    Autologous blood donation in elective surgery of colorectal cancer - is it possible?

    Reports from recent studies indicate a causal relationship between all transfusion, infectious complications and recurrent disease in surgical patients. The aim of the presented study is to elucidate the possibilities and indications for autologous haemotransfusion in the elective surgery of the colorectal cancer. Retrospective analysis was per formed over 724 electively operated patients with colorectal cancer. The prospective part includes observation of 20 radically operated patients. In these patients autologous haemotransfusion was per - formed. As indications for autologous haemotransfusion we consider age up to 80 years, hemoglobin levels at least 130 g/l, plasma protein > 65 g/l, body weight > 50 kg, good general condition and absence of decompensated cardiovascular or endocrine dis eases. In two patients with hemoglobin level between 90 - 110 g/l we per formed stimulation by means of Erythropoietin. According to retrospective data ad mission hemoglobin levels varies from 54 g/l to 175 g/l (mean 119 g/l ± 22, 6). From all electively operated patients 33, 9% had hemoglobin values over 130 g/l. The frequency of post-operative infectious complications increases from 9.1% in non-transfused patients to 38,7 % in patients with more than 4 units of allogenic blood trans fused. The prospective part of the study is designed to establish feasibility of the autologous haemotransfusion in clinical practice. The fall of hemoglobin values on the next day after donation varies from 11g/l to 19g/l (mean 15.5 g/l ± 2,19). Erythropoietin stimulation was not found to increase hemoglobin values significantly. There were no complications in the post-operative period in autologous haemotransfusion group with exception of one superficial wound infection. Autologous haemotransfusion is feasible in approximately 1/3 of electively operated patients with colorectal cancer with no ad verse effects or post-operative complications. It de creases the necessity of allogenic bioproducts and hence the risks related. Autologous haemotransfusion is easy to perform and propose ben e fits both from medical and logistic nature. Concerning to stimulation with Erythropoetin in our opinion it is without proven short term efficacy in general surgery so far

    Directed current due to broken time-space symmetry

    We consider the classical dynamics of a particle in a one-dimensional space-periodic potential U(X) = U(X+2\pi) under the influence of a time-periodic space-homogeneous external field E(t)=E(t+T). If E(t) is neither symmetric function of t nor antisymmetric under time shifts E(t±T/2)E(t)E(t \pm T/2) \neq -E(t), an ensemble of trajectories with zero current at t=0 yields a nonzero finite current as tt\to \infty. We explain this effect using symmetry considerations and perturbation theory. Finally we add dissipation (friction) and demonstrate that the resulting set of attractors keeps the broken symmetry property in the basins of attraction and leads to directed currents as well.Comment: 2 figure

    Pseudomonas germanica sp. nov., isolated from Iris germanica rhizomes

    Through bacterial plant-endophyte extraction from rhizomes of Iris germanica plant, a Gram-stain-negative, aerobic, catalase- and oxidase-positive gammaproteobacterial strain, referred to as FIT28T, was isolated. FIT28T shows vigorous growth on nutrient rich media within the temperature range of 4-35 °C, with optimal growth at 28 °C, a wide pH tolerance from pH 5 to 11, and salt tolerance up to 6 % (w/v) NaCl. Colonies are white-yellow and quickly become mucoid. The results of analysis of the 16S rRNA gene sequence placed the strain within the genus Pseudomonas, and multilocus sequence analysis (MLSA) using 16S rRNA, rpoB, gyrB and rpoD concatenated sequences revealed that the closest relatives of FIT28T are Pseudomonas zeae OE48.2T, 'Pseudomonas crudilactis' UCMA 17988, Pseudomonas tensinigenes ZA5.3T, Pseudomonas helmanticensis OHA11T, Pseudomonas baetica a390T, Pseudomonas iridis P42T, Pseudomonas atagonensis PS14T and Pseudomonas koreensis Ps 9-14T, within the Pseudomonas koreensis subgroup of the Pseudomonas fluorescens lineage. The genome size of FIT28T is about 6.7 Mb with 59.09 mol% DNA G+C content. Average nucleotide identity (ANI) and digital DNA-DNA hybridization (dDDH) values calculated from the genomic sequences of FIT28T, and the closely related P. zeae OE48.2T are 95.23 and 63.4 %, respectively. Biochemical, metabolic and chemotaxonomic studies further support our proposal that Pseudomonas germanica sp. nov., should be considered a novel species of the genus Pseudomonas. Hence, the type strain FIT28T (=LMG 32353T=DSM 112698T) has been deposited in public cell-type culture centres

    Pseudomonas fitomaticsae sp. nov., isolated at Marimurtra Botanical Garden in Blanes, Catalonia, Spain

    In the framework of the research project called fitomatics, we have isolated and characterized a bacterial plant-endophyte from the rhizomes of Iris germanica, hereafter referred to as strain FIT81T. The bacterium is Gram negative, rod-shaped with lophotrichous flagella, and catalase- and oxidase-positive. The optimal growth temperature of strain FIT81T is 28 °C, although it can grow within a temperature range of 4-32 °C. The pH growth tolerance ranges between pH 5 and 10, and it tolerates 4% (w/v) NaCl. A 16S rRNA phylogenetic analysis positioned strain FIT81T within the genus Pseudomonas, and multilocus sequence analysis revealed that Pseudomonas gozinkensis IzPS32dT, Pseudomonas glycinae MS586T, Pseudomonas allokribbensis IzPS23T, 'Pseudomonas kribbensis' 46-2 and Pseudomonas koreensis PS9-14T are the top five most closely related species, which were selected for further genome-to-genome comparisons, as well as for physiological and chemotaxonomic characterization. The genome size of strain FIT81T is 6 492 796 base-pairs long, with 60.6 mol% of G+C content. Average nucleotide identity and digital DNA-DNA hybridization analyses yielded values of 93.6 and 56.1%, respectively, when the FIT81T genome was compared to that of the closest type strain P. gozinkensis IzPS32dT. Taken together, the obtained genomic, physiologic and chemotaxonomic data indicate that strain FIT81T is different from its closest relative species, which lead us to suggest that it is a novel species to be included in the list of type strains with the name Pseudomonas fitomaticsae sp. nov.     Применение обогащенной тромбоцитами плазмы с целью профилактики поздних осложнений и болевого синдрома после герниопластики

    Цель. За счет применения обогащенной тромбоцитами плазмы (ОTП) снизить степень выраженности поздних осложнений у больных, оперированных по поводу инцизионных вентральных грыж. Материалы и методы. В клиническое исследование, проведенное в условиях хирургического отделения Центра реконструктивной и восстановительной медицины (Университетская клиника) Одесского национального медицинского университета с 2013 по 2019 г., включены 84 больных, оперированных по поводу послеоперационных вентральных грыж. Средний возраст больных составил (60,1 ± 7,6) года, размер грыжевых дефектов - 8,2 см, средний индекс массы тела – 31,8 кг/м2. Больным по показаниям выполнен стандартный объем оперативного вмешательства: грыжесечение, герниопластика передней брюшной стенки облегченным (80 г/м2) полипропиленовым имплантатом. Пациентам основной группы после выполнения пластики зону имплантации протеза инфильтрировали активированной аутологичной ОТП, которая была заготовлена путем двойного центрифугирования, в объеме 14 – 38 мл. Пациентам контрольной группы инфильтрацию ОТП не выполняли. Качество жизни пациентов оценивали с помощью стандартизированного опросника SF-36 до операции, а также в послеоперационном периоде. С целью дифференцирования нейропатической боли применяли опросники DN4 и ID Pain. Результаты. У пациентов обеих групп в послеоперационном периоде отмечали повышение показателей качества жизни. Статистическая достоверность различий показателей была выше у пациентов с надапоневротическим расположением протеза. Показатели физического здоровья и ассоциированные с ними показатели социальной активности, общего состояния здоровья демонстрировали значимо более высокий уровень качества жизни у пациентов основной группы. Показатель общего состояния здоровья у больных после надапоневротической герниопластики с применением ОТП был достоверно выше, чем в контрольной группе – соответственно 69,3 ± 3,8 и 59,7 ± 4,1 (p < 0,05). Выводы. Техника инфильтрации мягких тканей, контактирующих с полипропиленовым протезом, безопасна, достоверно не увеличивает риск возникновения локальных или системных осложнений. Предложенная методика за счет модификации репаративных процессов в зоне герниопластики способствует интеграции протеза, снижает риск развития и степень выраженности биомеханических нарушений, хронического болевого синдрома, что положительно сказывается на качестве жизни пациентов. Эффективность техники статистически достоверно выше у пациентов после надапоневротической герниопластики

    Tomato STEROL GLYCOSYLTRANSFERASE 1 silencing unveils a major role of steryl glycosides in plant and fruit development

    Free and glycosylated sterols localize in the plant cell plasma membrane, where in combination with other lipids regulate its structure and function. The role of glycosylated sterols in regulating membrane-associated biological processes is more relevant in plants like tomato (Solanum lycopersicum), in which glycosylated sterols are the predominant sterols. A proper ratio of free sterols versus glycosylated sterols has proven to be essential for proper plant performance in several species, but almost nothing is known in tomato. To assess the role of glycosylated sterols in tomato plant and fruit development, we generated transgenic lines of tomato cultivar Micro-Tom expressing two different amiRNAs devised to silence STEROL GLYCOSYLTRANSFERASE 1, the most actively expressed of the four genes encoding sterol glycosyltransferases in this plant. STEROL GLYCOSYLTRANSFERASE 1 gene silencing caused moderate plant dwarfism and reduced fruit size. Analysis of the profile of glycosylated sterols throughout fruit development demonstrated that the maintenance of proper levels of these compounds during the early stages of fruit development is essential for normal fruit growth, since reduced levels of glycosylated sterols trigger a transcriptional downregulatory response that affects genes involved in processes that are critical for proper fruit development, such as seed filling, cell wall extension and auxin signaling

    Magnetoelectric ordering of BiFeO3 from the perspective of crystal chemistry

    In this paper we examine the role of crystal chemistry factors in creating conditions for formation of magnetoelectric ordering in BiFeO3. It is generally accepted that the main reason of the ferroelectric distortion in BiFeO3 is concerned with a stereochemical activity of the Bi lone pair. However, the lone pair is stereochemically active in the paraelectric orthorhombic beta-phase as well. We demonstrate that a crucial role in emerging of phase transitions of the metal-insulator, paraelectric-ferroelectric and magnetic disorder-order types belongs to the change of the degree of the lone pair stereochemical activity - its consecutive increase with the temperature decrease. Using the structural data, we calculated the sign and strength of magnetic couplings in BiFeO3 in the range from 945 C down to 25 C and found the couplings, which undergo the antiferromagnetic-ferromagnetic transition with the temperature decrease and give rise to the antiferromagnetic ordering and its delay in regard to temperature, as compared to the ferroelectric ordering. We discuss the reasons of emerging of the spatially modulated spin structure and its suppression by doping with La3+.Comment: 18 pages, 5 figures, 3 table
