URB597 Administration decreases neuroinflammation and promotes autophagy in Alzheimer's disease models

In recent years, much attention has been focused on the endocannabinoid system as a therapeutic target for neurodegenerative diseases, including Alzheimer's disease (AD). The first endocannabinoid to be characterized was anandamide (N -arachidonoyl ethanolamine, AEA), which is hydrolyzed in postsynaptic neurons by the fatty acid amide hydrolase (FAAH). Notably, the post-mortem examination of brains from AD patients has revealed several alterations of the endocannabinoid system reporting a decrease in cortical AEA levels and a parallel increase in FAAH activity. On this ground, by using both cellular and a transgenic mouse model of AD, we aimed at determining whether pharmacological inhibition of FAAH via URB597 administration may affect microglia polarization and provide beneficial effects in restoring autophagic process and reducing amyloid-β (Aβ) aggregates, which are all neuropathological features of AD. The evaluation of morphological and major markers of microglial activation (iNOS and ARG-1) showed the ability of URB597 to revert microglial activation towards an anti-inflammatory condition, which was associated with the decrease of pro-inflammatory cytokines and an increase of anti-inflammatory cytokines together with the restoration of key factors involved in autophagic processes. These data were confirmed in vivo in the Tg2576 mice where the broad-spectrum action of endocannabinoids ranging from the anti-inflammatory action to the restoration of autophagic flux was demonstrated by the increase of proteins such as ATG7, Beclin1, LC3 and activation of ULK1 signaling following mTOR decrease after URB597 treatment. Moreover, treatment with URB597 decreases the expression of BACE1 through the modulation of CB2 receptors, leading to the reduction of amyloid plaques. Taken together, these data suggest that inhibition of FAAH can drive the polarization of microglia towards an anti-inflammatory phenotype and demonstrate that the modulation of the endocannabinoid system in AD, via URB597 administration, could be a potential and effective therapy not only to fight neuroinflammation but also to regulate the autophagic machinery

A multimethodological approach for the valorization of “Senatore Cappelli” wheat milling by-products as a source of bioactive compounds and nutraceutical activity

Wheat is the third most cultivated cereal in the world and represents the major contributor to human nutrition. Milling wheat by-products such as husks (17–20% of the total processing output weight), even if still containing high-value-added bioactive compounds, are often left untreated or unused, thus resulting in environmental and human health burdens. In these regards, the present study is aimed at evaluating in a multimethodological approach the nutraceutical properties of durum wheat husks belonging to the ancient cultivar “Senatore Cappelli”, thus assessing their potential as bioactive compound sources in terms of phytochemical, cytotoxic, and nutraceutical properties. By means of HPLC-FD analyses, wheat husk samples analyzed revealed a higher content of serotonin, amounting to 35% of the total BAs, and were confirmed to occur at biogenic amines quality index (BAQI) values <10 mg/100 g. In addition, spectrophotometric assays showed a signif- icant variable content in the phenolic (189.71–351.14 mg GAE/100 g) and antioxidant compounds (31.23–37.84 mg TE/100 g) within the wheat husk samples analyzed, according to the different cul- tivar areas of origin. Considering wheat husk extracts’ anti-inflammatory and antioxidant activity, in vitro analyses were performed on BV-2 murine microglia cells cultured in the presence or absence of LPS, thus evaluating their ability to promote microglia polarization towards an anti-inflammatory phenotype. Cytotoxicity assays showed that wheat extracts do not affect microglia viability. Wheat husks activity on microglial polarization was assessed by analyzing the expression of M1 and M2 markers’ mRNA by RT-PCR. Wheat husk antioxidant activity was assessed by analysis of NRF2 and SOD1 mRNA expression. Moreover, the sustainability assessment for the recovery of bioactive components from wheat by-products was carried out by applying the life cycle assessment (LCA) methodology using SimaPro v9.2.2. software

Daily Interpersonal Stress and the Stressor–Vulnerability Model of Alcohol Use

    Get PDF
We used an experience sampling design to examine the within–person, within–day associations among interpersonal stress, negative affect, and alcohol use, and how these associations varied as a function of alcohol–outcome expectancies (AOEs), avoidance coping style, sex, and neuroticism. Ninety–eight community adult drinkers who wanted to reduce their alcohol consumption (49 women) reported for 21 days on their interpersonal stress and affect (three times per day), and alcohol use (as it occurred) using hand–held computers. Several individual difference factors interacted with daytime interpersonal stress and afternoon negative affect in predicting nighttime alcohol use, with individuals high in careless unconcern AOEs or low in impairment AOEs demonstrating stronger positive associations between daytime stress and negative affect and nighttime drinking. Daytime drinking and individual difference factors also interacted in predicting nighttime interpersonal stress, with individuals high in careless unconcern AOEs or those low in impairment AOEs or avoidance coping style demonstrating the strongest positive associations between daytime drinking and nighttime stress. The interactive effects in predicting drinking outcomes were generally limited to days on which some interpersonal stress occurred

1 - Introductions of exotic macroalgae have increased recently on European shores. Each introductioninvolves at least one vector of transfer. For macroalgae, the potential vectors are aquaculture(intentional or accidental introduction), fouling on hulls, ballast water, aquarium trading, fishingnets.2 - Coastal lagoons, including Venice and Thau Lagoon, developed into major hotspots of marinemacrophyte introductions in the Mediterranean Sea. Moreover, in shallow lagoons and estuaries, eutrophication processes have resulted in the development of macroalgal biomass.3 - The most characteristic species of these macroalgal communities include members of the ulvophyceangenus Ulva L. In foliose Ulvales, simple morphology and anatomy, rampant convergence, remarkabledegrees of phenotypic plasticity in response to environmental factors tend to confound attempts atidentification and make cryptic introduction difficult to detect.4 - During a survey of macroalgal biodiversity in Venice Lagoon, among the pool of exotic speciesfound, there was an Ulva differing from the Atlantic and Mediterranean species in both vegetativeand reproductive features. Detailed observations consented us to identify the taxon as Ulva pertusaKjellman, previously reported in the Mediterranean only for Thau Lagoon

Perinatal S-Adenosylmethionine Supplementation Represses PSEN1 Expression by the Cellular Epigenetic Memory of CpG and Non-CpG Methylation in Adult TgCRD8 Mice

DNA methylation, the main epigenetic modification regulating gene expression, plays a role in the pathophysiology of neurodegeneration. Previous evidence indicates that 50 -flanking hypomethylation of PSEN1, a gene involved in the amyloidogenic pathway in Alzheimer’s dis- ease (AD), boosts the AD-like phenotype in transgenic TgCRND8 mice. Supplementation with S-adenosylmethionine (SAM), the methyl donor in the DNA methylation reactions, reverts the patho- logical phenotype. Several studies indicate that epigenetic signatures, driving the shift between normal and diseased aging, can be acquired during the first stages of life, even in utero, and manifest phenotypically later on in life. Therefore, we decided to test whether SAM supplementation during the perinatal period (i.e., supplementing the mothers from mating to weaning) could exert a protec- tive role towards AD-like symptom manifestation. We therefore compared the effect of post-weaning vs. perinatal SAM treatment in TgCRND8 mice by assessing PSEN1 methylation and expression and the development of amyloid plaques. We found that short-term perinatal supplementation was as effective as the longer post-weaning supplementation in repressing PSEN1 expression and amyloid deposition in adult mice. These results highlight the importance of epigenetic memory and methyl donor availability during early life to promote healthy aging and stress the functional role of non-CpG methylation
