled by post-translational modifications and the corresponding enzymatic machinery. Specifically, SUMO-targeted ubiquitin ligases (STUbLs) have emerged as key players in nuclear quality control, genome maintenance, and transcription. However, how STUbLs select specific substrates among myriads of SUMOylated proteins on chromatin remains unclear. Here, we reveal a remarkable co-localization of the budding yeast STUbL Slx5/Slx8 and ubiquitin at seven genomic loci that we term "ubiquitin hotspots". Ubiquitylation at these sites depends on Slx5/Slx8 and protein turnover on the Cdc48 segregase. We identify the transcription factor-like Ymr111c/Euc1 to associate with these sites and to be a critical determinant of ubiquitylation. Euc1 specifically targets Slx5/Slx8 to ubiquitin hotspots via bipartite binding of Slx5 that involves the Slx5 SUMO-interacting motifs and an additional, novel substrate recognition domain. Interestingly, the Euc1-ubiquitin hotspot pathway acts redundantly with chromatin modifiers of the H2A.Z and Rpd3L pathways in specific stress responses. Thus, our data suggest that STUbL-dependent ubiquitin hotspots shape chromatin during stress adaptation
A generalization of the van-der-Pol oscillator underlies active signal amplification in Drosophila hearing
The antennal hearing organs of the fruit fly Drosophila melanogaster boost their sensitivity by an active mechanical process that, analogous to the cochlear amplifier of vertebrates, resides in the motility of mechanosensory cells. This process nonlinearly improves the sensitivity of hearing and occasionally gives rise to self-sustained oscillations in the absence of sound. Time series analysis of self-sustained oscillations now unveils that the underlying dynamical system is well described by a generalization of the van-der-Pol oscillator. From the dynamic equations, the underlying amplification dynamics can explicitly be derived. According to the model, oscillations emerge from a combination of negative damping, which reflects active amplification, and a nonlinear restoring force that dictates the amplitude of the oscillations. Hence, active amplification in fly hearing seems to rely on the negative damping mechanism initially proposed for the cochlear amplifier of vertebrate
Electronic structure of the (111) and (-1-1-1) surfaces of cubic BN: A local-density-functional ab initio study
We present ab initio local-density-functional electronic structure
calculations for the (111) and (-1-1-1) surfaces of cubic BN. The energetically
stable reconstructions, namely the N adatom, N3 triangle models on the (111),
the (2x1), boron and nitrogen triangle patterns on the (-1-1-1) surface are
investigated. Band structure and properties of the surface states are discussed

Speckle Control with a remapped-pupil PIAA-coronagraph
The PIAA is a now well demonstrated high contrast technique that uses an
intermediate remapping of the pupil for high contrast coronagraphy
(apodization), before restoring it to recover classical imaging capabilities.
This paper presents the first demonstration of complete speckle control loop
with one such PIAA coronagraph. We show the presence of a complete set of
remapping optics (the so-called PIAA and matching inverse PIAA) is transparent
to the wavefront control algorithm. Simple focal plane based wavefront control
algorithms can thus be employed, without the need to model remapping effects.
Using the Subaru Coronagraphic Extreme AO (SCExAO) instrument built for the
Subaru Telescope, we show that a complete PIAA-coronagraph is compatible with a
simple implementation of a speckle nulling technique, and demonstrate the
benefit of the PIAA for high contrast imaging at small angular separation.Comment: 6 figures, submitted to PAS
Upper Limits On Periodic, Pulsed Radio Emission from the X-Ray Point Source in Cassiopeia A
The Chandra X-ray Observatory recently discovered an X-ray point source near
the center of Cassiopeia A, the youngest known Galactic supernova remnant. We
have conducted a sensitive search for radio pulsations from this source with
the Very Large Array, taking advantage of the high angular resolution of the
array to resolve out the emission from the remnant itself. No convincing
signatures of a dispersed, periodic source or of isolated dispersed pulses were
found, whether for an isolated or a binary source. We derive upper limits of 30
and 1.3 mJy at 327 and 1435 MHz for the phase-averaged pulsed flux density from
this source. The corresponding luminosity limits are lower than those for any
pulsar with age less than 10^4 years. The sensitivities of our search to single
pulses were 25 and 1.0 Jy at 327 and 1435 MHz. For comparison, the Crab pulsar
emits roughly 80 pulses per minute with flux densities greater than 100 Jy at
327 MHz and 8 pulses per minute with flux densities greater than 50 Jy at 1435
MHz. These limits are consistent with the suggestion that the X-ray point
source in Cas A adds to the growing number of neutron stars which are not radio

Mid-infrared laser light nulling experiment using single-mode conductive waveguides
Aims: In the context of space interferometry missions devoted to the search
of exo-Earths, this paper investigates the capabilities of new single mode
conductive waveguides at providing modal filtering in an infrared and
monochromatic nulling experiment; Methods: A Michelson laser interferometer
with a co-axial beam combination scheme at 10.6 microns is used. After
introducing a Pi phase shift using a translating mirror, dynamic and static
measurements of the nulling ratio are performed in the two cases where modal
filtering is implemented and suppressed. No additional active control of the
wavefront errors is involved. Results: We achieve on average a statistical
nulling ratio of 2.5e-4 with a 1-sigma upper limit of 6e-4, while a best null
of 5.6e-5 is obtained in static mode. At the moment, the impact of external
vibrations limits our ability to maintain the null to 10 to 20 seconds.;
Conclusions: A positive effect of SM conductive waveguide on modal filtering
has been observed in this study. Further improvement of the null should be

Family memories in the home: contrasting physical and digital mementos
We carried out fieldwork to characterise and compare physical and digital mementos in the home. Physical mementos are highly valued, heterogeneous and support different types of recollection. Contrary to expectations, we found physical mementos are not purely representational, and can involve appropriating common objects and more idiosyncratic forms. In contrast, digital mementos were initially perceived as less valuable, although participants later reconsidered this. Digital mementos were somewhat limited in function and expression, largely involving representational photos and videos, and infrequently accessed. We explain these digital limitations and conclude with design guidelines for digital mementos, including better techniques for accessing and integrating these into everyday life, allowing them to acquire the symbolic associations and lasting value that characterise their physical counterparts
