Self-sorting of two imine-based metal complexes: Balancing kinetics and thermodynamics in constitutional dynamic networks

A major hurdle in the development of complex constitutional dynamic networks (CDNs) is the lack of strategies to simultaneously control the output of two (or more) interconnected dynamic processes over several species, namely reversible covalent imine bond formation and dynamic metal–ligand coordination. We have studied in detail the self-sorting process of 11 constitutional dynamic libraries containing two different amines, aldehydes and metal salts into two imine-based metal complexes, having no overlap in terms of their compositions. This study allowed us to determine the factors influencing the fidelity of this process (concentration, electronic and steric parameters of the organic components, and nature of the metal cations). In all 11 systems, the outcome of the process was primarily determined by the ability of the octahedral metal ion to select its pair of components from the initial pool of components, with the composition of the weaker tetrahedral complex being imposed by the components rejected by the octahedral metal ions. Different octahedral metal ions required different levels of precision in the “assembling instructions” provided by the organic components of the CDN to guide it towards a sorted output. The concentration of the reaction mixture, and the electronic and steric properties of the initial components of the library were all found to influence the lifetime of unwanted metastable intermediates formed during the assembling of the two complexes

Triple Self-Sorting in Constitutional Dynamic Networks: Parallel Generation of Imine-Based CuI, FeII, and ZnII Complexes

Three imine‐based metal complexes, having no overlap in terms of their compositions, have been simultaneously generated from the self‐sorting of a constitutional dynamic library (CDL) containing three amines, three aldehydes, and three metal salts. The hierarchical ordering of the stability of the three metal complexes assembled and the leveraging of the antagonistic and agonistic relationships existing between the constituents within the constitutional dynamic network corresponding to the CDL were pivotal in achieving the sorting. Examination of the process by NMR spectroscopy showed that the self‐sorting of the FeII and ZnII complexes depended on an interplay between the thermodynamic driving forces and a kinetic trap involved in their assembly. These results also exemplify the concept of “simplexity”—the fact that the output of a self‐assembling system may be simplified by increasing its initial compositional complexity—as the two complexes could self‐sort only in the presence of the third pair of organic components, those of the CuI complex

    Get PDF
COVID-19 is a disease that has severe manifestations in some patients, who need support to improve their ventilatory parameters, currently opting for non-invasive ventilation (NIV) and pronation strategies to improve lung oxygenation. OBJECTIVE. The main objective of this research is to review the literature on the use of non-invasive ventilation plus pronation in patients with COVID-19 related to a decrease in mortality available until July 2020. METHODS. A narrative review study was carried out, performing a search in PUBMED / Medline and SCOPUS, which allowed us to extract the relevant studies that were selected for this review. RESULTS. It was found that 20% -25% of COVID-19 patients, NIV and awake pronation can stabilize their respiratory status and avoid the need for intubation. CONCLUSIONS. We must bear in mind that when using devices associated with NIV, they must be performed in negative pressure rooms or with high-energy particle accumulator filters together with the appropriate protective equipment. The prone position alters the mechanics and physiology of gas exchange to result in better oxygenation.Tesi

Novel Inducers of the Envelope Stress Response BaeSR in Salmonella Typhimurium: BaeR Is Critically Required for Tungstate Waste Disposal

    Get PDF
The RpoE and CpxR regulated envelope stress responses are extremely important for SalmonellaTyphimurium to cause infection in a range of hosts. Until now the role for BaeSR in both the Salmonella Typhimurium response to stress and its contribution to infection have not been fully elucidated. Here we demonstrate stationary phase growth, iron and sodium tungstate as novel inducers of the BaeRregulon, with BaeR critically required for Salmonella resistance to sodium tungstate. We show that functional overlap between the resistance nodulation-cell division (RND) multidrug transporters, MdtA, AcrD and AcrB exists for the waste disposal of tungstate from the cell. We also point to a role for enterobactinsiderophores in the protection of enteric organisms from tungstate, akin to the scenario in nitrogen fixing bacteria. Surprisingly, BaeR is the first envelope stress response pathway investigated in S. Typhimurium that is not required for murine typhoid in either ityS or ityR mouse backgrounds. BaeR is therefore either required for survival in larger mammals such as pigs or calves, an avian host such as chickens, or survival out with the host altogether where Salmonella and related enterics must survive in soil and water

The 1684 bombing of Genoa by the French fleet had huge consequences in Europe and confirmed the hegemony of Louis XIV. Unexpectedly, a work published by the Genoese writer Giovanni Paolo Marana, Dialogue de Genes et d’Algers, Villes Foudroyées par les Armes Invincibles de Louis le Grand, l’année 1684, was taken to be a prosopopoeia of two beseiged and conquered cities. In it, Algiers tries to convince Genoa of the superiority of its new French lord, whose bellicose objectives allow just two reactions: submission or resistance. Genoa’s desperate revolt is depicted not as heroic but rather as an affront to divine will, and the vanquished, destroyed city issues its humiliated apologetic through this articulate exchange. The list of all Genoa’s diplomatic failings lead to the conclusion that punishment was deserved, and the rebellious city is destroyed. Published simultaneously by court order in Italian and French, this fictional work of propaganda was aimed at readers throughout Europe, and its impact was greater than it would have been had it depicted Genoa as the enemy. This “dialogue” is evidence of the king’s desire to assert his legitimacy in the Mediterranean

ZraP is a periplasmic molecular chaperone and a repressor of the zinc-responsive two-component regulator ZraSR

The bacterial envelope is the interface with the surrounding environment and is consequently subjected to a barrage of noxious agents including a range of compounds with antimicrobial activity. The ESR (envelope stress response) pathways of enteric bacteria are critical for maintenance of the envelope against these antimicrobial agents. In the present study, we demonstrate that the periplasmic protein ZraP contributes to envelope homoeostasis and assign both chaperone and regulatory function to ZraP from Salmonella Typhimurium. The ZraP chaperone mechanism is catalytic and independent of ATP; the chaperone activity is dependent on the presence of zinc, which is shown to be responsible for the stabilization of an oligomeric ZraP complex. Furthermore, ZraP can act to repress the two-component regulatory system ZraSR, which itself is responsive to zinc concentrations. Through structural homology, ZraP is a member of the bacterial CpxP family of periplasmic proteins, which also consists of CpxP and Spy. We demonstrate environmental co-expression of the CpxP family and identify an important role for these proteins in Salmonella's defence against the cationic antimicrobial peptide polymyxin B
