    Oceanic Turbulence and Stochastic Models from Subsurface Lagrangian Data for the Northwest Atlantic Ocean

    The historical dataset provided by 700-m acoustically tracked floats is analyzed in different regions of the northwestern Atlantic Ocean. The goal is to characterize the main properties of the mesoscale turbulence and to explore Lagrangian stochastic models capable of describing them. The data analysis is carried out mostly in terms of Lagrangian velocity autocovariance and cross-covariance functions. In the Gulf Stream recirculation and extension regions, the autocovariances and cross covariances exhibit significant oscillatory patterns on time scales comparable to the Lagrangian decorrelation time scale. They are indicative of sub- and superdiffusive behaviors in the mean spreading of water particles. The main result of the paper is that the properties of Lagrangian data can be considered as a superposition of two different regimes associated with looping and nonlooping trajectories and that both regimes can be parameterized using a simple first-order Lagrangian stochastic model with spin parameter V. The spin couples the zonal and meridional velocity components, reproducing the effects of rotating coherent structures such as vortices and mesoscale eddies. It is considered as a random parameter whose probability distribution is approximately bimodal, reflecting the distribution of loopers (finite V) and nonloopers (zero V). This simple model is found to be very effective in reproducing the statistical properties of the data. 1

    More than teacher directed or child initiated: Preschool curriculum type, parent involvement, and children's outcomes in the child-parent centers.

    This study investigated the contributions of curriculum approach and parent involvement to the short- and long-term effects of preschool participation in the Title I Chicago Child-Parent Centers. Data came from the complete cohort of 989 low-income children (93% African American) in the Chicago Longitudinal Study, who attended preschool in the 20 Child-Parent Centers in 1983-1985 and kindergarten in 1985-1986. We found that implementation of an instructional approach rated high by Head Teachers in teacher-directed and child-initiated activities was most consistently associated with children’s outcomes, including school readiness at kindergarten entry, reading achievement in third and eighth grades, and avoidance of grade retention. Parent involvement in school activities, as rated by teachers and by parents, was independently associated with child outcomes from school readiness at kindergarten entry to eighth grade reading achievement and grade retention above and beyond the influence of curriculum approach. Findings indicate that instructional approaches that blend a teacher-directed focus with child-initiated activities and parental school involvement are origins of the long-term effects of participation in the Child-Parent Centers

    Family profiles and educational attainment

    The educational attainment of participants in the Chicago Longitudinal Study (93% Black and 7% Hispanic) was examined. Cluster analysis on measures of human capital resources, family dynamics and demographics was used to identify four distinct profiles of families. In general, children who had family profiles characterized by higher levels of human capital resources and more favorable scores on indictors of family functioning were more likely than other children to have higher educational attainment. Moreover, children who experienced a positive change in family profile characteristics between the ages of 8 and 12 were more likely than other children to have higher educational attainment. Implications for intervention and research are discussed

    Stress and Strain in Flat Piling of Disks

    We have created a flat piling of disks in a numerical experiment using the Distinct Element Method (DEM) by depositing them under gravity. In the resulting pile, we then measured increments in stress and strain that were associated with a small decrease in gravity. We first describe the stress in terms of the strain using isotropic elasticity theory. Then, from a micro-mechanical view point, we calculate the relation between the stress and strain using the mean strain assumption. We compare the predicted values of Young's modulus and Poisson's ratio with those that were measured in the numerical experiment.Comment: 9 pages, 1 table, 8 figures, and 2 pages for captions of figure

    Macrophage proliferation distinguishes 2 subgroups of knee osteoarthritis patients

    Osteoarthritis (OA) is a leading cause of disability, globally. Despite an emerging role for synovial inflammation in OA pathogenesis, attempts to target inflammation therapeutically have had limited success. A better understanding of the cellular and molecular processes occurring in the OA synovium is needed to develop novel therapeutics. We investigated macrophage phenotype and gene expression in synovial tissue of OA and inflammatory-arthritis (IA) patients. Compared with IA, OA synovial tissue contained higher but variable proportions of macrophages (P < 0.001). These macrophages exhibited an activated phenotype, expressing folate receptor-2 and CD86, and displayed high phagocytic capacity. RNA sequencing of synovial macrophages revealed 2 OA subgroups. Inflammatory-like OA (iOA) macrophages are closely aligned to IA macrophages and are characterized by a cell proliferation signature. In contrast, classical OA (cOA) macrophages display cartilage remodeling features. Supporting these findings, when compared with cOA, iOA synovial tissue contained higher proportions of macrophages (P < 0.01), expressing higher levels of the proliferation marker Ki67 (P < 0.01). These data provide new insight into the heterogeneity of OA synovial tissue and suggest distinct roles of macrophages in pathogenesis. Our findings could lead to the stratification of OA patients for suitable disease-modifying treatments and the identification of novel therapeutic targets

    The formation of broad emission line regions in supernova-QSO wind interactions: II. 2D calculations

    One aspect of supernova remnant evolution that is relatively unstudied is the influence of an AGN environment. A high density ambient medium and a nearby powerful continuum source will assist the cooling of shocked ejecta and swept-up gas. Motion of the surrounding medium relative to the remnant will also affect the remnant morphology. In an extension to previous work we have performed 2D hydrodynamical calculations of SNR evolution in an AGN environment, and have determined the evolutionary behaviour of cold gas in the remnant. The cold gas will contribute to the observed broad line emission in AGNs, and we present preliminary theoretical line profiles from our calculations. A more detailed comparison with observations will be performed in future work. The SNR-AGN interaction may be also useful as a diagnostic of AGN winds

    Understanding VPAC receptor family peptide binding and selectivity

    The vasoactive intestinal peptide (VIP) and pituitary adenylate cyclase-activating polypeptide (PACAP) receptors are key regulators of neurological processes. Despite recent structural data, a comprehensive understanding of peptide binding and selectivity among different subfamily receptors is lacking. Here, we determine structures of active, Gs-coupled, VIP-VPAC1R, PACAP27-VPAC1R, and PACAP27-PAC1R complexes. Cryo-EM structural analyses and molecular dynamics simulations (MDSs) reveal fewer stable interactions between VPAC1R and VIP than for PACAP27, more extensive dynamics of VIP interaction with extracellular loop 3, and receptor-dependent differences in interactions of conserved N-terminal peptide residues with the receptor core. MD of VIP modelled into PAC1R predicts more transient VIP-PAC1R interactions in the receptor core, compared to VIP-VPAC1R, which may underlie the selectivity of VIP for VPAC1R over PAC1R. Collectively, our work improves molecular understanding of peptide engagement with the PAC1R and VPAC1R that may benefit the development of novel selective agonists
