Is Gliese 581d habitable? Some constraints from radiative-convective climate modeling
The recently discovered exoplanet Gl581d is extremely close to the outer edge
of its system's habitable zone, which has led to much speculation on its
possible climate. We have performed a range of simulations to assess whether,
given simple combinations of chemically stable greenhouse gases, the planet
could sustain liquid water on its surface. For best estimates of the surface
gravity, surface albedo and cloud coverage, we find that less than 10 bars of
CO2 is sufficient to maintain a global mean temperature above the melting point
of water. Furthermore, even with the most conservative choices of these
parameters, we calculate temperatures above the water melting point for CO2
partial pressures greater than about 40 bar. However, we note that as Gl581d is
probably in a tidally resonant orbit, further simulations in 3D are required to
test whether such atmospheric conditions are stable against the collapse of CO2

Global modelling of the early Martian climate under a denser CO2 atmosphere: Water cycle and ice evolution
We discuss 3D global simulations of the early Martian climate that we have
performed assuming a faint young Sun and denser CO2 atmosphere. We include a
self-consistent representation of the water cycle, with atmosphere-surface
interactions, atmospheric transport, and the radiative effects of CO2 and H2O
gas and clouds taken into account. We find that for atmospheric pressures
greater than a fraction of a bar, the adiabatic cooling effect causes
temperatures in the southern highland valley network regions to fall
significantly below the global average. Long-term climate evolution simulations
indicate that in these circumstances, water ice is transported to the highlands
from low-lying regions for a wide range of orbital obliquities, regardless of
the extent of the Tharsis bulge. In addition, an extended water ice cap forms
on the southern pole, approximately corresponding to the location of the
Noachian/Hesperian era Dorsa Argentea Formation. Even for a multiple-bar CO2
atmosphere, conditions are too cold to allow long-term surface liquid water.
Limited melting occurs on warm summer days in some locations, but only for
surface albedo and thermal inertia conditions that may be unrealistic for water
ice. Nonetheless, meteorite impacts and volcanism could potentially cause
intense episodic melting under such conditions. Because ice migration to higher
altitudes is a robust mechanism for recharging highland water sources after
such events, we suggest that this globally sub-zero, `icy highlands' scenario
for the late Noachian climate may be sufficient to explain most of the fluvial
geology without the need to invoke additional long-term warming mechanisms or

Receptor and secreted targets of Wnt-1/β-catenin signalling in mouse mammary epithelial cells
BACKGROUND: Deregulation of the Wnt/ β-catenin signal transduction pathway has been implicated in the pathogenesis of tumours in the mammary gland, colon and other tissues. Mutations in components of this pathway result in β-catenin stabilization and accumulation, and the aberrant modulation of β-catenin/TCF target genes. Such alterations in the cellular transcriptional profile are believed to underlie the pathogenesis of these cancers. We have sought to identify novel target genes of this pathway in mouse mammary epithelial cells. METHODS: Gene expression microarray analysis of mouse mammary epithelial cells inducibly expressing a constitutively active mutant of β-catenin was used to identify target genes of this pathway. RESULTS: The differential expression in response to ΔNβ-catenin for five putative target genes, Autotaxin, Extracellular Matrix Protein 1 (Ecm1), CD14, Hypoxia-inducible gene 2 (Hig2) and Receptor Activity Modifying Protein 3 (RAMP3), was independently validated by northern blotting. Each of these genes encodes either a receptor or a secreted protein, modulation of which may underlie the interactions between Wnt/β-catenin tumour cells and between the tumour and its microenvironment. One of these genes, Hig2, previously shown to be induced by both hypoxia and glucose deprivation in human cervical carcinoma cells, was strongly repressed upon ΔNβ-catenin induction. The predicted N-terminus of Hig2 contains a putative signal peptide suggesting it might be secreted. Consistent with this, a Hig2-EGFP fusion protein was able to enter the secretory pathway and was detected in conditioned medium. Mutation of critical residues in the putative signal sequence abolished its secretion. The expression of human HIG2 was examined in a panel of human tumours and was found to be significantly downregulated in kidney tumours compared to normal adjacent tissue. CONCLUSIONS: HIG2 represents a novel non-cell autonomous target of the Wnt pathway which is potentially involved in human cancer
Systemic therapy of Cushing’s syndrome
Cushing’s disease (CD) in a stricter sense derives from pathologic adrenocorticotropic hormone (ACTH) secretion usually triggered by micro- or macroadenoma of the pituitary gland. It is, thus, a form of secondary hypercortisolism. In contrast, Cushing’s syndrome (CS) describes the complexity of clinical consequences triggered by excessive cortisol blood levels over extended periods of time irrespective of their origin. CS is a rare disease according to the European orphan regulation affecting not more than 5/10,000 persons in Europe. CD most commonly affects adults aged 20–50 years with a marked female preponderance (1:5 ratio of male vs. female). Patient presentation and clinical symptoms substantially vary depending on duration and plasma levels of cortisol. In 80% of cases CS is ACTH-dependent and in 20% of cases it is ACTH-independent, respectively. Endogenous CS usually is a result of a pituitary tumor. Clinical manifestation of CS, apart from corticotropin-releasing hormone (CRH-), ACTH-, and cortisol-producing (malign and benign) tumors may also be by exogenous glucocorticoid intake. Diagnosis of hypercortisolism (irrespective of its origin) comprises the following: Complete blood count including serum electrolytes, blood sugar etc., urinary free cortisol (UFC) from 24 h-urine sampling and circadian profile of plasma cortisol, plasma ACTH, dehydroepiandrosterone, testosterone itself, and urine steroid profile, Low-Dose-Dexamethasone-Test, High-Dose-Dexamethasone-Test, after endocrine diagnostic tests: magnetic resonance imaging (MRI), ultra-sound, computer tomography (CT) and other localization diagnostics. First-line therapy is trans-sphenoidal surgery (TSS) of the pituitary adenoma (in case of ACTH-producing tumors). In patients not amenable for surgery radiotherapy remains an option. Pharmacological therapy applies when these two options are not amenable or refused. In cases when pharmacological therapy becomes necessary, Pasireotide should be used in first-line in CD. CS patients are at an overall 4-fold higher mortality rate than age- and gender-matched subjects in the general population. Differences in stress tolerance and brood size between a non-indigenous and an indigenous gammarid in the northern Baltic Sea
Differences in stress tolerance and reproductive traits may drive the competitive hierarchy between nonindigenous and indigenous species and turn the former ones into successful invaders. In the northern Baltic Sea, the non-indigenous Gammarus tigrinus is a recent invader of littoral ecosystems and now occupies comparable ecological niches as the indigenous G. zaddachi. In laboratory experiments on specimens collected between June and August 2009 around Tva¨rminne in southern Finland (59°500N/23°150E), the tolerances towards heat stress and hypoxia were determined for the two species using lethal time, LT50, as response variable. The brood size of the two species was also studied and some observations were made on maturation of juveniles. Gammarus tigrinus was more resistant to hypoxia and survived at higher temperatures than G. zaddachi. Brood size was also greater in G. tigrinus than in G. zaddachi and G. tigrinus matured at a smaller size and earlier than G. zaddachi. Hence, there are clear competitive advantages for the non-indigenous G. tigrinus compared to the indigenous G. zaddachi, and these may be further strengthened through ongoing environmental changes related to increased eutrophication and a warming climate in the Baltic Sea region
