Assessment of Left Atrial Deformation and Function by 2-Dimensional Speckle Tracking Echocardiography in Healthy Dogs and Dogs With Myxomatous Mitral Valve Disease
open7noBackground: The assessment of left atrial (LA) function by 2-dimensional speckle tracking echocardiography (STE) holds important clinical implications in human medicine. Few similar data are available in dogs. Objectives: To assess LA function by STE in dogs with and without myxomatous mitral valve disease (MMVD), analyzing LA areas, systolic function, and strain. Animals: One hundred and fifty dogs were divided according to the American College of Veterinary Internal Medicine classification of heart failure: 23 dogs in class A, 52 in class B1, 36 in class B2, and 39 in class C + D. Methods: Prospective observational study. Conventional morphologic and Doppler variables, LA areas, and STE-based LA strain analysis were performed in all dogs and results were compared among groups. Correlation analysis was carried out between LA STE variables and other echocardiographic variables. Results: Variability study showed good reproducibility for all the tested variables (coefficient of variation <16%). Left atrial areas, fractional area change, peak atrial longitudinal strain (PALS), peak atrial contraction strain, and contraction strain index (CSI) differed significantly between groups B2 and C + D and all the other groups (overall P < .001), whereas only PALS differed between groups B1 and A (P = .01). Left atrial areas increased with progression of the disease, whereas LA functional parameters decreased. Only CSI increased nonsignificantly from group A to group B1 and then progressively decreased. Thirty-one significant correlations (P < .001, r > .3) were found between conventional left heart echocardiographic variables and LA areas and strain variables. IMPA versus Cloud Analysis and IDA: Different Methods to Evaluate Structural Seismic Fragility
Well-known methods for seismic performance assessment, such as incremental dynamic analysis (IDA), multi-stripes analysis (MSA) and the cloud method, involve nonlinear response time-history analyses to characterize the relationship between the chosen damage measure versus intensity measure. Over the past two decades, many authors have proposed simplified procedures or nonlinear static approaches to develop fragility. In these procedures, the capacity of the system is evaluated by nonlinear static procedures (i.e., the capacity spectrum method (CSM), the N2 method, modal pushover analysis (MPA)) and the demand is derived by response spectra. In addition to the familiar ones, incremental modal pushover analysis (IMPA) is a novel nonlinear static procedure proposed in recent years, and it is used in this research to present an IM-based fragility estimation. The accuracy and effectiveness of different methods to assess vulnerability are investigated by comparing fragility curves derived by MPA-based cloud analysis, IMPA and cloud analysis against IDA. The comparison gives valuable insights on the influence of scaling on different sets of records; however, a more extended validation is needed to confirm the obtained results and draw more general conclusions. Results arise from two relatively small bins of record motions differing by ranges of Joyner-Boore distance and scattered in a range of magnitude are presented

:.Diabetes mellitus is a major of health problems. Incomprehension of patients about therapy increase noncompliance in taking insulin. Hypoglycemia is a condition which blood glucose below normal or less than 70 mg/dl. The purpose of this research is to know about the knowledge relationship with the use of insulin with hypoglycemia in patients in the clinic interna of GMIM Pancaran Kasih Manado hospital. This research is a descriptive analytic with cross sectional design. The sampling method used to total sampling that consist of 60 respondents.The research instrument is an interview of the 10 statements about the use of insulin and knowledge and questionnaire 7 questions about hypoglicemia .Theanswers already collected is being analized using the Chi-square test and counted by SPSS. The result of this research which is gain the trusted level of 95% show value p=0,005. This pvalue is smaller than á = 0,05. The conclusion of this research is showed that a significant relationshipabout the knowledge relationship with the use of insulin with hypoglycemia n patients in the clinic interna of GMIM Pancaran Kasih Manado hospital
On the thickness of the double layer in ionic liquids
In this study, we examined the thickness of the electrical double layer (EDL)
in ionic liquids using density functional theory (DFT) calculations and
molecular dynamics (MD) simulations. We focused on the BF4- anion adsorption
from 1-ethyl-3-methylimidazolium tetrafluoroborate (EMImBF4) ionic liquid on
the Au(111) surface. At both DFT and MD levels, we evaluated the
capacitance-potential dependence for the Helmholtz model of the interface.
Using MD simulations, we also explored a more realistic, multilayer EDL model
accounting for the ion layering. Concurrent analysis of the DFT and MD results
provides a ground for thinking whether the electrical double layer in ionic
liquids is one- or multi-ionic-layer thick
NFV service dynamicity with a DevOps approach : demonstrating zero-touch deployment & operations
Next generation network services will be realized by NFV-based microservices to enable greater dynamics in deployment and operations. Here, we present a demonstrator that realizes this concept using the NFV platform built in the EU FP7 project UNIFY. Using the example of an Elastic Router service, we show automated deployment and configuration of service components as well as corresponding monitoring components facilitating automated scaling of the entire service. We also demonstrate automatic execution of troubleshooting and debugging actions. Operations of the service are inspired by DevOps principles, enabling quick detection of operational conditions and fast corrective actions. This demo conveys essential insights on how the life-cycle of an NFV-based network service may be realized in future NFV platforms
Paradigm of biased PAR1 (protease-activated receptor-1) activation and inhibition in endothelial cells dissected by phosphoproteomics
Thrombin is the key serine protease of the coagulation cascade and mediates cellular responses by activation of PARs (protease-activated receptors). The predominant thrombin receptor is PAR1, and in endothelial cells (ECs), thrombin dynamically regulates a plethora of phosphorylation events. However, it has remained unclear whether thrombin signaling is exclusively mediated through PAR1. Furthermore, mechanistic insight into activation and inhibition of PAR1-mediated EC signaling is lacking. In addition, signaling networks of biased PAR1 activation after differential cleavage of the PAR1 N terminus have remained an unresolved issue. Here, we used a quantitative phosphoproteomics approach to show that classical and peptide activation of PAR1 induce highly similar signaling, that low thrombin concentrations initiate only limited phosphoregulation, and that the PAR1 inhibitors vorapaxar and parmodulin-2 demonstrate distinct antagonistic properties. Subsequent analysis of the thrombin-regulated phosphosites in the presence of PAR1 inhibitors revealed that biased activation of PAR1 is not solely linked to a specific G-protein downstream of PAR1. In addition, we showed that only the canonical thrombin PAR1 tethered ligand induces extensive early phosphoregulation in ECs. Our study provides detailed insight in the signaling mechanisms downstream of PAR1. Our data demonstrate that thrombin-induced EC phosphoregulation is mediated exclusively through PAR1, that thrombin and thrombin-tethered ligand peptide induce similar phosphoregulation, and that only canonical PAR1 cleavage by thrombin generates a tethered ligand that potently induces early signaling. Furthermore, platelet PAR1 inhibitors directly affect EC signaling, indicating that it will be a challenge to design a PAR1 antagonist that will target only those pathways responsible for tissue pathology
Maternal modulation of paternal effects on offspring development.
The paternal transmission of environmentally induced phenotypes across generations has been reported to occur following a number of qualitatively different exposures and appear to be driven, at least in part, by epigenetic factors that are inherited via the sperm. However, previous studies of paternal germline transmission have not addressed the role of mothers in the propagation of paternal effects to offspring. We hypothesized that paternal exposure to nutritional restriction would impact male mate quality and subsequent maternal reproductive investment with consequences for the transmission of paternal germline effects. In the current report, using embryo transfer in mice, we demonstrate that sperm factors in adult food restricted males can influence growth rate, hypothalamic gene expression and behaviour in female offspring. However, under natural mating conditions females mated with food restricted males show increased pre- and postnatal care, and phenotypic outcomes observed during embryo transfer conditions are absent or reversed. We demonstrate that these compensatory changes in maternal investment are associated with a reduced mate preference for food restricted males and elevated gene expression within the maternal hypothalamus. Therefore, paternal experience can influence offspring development via germline inheritance, but mothers can serve as a modulating factor in determining the impact of paternal influences on offspring development
