Viral vectors encoding endomorphins and serine histogranin attenuate neuropathic pain symptoms after spinal cord injury in rats
Background: The treatment of spinal cord injury (SCI)-induced neuropathic pain presents a challenging healthcare problem. The lack of available robust pharmacological treatments underscores the need for novel therapeutic methods and approaches. Due to the complex character of neuropathic pain following SCI, therapies targeting multiple mechanisms may be a better choice for obtaining sufficient long-term pain relief. Previous studies in our lab showed analgesic effects using combinations of an NMDA antagonist peptide Ser1histogranin (SHG), and the mu-opioid peptides endomorphins (EMs), in several pain models. As an alternative to drug therapy, this study evaluated the analgesic potential of these peptides when delivered via gene therapy. Results: Lentiviruses encoding SHG and EM-1 and EM-2 were intraspinally injected, either singly or in combination, into rats with clip compression SCI 2weeks following injury. Treated animals showed significant reduction in mechanical and thermal hypersensitivity, compared to control groups injected with GFP vector only. The antinociceptive effects of individually injected components were modest, but the combination of EMs and SHG produced robust and sustained antinociception. The onset of the analgesic effects was observed between 1-5 weeks post-injection and sustained without decrement for at least 7weeks. No adverse effects on locomotor function were observed. The involvement of SHG and EMs in the observed antinociception was confirmed by pharmacologic inhibition using intrathecal injection of either the opioid antagonist naloxone or an anti-SHG antibody. Immunohistochemical analysis showed the presence of SHG and EMs in the spinal cord of treated animals, and immunodot-blot analysis of CSF confirmed the presence of these peptides in injected animals. In a separate group of rats, delayed injection of viral vectors was performed in order to mimic a more likely clinical scenario. Comparable and sustained antinociceptive effects were observed in these animals using the SHG-EMs combination vectors compared to the group with early intervention. Conclusions: Findings from this study support the potential for direct gene therapy to provide a robust and sustained alleviation of chronic neuropathic pain following SCI. The combination strategy utilizing potent mu-opioid peptides with a naturally-derived NMDA antagonist may produce additive or synergistic analgesic effects without the tolerance development for long-term management of persistent pain. © 2015 Nasirinezhad et al

The Pure Virtual Braid Group Is Quadratic
If an augmented algebra K over Q is filtered by powers of its augmentation
ideal I, the associated graded algebra grK need not in general be quadratic:
although it is generated in degree 1, its relations may not be generated by
homogeneous relations of degree 2. In this paper we give a sufficient criterion
(called the PVH Criterion) for grK to be quadratic. When K is the group algebra
of a group G, quadraticity is known to be equivalent to the existence of a (not
necessarily homomorphic) universal finite type invariant for G. Thus the PVH
Criterion also implies the existence of such a universal finite type invariant
for the group G. We apply the PVH Criterion to the group algebra of the pure
virtual braid group (also known as the quasi-triangular group), and show that
the corresponding associated graded algebra is quadratic, and hence that these

Curved Koszul duality theory
38 pagesInternational audienceWe extend the bar-cobar adjunction to operads and properads, not necessarily augmented. Due to the default of augmentation, the objects of the dual category are endowed with a curvature. We handle the lack of augmentation by extending the category of coproperads to include objects endowed with a curvature. As usual, the bar-cobar construction gives a (large) cofibrant resolution for any properad, such as the properad encoding unital and counital Frobenius algebras, a notion which appears in 2d-TQFT. We also define a curved Koszul duality theory for operads or properads presented with quadratic, linear and constant relations, which provides the possibility for smaller relations. We apply this new theory to study the homotopy theory and the cohomology theory of unital associative algebras

The Variation of Mechanical Properties of M300 Maraging Steel Manufactured with Varying Process Parameters in Laser Powder Bed Fusion
Laser power bed fusion (L-PBF) is a type of additive manufacturing (AM) that uses layers
of powdered metal and a laser to manufacture a part in a layer-by-layer fashion. L-PBF uses a
variety of process parameters that ultimately determine the overall quality and mechanical
properties of a print. The ability to alter parameters allows for the utilization of various metals in
this form of AM. Maraging 300 steel (M300) is a material of particular interest due to its combined
tensile strength and high strength-to-weight ratio. By using an assortment of parameters and
comparing the resulting mechanical properties it can be determined which process parameters
result in a more favorable part to be used in a variety of applications. A favorable process parameter
set was selected for future use. This study aims to determine which process parameters result in
the best overall mechanical properties of M300 manufactured using L-PBF.Mechanical Engineerin
Supersymmetric Deformations of Maximally Supersymmetric Gauge Theories
We study supersymmetric and super Poincar\'e invariant deformations of
ten-dimensional super Yang-Mills theory and of its dimensional reductions. We
describe all infinitesimal super Poincar\'e invariant deformations of equations
of motion of ten-dimensional super Yang-Mills theory and its reduction to a
point; we discuss the extension of them to formal deformations. Our methods are
based on homological algebra, in particular, on the theory of L-infinity and
A-infinity algebras. The exposition of this theory as well as of some basic
facts about Lie algebra homology and Hochschild homology is given in

Real World Data in Adaptive Biomedical Innovation: A Framework for Generating Evidence Fit for Decision-Making
Analyses of healthcare databases (claims, electronic health records [EHRs]) are useful supplements to clinical trials for generating evidence on the effectiveness, harm, use, and value of medical products in routine care. A constant stream of data from the routine operation of modern healthcare systems, which can be analyzed in rapid cycles, enables incremental evidence development to support accelerated and appropriate access to innovative medicines. Evidentiary needs by regulators, Health Technology Assessment, payers, clinicians, and patients after marketing authorization comprise (1) monitoring of medication performance in routine care, including the materialized effectiveness, harm, and value; (2) identifying new patient strata with added value or unacceptable harms; and (3) monitoring targeted utilization. Adaptive biomedical innovation (ABI) with rapid cycle database analytics is successfully enabled if evidence is meaningful, valid, expedited, and transparent. These principles will bring rigor and credibility to current efforts to increase research efficiency while upholding evidentiary standards required for effective decision-making in healthcare
