    iLIF: illumination by Laser-Induced Fluorescence for single flash imaging on a nanoseconds timescale \ud

    The challenge in visualizing fast microscale fluid motion phenomena is to record high-quality images free of motion-blur. Here, we present an illumination technique based on laser-induced fluorescence which delivers high-intensity light pulses of 7 ns. The light source consists of a Q-switched Nd:YAG laser and a laser dye solution incorporated into a total internal reflection lens, resulting in a uni-directional light beam with a millimeter-sized circular aperture and 3° divergence. The laser coherence, considered undesirable for imaging purposes, is reduced while maintaining a nanoseconds pulse duration. The properties of the illumination by laser-induced fluorescence (iLIF) are quantified, and a comparison is made with other high-intensity pulsed and continuous light source

    Controlled impact of a disk on a water surface: Cavity dynamics

    In this paper we study the transient surface cavity which is created by the controlled impact of a disk of radius h0 on a water surface at Froude numbers below 200. The dynamics of the transient free surface is recorded by high speed imaging and compared to boundary integral simulations. An excellent agreement is found between both. The flow surrounding the cavity is measured with high speed particle image velocimetry and is found to also agree perfectly with the flow field obtained from the simulations. We present a simple model for the radial dynamics of the cavity based on the collapse of an infinite cylinder. This model accounts for the observed asymmetry of the radial dynamics between the expansion and contraction phase of the cavity. It reproduces the scaling of the closure depth and total depth of the cavity which are both found to scale roughly proportional to Fr^{1/2} with a weakly Froude number dependent prefactor. In addition, the model accurately captures the dynamics of the minimal radius of the cavity, the scaling of the volume Vbubble of air entrained by the process, namely Vbubble/h0^3 proportional (1 + 0.26Fr^{1/2})Fr^{1/2}, and gives insight into the axial asymmetry of the pinch-off process.Comment: 34 pages, 17 figures, submitted to the Journal of Fluid Mechanics.     Non-continuous Froude number scaling for the closure depth of a cylindrical cavity

    A long, smooth cylinder is dragged through a water surface to create a cavity with an initially cylindrical shape. This surface void then collapses due to the hydrostatic pressure, leading to a rapid and axisymmetric pinch-off in a single point. Surprisingly, the depth at which this pinch-off takes place does not follow the expected Froude1/3^{1/3} power-law. Instead, it displays two distinct scaling regimes separated by discrete jumps, both in experiment and in numerical simulations (employing a boundary integral code). We quantitatively explain the above behavior as a capillary waves effect. These waves are created when the top of the cylinder passes the water surface. Our work thus gives further evidence for the non-universality of the void collapse

    Normative data for the Dutch version of the Penn State Worry Questionnaire.

    Worry is a common symptom in various psychiatric problems and the key symptom of generalised anxiety disorder (GAD). The Penn State Worry Questionnaire (PSWQ) is the most widely used self-report scale for measuring worry. The present study provides normative data for the Dutch version of the PSWQ for a large community sample and a clinically referred sample of patients with GAD. Norms are not only provided for the original 16-item version, but also for an abbreviated 11-item version, which only consists of the positively worded items and has been shown to be a promising alternative to the full-length version. The percentile scores obtained for the community sample and the clinical GAD sample did not show much overlap, and this appeared true for the full-length as well as the abbreviated version of the PSWQ. These normative data seem suitable for differentiating between normal and abnormal manifestations of worrying and for evaluating the efficacy of treatments for GAD.     Repertoire Analysis of B-Cells Located in Striated Ducts of Salivary Glands of Patients With Sjogren's Syndrome

    A major complication of primary Sjögren's syndrome (pSS) is development of mucosa associated lymphoid tissue (MALT) B-cell lymphoma, particularly in salivary glands. These lymphomas express FcRL4 and are characteristically associated with lymphoepithelial lesions. Neoplastic B-cells may be derived from non-neoplastic glandular intraductal B-cells, also virtually all expressing FcRL4. A characteristic feature of MALT lymphomas is the production of rheumatoid factors (RFs), which are largely encoded by stereotypic immunoglobulin variable heavy chain (IGHV) sequences. The aim of this study was to examine whether there is a relationship between the intraductal and periductal B-cells and whether the intraductal B-cells are selected for RF. RNA was extracted from laser-microdissected infiltrated ductal areas and periductal infiltrates from frozen parotid gland tissue sections of 5 pSS patients. PCR amplified IGHV transcripts were cloned into pCR™4-TOPO vector and subsequently sequenced. Microdissected ducts yielded 96 unique IGHV sequences derived from intraductal B-cells, while 119 unique IGHV sequences were obtained from periductal infiltrates. No major difference in VH-gene usage was observed between intraductal and periductal B-cells. Nearly all (>90%) IGHV sequences derived from both intraductal and periductal B-cells were mutated. Clonal expansions as defined by shared VDJ rearrangements were also present among both intraductal and periductal B-cells: in total 32 clones were found, from which 12 were located within ducts, 15 in periductal areas, and five clones shared members in both areas. We observed 12 IGHV rearrangements encoding for RF sequences from which two were derived from intraductal B-cells and 10 from periductal B-cells. Nine RF sequences were part of a clone. Together these findings indicate that intraductal and periductal B-cells are closely related to each other. Intraductal B-cells are most likely derived from periductal B-cells. We did not obtain evidence that RF-specific B-cells are enriched within the striated ducts. We speculate that in principle any activated B-cell can enter the striated ducts from the periductal infiltrate, irrespective of its antigenic specificity. Within the ducts, these B-cells may receive additional activation and proliferation signals, to further expand at these sites and by acquisition of driver-mutations develop toward lymphoma

    Interprofessional Consensus Regarding Design Requirements for Liquid-Based Perinatal Life Support (PLS) Technology

    Liquid-based perinatal life support (PLS) technology will probably be applied in a first-in-human study within the next decade. Research and development of PLS technology should not only address technical issues, but also consider socio-ethical and legal aspects, its application area, and the corresponding design implications. This paper represents the consensus opinion of a group of healthcare professionals, designers, ethicists, researchers and patient representatives, who have expertise in tertiary obstetric and neonatal care, bio-ethics, experimental perinatal animal models for physiologic research, biomedical modeling, monitoring, and design. The aim of this paper is to provide a framework for research and development of PLS technology. These requirements are considering the possible respective user perspectives, with the aim to co-create a PLS system that facilitates physiological growth and development for extremely preterm born infants

    Evolutionary Analysis of Inter-Farm Transmission Dynamics in a Highly Pathogenic Avian Influenza Epidemic

    Phylogenetic studies have largely contributed to better understand the emergence, spread and evolution of highly pathogenic avian influenza during epidemics, but sampling of genetic data has never been detailed enough to allow mapping of the spatiotemporal spread of avian influenza viruses during a single epidemic. Here, we present genetic data of H7N7 viruses produced from 72% of the poultry farms infected during the 2003 epidemic in the Netherlands. We use phylogenetic analyses to unravel the pathways of virus transmission between farms and between infected areas. In addition, we investigated the evolutionary processes shaping viral genetic diversity, and assess how they could have affected our phylogenetic analyses. Our results show that the H7N7 virus was characterized by a high level of genetic diversity driven mainly by a high neutral substitution rate, purifying selection and limited positive selection. We also identified potential reassortment in the three genes that we have tested, but they had only a limited effect on the resolution of the inter-farm transmission network. Clonal sequencing analyses performed on six farm samples showed that at least one farm sample presented very complex virus diversity and was probably at the origin of chronological anomalies in the transmission network. However, most virus sequences could be grouped within clearly defined and chronologically sound clusters of infection and some likely transmission events between farms located 0.8–13 Km apart were identified. In addition, three farms were found as most likely source of virus introduction in distantly located new areas. These long distance transmission events were likely facilitated by human-mediated transport, underlining the need for strict enforcement of biosafety measures during outbreaks. This study shows that in-depth genetic analysis of virus outbreaks at multiple scales can provide critical information on virus transmission dynamics and can be used to increase our capacity to efficiently control epidemics
