    Since in the 80's as the technologic boom took place, its advances have been applied in many fields, including communications. More specifically, the telephone network, typically analog, has migrated in recent years to digital. In this context, it is noted that communication needs have changed. Until a few years ago the vast majority of traffic was voice traffic. However, today that percentage is minimal and there is a growing demand of data traffic thanks to the great evolution of the Internet. Here data becomes a priority, as well as its technology, its protocols, and people start thinking about how to manage traffic and merge both in one. From this comes the idea of transmitting voice over Internet Protocol (VoIP). At the end of the 90's technology had advanced enough to create solutions that enabled a computer to work as a telephone exchange (PBX) and a few years later extensive services to VoIP users began to be offered. Therefore, we try to ease its use by creating user-friendly graphical interfaces, which do not require extensive knowledge in how to configure telephony systems. Askozia PBX is a project that aims to bring VoIP technology to businesses and average users. And this is where my project fits in. Within this GUI, there will be the possibility to display the system configuration file and make changes that are desired. In other words, the project is based on the development and integration of an embedded editor in the graphical user interface for managing a PBX, that aims to make live changes in settings possible. I will explain in detail the process of integration and reduction of code required to turn my project into a package available for Askozia. __________________________________________________________________________________________________________Desde que en en los 80 tuviera lugar el boom tecnológico, sus avances han sido aplicados en muchos campos, incluidos las comunicaciones. Mas específicamente, la red de telefonía, que hasta hace poco era analógica, ha evolucionado recientemente a digital. De este contexto se desprende que las necesidades comunicativas han cambiado. Hasta hace unos años, la mayoría del tráfico era tráfico de voz; sin embargo hoy dicho porcentaje es mínimo, habiendo una creciente demanda de tráfico de datos, gracias a la gran evolución de Internet. Aquí los datos se convierten en una prioridad, así como su tecnología y sus protocolos. Por esto se empezó a pensar como tratar ambos tráficos y unirlos en uno solo. De aquí viene la idea de trasmitir voz sobre tecnología IP. (VoIP) A finales de los 90 la tecnología había avanzado lo suficiente como para crear soluciones que permitieran a un ordenador comportarse como una centralita telefónica (PBX), y pocos años después, empezaron a ser ofrecidos servicios extensivos a los usuarios de VoIP. Por lo tanto, intentamos facilitar el uso de dichas PBX virtuales, creando interfaces gráficas amigables, que no requieran un gran conocimiento sobre como configurar un sistema telefónico. AskoziaPBX es un proyecto cuyos objetivos son acercar la tecnología VoIP a usuarios medios. Aquí es donde mi proyecto encaja. Con este interfaz gráfico, se dará a posibilidad de mostrar el archivo de configuración del sistema, y poder llevar a cabo los cambios deseados. En otras palabras, el proyecto esta basado en el desarrollo e integración de un editor empotrado en el interfaz gráfico, para gestionar la configuración de la PBX, de forma que se puedan realizar cambios de forma dinámica.     Clozapine and paliperidone palmitate antipsychotic combination in treatment-resistant schizophrenia and other psychotic disorders: A retrospective 6-month mirror-image study

    Background: Around 30% of patients with schizophrenia are considered treatment resistant (TRS). Only around 40% of TRS patients respond to clozapine. Long acting injectable antipsychotics could be a useful augmentation strategy for nonresponders. Methods: We conducted a multicenter, observational, naturalistic, retrospective, 6-month mirror-image study to evaluate the efficacy and tolerability of clozapine and paliperidone palmitate association in 50 patients with TRS and other psychotic disorders. Clinical outcomes and side effects were systematically assessed. Results: Six months after starting the combined treatment, participants showed a significant relief of symptoms, decreasing the Brief Psychiatric Rating Scale total score from 18.32 ± 7.71 to 7.84 ± 5.16 (p < 0.001). The number of hospitalizations, the length of hospital stays and the number of visits to emergency services also decreased, while an increase of the functionality was observed (Personal and Social Performance total score increased from 46.06 ± 118.7 to 60.86 ± 18.68, p < 0.001). There was also a significant decrease in the number and severity of side effects with the combination therapy, decreasing the Udvalg for Kliniske Undersogelser total score from 10.76 ± 8.04 to 8.82 ± 6.63 (p = 0.004). Conclusions: This study provides the first evidence that combining clozapine with paliperidone palmitate in patients with TRS and other psychotic disorders could be effective and safe, suggesting further research with randomized controlled trials of augmentation strategies for clozapine nonresponder patients. Policy Significance Statement: Patients with psychotic disorders such as schizophrenia show a variable response to antipsychotic treatments. Around 30% of patients are considered treatment resistant, indicated by insufficient symptom control to at least two different drugs. In these resistant cases, clozapine should be indicated, as it has shown to be superior to other options. However, only 40% of patients respond to clozapine, being necessary to establish which treatments could best potentiate clozapine action. Combining clozapine with long acting injectable antipsychotics, and particularly paliperidone palmitate, could be a useful strategy. We conducted a multicenter study of 50 patients with treatment-resistant schizophrenia and other psychotic disorders comparing the efficacy and tolerability in the 6 month-period prior and after starting the clozapine and paliperidone palmitate association. Our study suggests that this combination could be effective and safer, laying the groundwork for future clinical trials with this combination

    An AGAMOUS-related MADS-box gene, XAL1 (AGL12), regulates root meristem cell proliferation and flowering transition in Arabidopsis

This article shows that the sister gene of the AGAMOUS (AG) clade, AGL12, has an important role in root development as well as in flowering transition. We isolated three mutant alleles for AGL12, which is renamed here as XAANTAL1 (XAL1): Two alleles, xal1-1 and xal1-2, are in Columbia ecotype and xal1-3 is in Landsberg erecta ecotype. All alleles have a short-root phenotype with a smaller meristem, lower rate of cell production, and abnormal root apical meristem organization. Interestingly, we also encountered a significantly longer cell cycle in the strongest xal1 alleles with respect to wild-type plants. Expression analyses confirmed the presence of XAL1 transcripts in roots, particularly in the phloem. Moreover, XAL1beta-glucuronidase expression was specifically up-regulated by auxins in this tissue. In addition, mRNA in situ hybridization showed that XAL1 transcripts were also found in leaves and floral meristems of wild-type plants. This expression correlates with the late-flowering phenotypes of the xal1 mutants grown under long days. Transcript expression analysis suggests that XAL1 is an upstream regulator of SOC, FLOWERING LOCUS T, and LFY. We propose that XAL1 may have similar roles in both root and aerial meristems that could explain the xal1 late-flowering phenotype.This work was supported by Consejo Nacional de Ciencia y Tecnología (CONACYT), México (grant nos.     The MADS-box XAANTAL1 increases proliferation at the Arabidopsis root stem-cell niche and participates in transition to differentiation by regulating cell-cycle components

    Background Morphogenesis depends on the concerted modulation of cell proliferation and differentiation. Such modulation is dynamically adjusted in response to various external and internal signals via complex transcriptional regulatory networks that mediate between such signals and regulation of cell-cycle and cellular responses (proliferation, growth, differentiation). In plants, which are sessile, the proliferation/differentiation balance is plastically adjusted during their life cycle and transcriptional networks are important in this process. MADS-box genes are key developmental regulators in eukaryotes, but their role in cell proliferation and differentiation modulation in plants remains poorly studied. Methods We characterize the XAL1 loss-of-function xal1-2 allele and overexpression lines using quantitative cellular and cytometry analyses to explore its role in cell cycle, proliferation, stem-cell patterning and transition to differentiation. We used quantitative PCR and cellular markers to explore if XAL1 regulates cell-cycle components and PLETHORA1 (PLT1) gene expression, as well as confocal microscopy to analyse stem-cell niche organization. Key Results We previously showed that XAANTAL1 (XAL1/AGL12) is necessary for Arabidopsis root development as a promoter of cell proliferation in the root apical meristem. Here, we demonstrate that XAL1 positively regulates the expression of PLT1 and important components of the cell cycle: CYCD3;1, CYCA2;3, CYCB1;1, CDKB1;1 and CDT1a. In addition, we show that xal1-2 mutant plants have a premature transition to differentiation with root hairs appearing closer to the root tip, while endoreplication in these plants is partially compromised. Coincidently, the final size of cortex cells in the mutant is shorter than wild-type cells. Finally, XAL1 overexpression-lines corroborate that this transcription factor is able to promote cell proliferation at the stem-cell niche. ConclusionXAL1 seems to be an important component of the networks that modulate cell proliferation/differentiation transition and stem-cell proliferation during Arabidopsis root development; it also regulates several cell-cycle components.This work constitutes a partial fulfilment of the Graduate Program ‘Doctorado en Ciencias Biomédicas’ of the Universidad Nacional Autónoma de México in which Karla V. Contextos, experiencias y voces es un libro colectivo que corona once trabajos de investigación realizados por diecisiete autores con diferentes adscripciones institucionales que se articulan a través de grupos de trabajo latinoamericanos en torno al debate actual sobre la Interculturalidad y Educación. Y con ello obligan al lector a pensar simultáneamente en un conjunto de movimientos o desplazamientos teórico-conceptuales, en el despliegue de posicionamientos éticos, filosóficos, políticos y jurídicos, y en el desarrollo de metodologías de investigación novedosas, que nos posibiliten revelar y actuar sobre una diversidad de experiencias, modelos y programas que buscan potenciar los procesos interculturales en el Sur.     The MADS transcription factor XAL2/AGL14 modulates auxin transport during Arabidopsis root development by regulating PIN expression

    Elucidating molecular links between cell-fate regulatory networks and dynamic patterning modules is a key for understanding development. Auxin is important for plant patterning, particularly in roots, where it establishes positional information for cell-fate decisions. PIN genes encode plasma membrane proteins that serve as auxin efflux transporters; mutations in members of this gene family exhibit smaller roots with altered root meristems and stem-cell patterning. Direct regulators of PIN transcription have remained elusive. Here, we establish that a MADS-box gene (XAANTAL2, XAL2/AGL14) controls auxin transport via PIN transcriptional regulation during Arabidopsis root development; mutations in this gene exhibit altered stem-cell patterning, root meristem size, and root growth. XAL2 is necessary for normal shootward and rootward auxin transport, as well as for maintaining normal auxin distribution within the root. Furthermore, this MADS-domain transcription factor upregulates PIN1 and PIN4 by direct binding to regulatory regions and it is required for PIN4-dependent auxin response.     COVID-19 Severity and Survival over Time in Patients with Hematologic Malignancies: A Population-Based Registry Study

    Mortality rates for COVID-19 have declined over time in the general population, but data in patients with hematologic malignancies are contradictory. We identified independent prognostic factors for COVID-19 severity and survival in unvaccinated patients with hematologic malignancies, compared mortality rates over time and versus non-cancer inpatients, and investigated post COVID-19 condition. Data were analyzed from 1166 consecutive, eligible patients with hematologic malignancies from the population-based HEMATO-MADRID registry, Spain, with COVID-19 prior to vaccination roll-out, stratified into early (February–June 2020; n = 769 (66%)) and later (July 2020–February 2021; n = 397 (34%)) cohorts. Propensity-score matched non-cancer patients were identified from the SEMI-COVID registry. A lower proportion of patients were hospitalized in the later waves (54.2%) compared to the earlier (88.6%), OR 0.15, 95%CI 0.11–0.20. The proportion of hospitalized patients admitted to the ICU was higher in the later cohort (103/215, 47.9%) compared with the early cohort (170/681, 25.0%, 2.77; 2.01–3.82). The reduced 30-day mortality between early and later cohorts of non-cancer inpatients (29.6% vs. 12.6%, OR 0.34; 0.22–0.53) was not paralleled in inpatients with hematologic malignancies (32.3% vs. 34.8%, OR 1.12; 0.81–1.5). Among evaluable patients, 27.3% had post COVID-19 condition.
