    Analysis of Li-ion battery degradation using self-organizing maps

    This paper proposes a new methodology to identify the different degradation processes of Li-Ion battery cells. The goal of this study is to determine if different degradation factors can be separated by waveform analysis from aged cells with similar remaining capacity. In contrast to other works, the proposed method identifies the past operating conditions in the cell, regardless of the actual State of Health. The methodology is based on a data-driven approach by using a SOM (Self-organizing map), an unsupervised neural network. To verify the hypothesis a SOM has been trained with laboratory data from whole data cycles, to classify cells concerning their degradation path and according to their discharge voltage patterns. Additionally, this new methodology based on the SOM allows discriminating groups of cells with different cycling conditions (based on depth of discharge, ambient temperature and discharge current). This research line is very promising for classification of used cells, not only depending on their current static parameters (capacity, impedance), but also the battery use in their past life. This will allow making predictions of the Remaining Useful Life (RUL) of a battery with greater precision

    A step back on the conservation of a highly threatened species: opposite signs of recovery on Pinna nobilis population from Mar Menor lagoon

    The endemic species Pinna nobilis is the most endangered Mediterranean bivalve, facing nearly extinction all over the Mediterranean Sea, hosting its last reservoirs in highly impacted coastal lagoons. Thus, knowledge about the populations' conservation status in these ecosystems is essential. In 2019, the Mar Menor lagoon’s population was considered as a highly vulnerable population (Nebot-Colomer et al., 2021) due to several ecological disasters. The present study represents a continuation of the previous work, which aims to evaluate the resilience of the population, by assessing its reproductive success and maintenance of the population. To do so, between 2019 to 2022, we installed between 23-45 larvae collectors, monitored 13 permanent individual’s plots, and conducted visual searches and censuses. Overall, densities remained stable over years, although the number of individuals alive monitored in permanent plots decrease each year. Moreover, none of the methodologies carried out detected the incorporation of recruits in the population. Our results pointed out to opposite signs of recovery of the species, increasing its vulnerability to future disturbances. Therefore, urgent management and conservation actions focused on restoring the ecosystem and protecting P. nobilis individuals are needed to avoid this population extinction

    The CACCC-binding protein KLF3/BKLF represses a subset of KLF1/EKLF target genes and is required for proper erythroid maturation in vivo

    The CACCC-box binding protein erythroid Kruppel-like factor (EKLF/KLF1) is a master regulator that directs the expression of many important erythroid genes. We have previously shown that EKLF drives transcription of the gene for a second KLF, basic Kruppel-like factor, or KLF3. We have now tested the in vivo role of KLF3 in erythroid cells by examining Klf3 knockout mice. KLF3-deficient adults exhibit a mild compensated anemia, including enlarged spleens, increased red pulp, and a higher percentage of erythroid progenitors, together with elevated reticulocytes and abnormal erythrocytes in the peripheral blood. Impaired erythroid maturation is also observed in the fetal liver. We have found that KLF3 levels rise as erythroid cells mature to become TER119(+). Consistent with this, microarray analysis of both TER119(-) and TER119(+) erythroid populations revealed that KLF3 is most critical at the later stages of erythroid maturation and is indeed primarily a transcriptional repressor. Notably, many of the genes repressed by KLF3 are also known to be activated by EKLF. However, the majority of these are not currently recognized as erythroid-cell-specific genes. These results reveal the molecular and physiological function of KLF3, defining it as a feedback repressor that counters the activity of EKLF at selected target genes to achieve normal erythropoiesis

    EcoTILLING in Capsicum species: searching for new virus resistances

    <p>Abstract</p> <p>Background</p> <p>The EcoTILLING technique allows polymorphisms in target genes of natural populations to be quickly analysed or identified and facilitates the screening of genebank collections for desired traits. We have developed an EcoTILLING platform to exploit <it>Capsicum </it>genetic resources. A perfect example of the utility of this EcoTILLING platform is its application in searching for new virus-resistant alleles in <it>Capsicum </it>genus. Mutations in translation initiation factors (eIF4E, eIF(iso)4E, eIF4G and eIF(iso)4G) break the cycle of several RNA viruses without affecting the plant life cycle, which makes these genes potential targets to screen for resistant germplasm.</p> <p>Results</p> <p>We developed and assayed a cDNA-based EcoTILLING platform with 233 cultivated accessions of the genus <it>Capsicum</it>. High variability in the coding sequences of the <it>eIF4E </it>and <it>eIF(iso)4E </it>genes was detected using the cDNA platform. After sequencing, 36 nucleotide changes were detected in the CDS of <it>eIF4E </it>and 26 in <it>eIF(iso)4E</it>. A total of 21 <it>eIF4E </it>haplotypes and 15 <it>eIF(iso)4E </it>haplotypes were identified. To evaluate the functional relevance of this variability, 31 possible eIF4E/eIF(iso)4E combinations were tested against <it>Potato virus Y</it>. The results showed that five new <it>eIF4E </it>variants (<it>pvr2<sup>10</sup></it>, <it>pvr2<sup>11</sup></it>, <it>pvr2<sup>12</sup></it>, <it>pvr2<sup>13 </sup></it>and <it>pvr2<sup>14</sup></it>) were related to PVY-resistance responses.</p> <p>Conclusions</p> <p>EcoTILLING was optimised in different <it>Capsicum </it>species to detect allelic variants of target genes. This work is the first to use cDNA instead of genomic DNA in EcoTILLING. This approach avoids intronic sequence problems and reduces the number of reactions. A high level of polymorphism has been identified for initiation factors, showing the high genetic variability present in our collection and its potential use for other traits, such as genes related to biotic or abiotic stresses, quality or production. Moreover, the new <it>eIF4E </it>and <it>eIF(iso)4E </it>alleles are an excellent collection for searching for new resistance against other RNA viruses.</p

    Brain structural correlates of sensory phenomena in patients with obsessive-compulsive disorder

    Background: sensory phenomena (SP) are uncomfortable feelings, including bodily sensations, sense of inner tension, 'just-right' perceptions, feelings of incompleteness, or 'urge-only' phenomena, which have been described to precede, trigger or accompany repetitive behaviours in individuals with obsessive-compulsive disorder (OCD). Sensory phenomena are also observed in individuals with tic disorders, and previous research suggests that sensorimotor cortex abnormalities underpin the presence of SP in such patients. However, to our knowledge, no studies have assessed the neural correlates of SP in patients with OCD. Methods: we assessed the presence of SP using the University of São Paulo Sensory Phenomena Scale in patients with OCD and healthy controls from specialized units in São Paulo, Brazil, and Barcelona. All participants underwent a structural magnetic resonance examination, and brain images were examined using DARTEL voxel-based morphometry. We evaluated grey matter volume differences between patients with and without SP and healthy controls within the sensorimotor and premotor cortices. Results: we included 106 patients with OCD and 87 controls in our study. Patients with SP (67% of the sample) showed grey matter volume increases in the left sensorimotor cortex in comparison to patients without SP and bilateral sensorimotor cortex grey matter volume increases in comparison to controls. No differences were observed between patients without SP and controls. Limitations: most patients were medicated. Participant recruitment and image acquisition were performed in 2 different centres. Conclusion: we have identified a structural correlate of SP in patients with OCD involving grey matter volume increases within the sensorimotor cortex; this finding is in agreement with those of tic disorder studies showing that abnormal activity and volume increases within this region are associated with the urges preceding tic onset
