    Otimização e validação de metodologia para determinação de Ivermectina em amostras de leite

    Dissertação (mestrado)—Universidade de Brasília, Instituto de Química, Programa de Pós-Graduação em Tecnologias Química e Biológica, 2014.O fármaco veterinário endectocida conhecido como ivermectina ou 22,23-dihidroavermectina B1a + 22,23-dihidroavermectina B1b, apesar da sua recente proibição, continua a ser usado largamente no Brasil tanto em bovinos como em suíno e bubalino, porém a grande maioria se deve aos bovinos, devido ao fato do Brasil possuir o segundo maior rebanho mundial. A problemática da utilização deste fármaco está associada a uma possível excreção da ivermectina no leite desses animais, o qual é amplamente utilizado na dieta diária dos brasileiros, e os efeitos devido à exposição crônica a esse fármaco ainda não são bem estudados. Este trabalho adaptou uma metodologia para determinação da ivermectina em leite de bovinos. Foi realizada a validação da metodologia com leitura de amostra de leite fortificada. A amostra de leite integral foi coletada direto do animal para que permanecesse a quantidade total de gordura, devido ao caráter lipofílico do fármaco. O método validado baseou-se em cromatografia líquida com detecção UV/VIS. A extração foi realizada em duas partes, a primeira líquido-líquido, onde há a separação das gorduras e proteínas, e a segunda em fase sólida com o auxílio de cartuchos SPE C18. Para a leitura das amostras utilizou-se cromatógrafo líquido, com detector de UV-Vis, com coluna Phenomenex C18, 10cm, ID 46µm. Os resultados de validação mostraram que o método é linear, com coeficiente de determinação (R2) igual à 0,991, seletivo, preciso, com coeficientes de variação entre 3,98 e 9,42%, exato, com valores de recuperação variando entre 98 e 110% e com limites de detecção e quantificação de 19,825 µg/L e 49,350 µg/L respectivamente. _______________________________________________________________________________________________ ABSTRACTThe veterinary drug called ivermectin or endectocide 22.23-22.23-dihydroavermectin B1a + B1b dihydroavermectin, despite its recent ban, continues to be used widely in Brazil both in cattle and in pigs and buffalo, but the vast majority is due to cattle, due to the fact that Brazil has the second largest herd. The issue of use of this drug is associated with a possible excretion of ivermectin in milk of these animals, which is widely used in daily diet of Brazilians, and the effects due to chronic exposure to this drug are not well studied. This study adapted a method for determining the milk of bovine ivermectin. Validate the methodology with sample reading of fortified milk was performed. The sample of whole milk was collected directly from the animal to remain on the total amount of fat, due to the lipophilic nature of the drug. The validated method was based on liquid chromatography with UV detection / VIS. The extraction was performed in two parts, the first liquid-liquid, where there is a separation of fat and protein, and the second solid phase with the aid of C18 SPE cartridges. To read the samples we used liquid chromatography with UV-Vis detector with Phenomenex C18, 10cm, 46μm ID column. The validation results showed that the method is linear, with a coefficient of determination (R2) equal to 0.991, selective, precise, with coefficients of variation between 3.98 and 9.42%, exact, with recovery values ranging between 98 and and 110% limits of detection and quantification of 19,825 mg / L and 49,350 mg / l respectively

    Vascular fibrosis in aging and hypertension: molecular mechanisms and clinical implications

    Aging is the primary risk factor underlying hypertension and incident cardiovascular disease. With aging, the vasculature undergoes structural and functional changes characterized by endothelial dysfunction, wall thickening, reduced distensibility, and arterial stiffening. Vascular stiffness results from fibrosis and extracellular matrix (ECM) remodelling, processes that are associated with aging and are amplified by hypertension. Some recently characterized molecular mechanisms underlying these processes include increased expression and activation of matrix metalloproteinases, activation of transforming growth factor-β1/SMAD signalling, upregulation of galectin-3, and activation of proinflammatory and profibrotic signalling pathways. These events can be induced by vasoactive agents, such as angiotensin II, endothelin-1, and aldosterone, which are increased in the vasculature during aging and hypertension. Complex interplay between the “aging process” and prohypertensive factors results in accelerated vascular remodelling and fibrosis and increased arterial stiffness, which is typically observed in hypertension. Because the vascular phenotype in a young hypertensive individual resembles that of an elderly otherwise healthy individual, the notion of “early” or “premature” vascular aging is now often used to describe hypertension-associated vascular disease. We review the vascular phenotype in aging and hypertension, focusing on arterial stiffness and vascular remodelling. We also highlight the clinical implications of these processes and discuss some novel molecular mechanisms of fibrosis and ECM reorganization

    Razão de uma escolha: a importância do periódico O Portuguez e do seu redactor João Bernardo da Rocha Loureiro no desencadear da Revolução Liberal portuguesa de 1820, que deu início ao processo que colocou fim à monarquia absoluta e desencadeou a desconstrução do Antigo Regime em Portugal. Nesta panorâmica, e tendo como horizonte de chegada a fruição da liberdade, pode dizer-se que, enquanto sistema de significações colectivas, o imaginário do redactor Rocha Loureiro parece actuar como reacção ao poder dissolvedor da inteligência, como regulador que satisfaz a busca apaixonada de um grupo ou sociedade que persegue a sua identidade da qual a liberdade é elemento constituinte, fundamental e fundamentador. A liberdade define-se pela ausência de limitação às garantias necessárias para o harmonioso desenvolvimento do indivíduo. Antes de mais a liberdade é, na sua essência, uma ausência de limitação, sendo simultaneamente uma determinação positiva e espontânea da vontade de liberdade em que cada um procura atingir o fim racional de que está possuído. O indivíduo deseja a liberdade com vista a alcançar o melhor possível do seu ser. In this overview, and having as horizon the arrival, and the enjoyment of freedom, it can be said that, while the collective meanings system, and the imaginary of the redactor Rocha Loureiro seems to act, as a reaction to the melted power of his intelligence, and as a regulator that satisfies the passionate pursuit, of a group or society, that pursues its own identity, from which freedom is an essential element component. Liberty is defined by the lack of the restraining guarantees that are necessary for the harmonium’s development of the human being. First of all, freedom is, in its essence, a lack of limitation, which could be at the same time a positive and a spontaneous determination, of freedom desire, in which, each one seeks to achieve the rational purpose , that owns. The human being, wants the freedom, in order to achieve the best possible of his own being.     Planning the territory of São Paulo state, Brazil, in the Democratic Period: Carvalho Pinto’s Action Plan (1959 – 1963)

    The postwar is marked, in Brazil, by initiatives of state planning and modern architecture that reach their apex in President Juscelino Kubitschek's Targets Plan (1956-1961) and its “synthesis-target”, Brasilia. Contemporary of these achievements is the Action Plan promoted by the governor of São Paulo state, Carvalho Pinto (1959-1963). Sharing the developmentist character that marks Brazilian state planning experience at the time, the Action Plan contains extensive physical targets aiming at the integration of the territory in the production process; relying on the broad participation of planners connected to the Économie et Humanisme Movement of the Dominican Father Louis-Joseph Lebret, the plan incorporates cojunctural analyses, methodologies and public policies that combine such investments in infrastructure and agricultural and industrial expansion with investments envisaging the "improvement of man's conditions". This paper focuses on this Paulista episode of state planning, considering the social scene of the period, providing an overview of its contents and evaluating its results. It makes notes on the economic and technological changes that occurred in the postwar period, and on the process of Brazilian industrialization in the 1950s and 1960s; it attends the conjunctural analyses of the Brazilian and Paulista economy made by the plan's authors, which reveal their perception of the economic and social process and inform their planning activity. It describes the planning process since its organization and constitution of its technical staff, to the strategies used in its elaboration. It provides a framework of the Action Plan that considers integrally: budget management techniques, funding sources, physical targets, public policies. It points out the political content of the plan, exploring its possibilities and limits, notably those related to the urban question and the housing question. It covers the political conditions that made planning almost an unanimity in those years, and the reasons that lead to non-institutionalization of the practice and to the discontinuation of the initiative, after the defeat in Carvalho Pinto's successor race and the subsequent outbreak of the civil-military coup (1964-1985). Thus, being considered in its broader dimensions, Carvalho Pinto’s Action Plan reveals the deep marks left by it in the São Paulo state territory

    Improving Random Access with NOMA in mMTC XL-MIMO

    The extra-large multiple-input multiple-output (XL-MIMO) architecture has been recognized as a technology for supporting the massive MTC (mMTC), providing very high-data rates in high-user density scenarios. However, the large dimension of the array increases the Rayleigh distance (dRayl), in addition to obstacles and scatters causing spatial non-stationarities and distinct visibility regions (VRs) across the XL array extension. We investigate the random access (RA) problem in crowded XL-MIMO scenarios; the proposed grant-based random access (GB-RA) protocol combining the advantage of non-orthogonal multiple access (NOMA) and strongest user collision resolutions in extra-large arrays (SUCRe-XL) named NOMA-XL can allow access of two or three colliding users in the same XL sub-array (SA) selecting the same pilot sequence. The received signal processing in a SA basis changes the dRayl, enabling the far-field planar wavefront propagation condition, while improving the system performance. The proposed NOMA-XL GB-RA protocol can reduce the number of attempts to access the mMTC network while improving the average sum rate, as the number of SA increases.Comment: 13 pages, 5 figures, 1 table, conference VTC 2023. arXiv admin note: substantial text overlap with arXiv:2303.0053

    Genomic and non-genomic effects of androgens in the cardiovascular system: clinical implications

    The principle steroidal androgens are testosterone and its metabolite 5α-dihydrotestosterone (DHT), which is converted from testosterone by the enzyme 5α-reductase. Through the classic pathway with androgens crossing the plasma membrane and binding to the androgen receptor (AR) or via mechanisms independent of the ligand-dependent transactivation function of nuclear receptors, testosterone induces genomic and non-genomic effects respectively. AR is widely distributed in several tissues, including vascular endothelial and smooth muscle cells. Androgens are essential for many developmental and physiological processes, especially in male reproductive tissues. It is now clear that androgens have multiple actions besides sex differentiation and sexual maturation and that many physiological systems are influenced by androgens, including regulation of cardiovascular function [nitric oxide (NO) release, Ca2+ mobilization, vascular apoptosis, hypertrophy, calcification, senescence and reactive oxygen species (ROS) generation].
