Slx5/Slx8-dependent ubiquitin hotspots on chromatin contribute to stress tolerance
Chromatin is a highly regulated environment, and protein association with chromatin is often controlled by post-translational modifications and the corresponding enzymatic machinery. Specifically, SUMO-targeted ubiquitin ligases (STUbLs) have emerged as key players in nuclear quality control, genome maintenance, and transcription. However, how STUbLs select specific substrates among myriads of SUMOylated proteins on chromatin remains unclear. Here, we reveal a remarkable co-localization of the budding yeast STUbL Slx5/Slx8 and ubiquitin at seven genomic loci that we term "ubiquitin hotspots". Ubiquitylation at these sites depends on Slx5/Slx8 and protein turnover on the Cdc48 segregase. We identify the transcription factor-like Ymr111c/Euc1 to associate with these sites and to be a critical determinant of ubiquitylation. Euc1 specifically targets Slx5/Slx8 to ubiquitin hotspots via bipartite binding of Slx5 that involves the Slx5 SUMO-interacting motifs and an additional, novel substrate recognition domain. Interestingly, the Euc1-ubiquitin hotspot pathway acts redundantly with chromatin modifiers of the H2A.Z and Rpd3L pathways in specific stress responses. Thus, our data suggest that STUbL-dependent ubiquitin hotspots shape chromatin during stress adaptation
Speckle Control with a remapped-pupil PIAA-coronagraph
The PIAA is a now well demonstrated high contrast technique that uses an
intermediate remapping of the pupil for high contrast coronagraphy
(apodization), before restoring it to recover classical imaging capabilities.
This paper presents the first demonstration of complete speckle control loop
with one such PIAA coronagraph. We show the presence of a complete set of
remapping optics (the so-called PIAA and matching inverse PIAA) is transparent
to the wavefront control algorithm. Simple focal plane based wavefront control
algorithms can thus be employed, without the need to model remapping effects.
Using the Subaru Coronagraphic Extreme AO (SCExAO) instrument built for the
Subaru Telescope, we show that a complete PIAA-coronagraph is compatible with a
simple implementation of a speckle nulling technique, and demonstrate the
benefit of the PIAA for high contrast imaging at small angular separation.Comment: 6 figures, submitted to PAS
A generalization of the van-der-Pol oscillator underlies active signal amplification in Drosophila hearing
The antennal hearing organs of the fruit fly Drosophila melanogaster boost their sensitivity by an active mechanical process that, analogous to the cochlear amplifier of vertebrates, resides in the motility of mechanosensory cells. This process nonlinearly improves the sensitivity of hearing and occasionally gives rise to self-sustained oscillations in the absence of sound. Time series analysis of self-sustained oscillations now unveils that the underlying dynamical system is well described by a generalization of the van-der-Pol oscillator. From the dynamic equations, the underlying amplification dynamics can explicitly be derived. According to the model, oscillations emerge from a combination of negative damping, which reflects active amplification, and a nonlinear restoring force that dictates the amplitude of the oscillations. Hence, active amplification in fly hearing seems to rely on the negative damping mechanism initially proposed for the cochlear amplifier of vertebrate
Years of RXTE Monitoring of Anomalous X-ray Pulsar 4U 0142+61: Long-Term Variability
We report on 10 years of monitoring of the 8.7-s Anomalous X-ray Pulsar 4U
0142+61 using the Rossi X-Ray Timing Explorer (RXTE). This pulsar exhibited
stable rotation from 2000 March until 2006 February: the RMS phase residual for
a spin-down model which includes nu, nudot, and nuddot is 2.3%. We report a
possible phase-coherent timing solution valid over a 10-yr span extending back
to March 1996. A glitch may have occured between 1998 and 2000, but is not
required by the existing timing data. The pulse profile has been evolving since
2000. In particular, the dip of emission between its two peaks got shallower
between 2002 and 2006, as if the profile were evolving back to its pre-2000
morphology, following an earlier event, which possibly also included the glitch
suggested by the timing data. These profile variations are seen in the 2-4 keV
band but not in 6-8 keV. We also detect a slow increase in the pulsed flux
between 2002 May and 2004 December, such that it has risen by 36+/-3% over 2.6
years in the 2-10 keV band. The pulsed flux variability and the narrow-band
pulse profile changes present interesting challenges to aspects of the magnetar

In-plane magnetic anisotropy of Fe atoms on BiSe(111)
The robustness of the gapless topological surface state hosted by a 3D
topological insulator against perturbations of magnetic origin has been the
focus of recent investigations. We present a comprehensive study of the
magnetic properties of Fe impurities on a prototypical 3D topological insulator
BiSe using local low temperature scanning tunneling microscopy and
integral x-ray magnetic circular dichroism techniques. Single Fe adatoms on the
BiSe surface, in the coverage range are heavily relaxed
into the surface and exhibit a magnetic easy axis within the surface-plane,
contrary to what was assumed in recent investigations on the opening of a gap.
Using \textit{ab initio} approaches, we demonstrate that an in-plane easy axis
arises from the combination of the crystal field and dynamic hybridization

Electronic structure of the (111) and (-1-1-1) surfaces of cubic BN: A local-density-functional ab initio study
We present ab initio local-density-functional electronic structure
calculations for the (111) and (-1-1-1) surfaces of cubic BN. The energetically
stable reconstructions, namely the N adatom, N3 triangle models on the (111),
the (2x1), boron and nitrogen triangle patterns on the (-1-1-1) surface are
investigated. Band structure and properties of the surface states are discussed
