Cryo-EM structures reveal intricate Fe-S cluster arrangement and charging in Rhodobacter capsulatus formate dehydrogenase

Metal-containing formate dehydrogenases (FDH) catalyse the reversible oxidation of formate to carbon dioxide at their molybdenum or tungsten active site. They display a diverse subunit and cofactor composition, but structural information on these enzymes is limited. Here we report the cryo-electron microscopic structures of the soluble Rhodobacter capsulatus FDH (RcFDH) as isolated and in the presence of reduced nicotinamide adenine dinucleotide (NADH). RcFDH assembles into a 360 kDa dimer of heterotetramers revealing a putative interconnection of electron pathway chains. Flusshochwasser werden in lokale und plotzliche Sturzfluten und in Hochwasser an groseren Flussen unterschieden. Fur verschiedene Hochwasserindikatoren und Flusseinzugsgebiete ergeben sich erhebliche Unterschiede, wobei sowohl uberwiegend aus Regen als auch uberwiegend aus Schmelzwasser gespeiste Hochwasserereignisse betrachtet werden. Besondere Aufmerksamkeit finden Hochwasserereignisse an Rhein, Elbe, Weser und Ems sowie die Entwicklung von Sturzfluten infolge von Extremniederschlagen kurzer Dauer, wobei die Beobachtungen und Trends zu Modellierungsergebnissen in Beziehung gesetzt werden. Auch die Notwendigkeit von Anpassungsmasnahmen aufgrund uberwiegend positiver Trends wird diskutiert

TRY plant trait database – enhanced coverage and open access

Plant traits - the morphological, anatomical, physiological, biochemical and phenological characteristics of plants - determine how plants respond to environmental factors, affect other trophic levels, and influence ecosystem properties and their benefits and detriments to people. Plant trait data thus represent the basis for a vast area of research spanning from evolutionary biology, community and functional ecology, to biodiversity conservation, ecosystem and landscape management, restoration, biogeography and earth system modelling. Since its foundation in 2007, the TRY database of plant traits has grown continuously. It now provides unprecedented data coverage under an open access data policy and is the main plant trait database used by the research community worldwide. Increasingly, the TRY database also supports new frontiers of trait‐based plant research, including the identification of data gaps and the subsequent mobilization or measurement of new data. To support this development, in this article we evaluate the extent of the trait data compiled in TRY and analyse emerging patterns of data coverage and representativeness. Best species coverage is achieved for categorical traits - almost complete coverage for ‘plant growth form’. However, most traits relevant for ecology and vegetation modelling are characterized by continuous intraspecific variation and trait–environmental relationships. These traits have to be measured on individual plants in their respective environment. Despite unprecedented data coverage, we observe a humbling lack of completeness and representativeness of these continuous traits in many aspects. We, therefore, conclude that reducing data gaps and biases in the TRY database remains a key challenge and requires a coordinated approach to data mobilization and trait measurements. This can only be achieved in collaboration with other initiatives

A seamless filter for daily to seasonal forecasts, with applications to Iran and Brazil

A digital filter is introduced which treats the problem of predictability versus time averaging in a continuous, seamless manner. This seamless filter (SF) is characterized by a unique smoothing rule that determines the strength of smoothing in dependence on lead time. The rule needs to be specified beforehand, either by expert knowledge or by user demand. As a result, skill curves are obtained that allow a predictability assessment across a whole range of time-scales, from daily to seasonal, in a uniform manner. The SF is applied to downscaled SEAS5 ensemble forecasts for two focus regions in or near the tropical belt, the river basins of the Karun in Iran and the São Francisco in Brazil. Both are characterized by strong seasonality and semi-aridity, so that predictability across various time-scales is in high demand. Among other things, it is found that from the start of the water year (autumn), areal precipitation is predictable with good skill for the Karun basin two and a half months ahead; for the São Francisco it is only one month, longer-term prediction skill is just above the critical level

Rhine flood stories: Spatio‐temporal analysis of historic and projected flood genesis in the Rhine River basin

AbstractThe genesis of floods in large river basins often is complex. Streamflow originating from precipitation and snowmelt and different tributaries can superimpose and cause high water levels, threatening cities and communities along the riverbanks. For better understanding the mechanisms (origin and composition) of flood events in large and complex basins, we capture and share the story behind major historic and projected streamflow peaks in the Rhine River basin. Our analysis is based on hydrological simulations with the mesoscale Hydrological Model forced with both meteorological observations and an ensemble of climate projections. The spatio‐temporal analysis of the flood events includes the assessment and mapping of antecedent liquid precipitation, snow cover changes, generated and routed runoff, areal extents of events, and the above‐average runoff from major sub‐basins up to 10 days before a streamflow peak. We introduce and assess the analytical setup by presenting the flood genesis of the two well‐known Rhine floods that occurred in January 1995 and May 1999. We share our extensive collection of event‐based Rhine River flood genesis, which can be used in‐ and outside the scientific community to explore the complexity and diversity of historic and projected flood genesis in the Rhine basin. An interactive web‐based viewer provides easy access to all major historic and projected streamflow peaks at four locations along the Rhine. The comparison of peak flow genesis depending on different warming levels elucidates the role of changes in snow cover and precipitation characteristics in the (pre‐)Alps for flood hazards along the entire channel of the Rhine. Furthermore, our results suggest a positive correlation between flood magnitudes and areal extents of an event. Further hydro‐climatological research is required to improve the understanding of the climatic impact on the Rhine and beyond.The genesis of riverine floods in large river basins often is complex.
