Development and validation of fluorescence live-cell imaging approaches to study flavivirus infection kinetics in animal cells
The genus Flavivirus in the family Flaviviridae comprises many clinically important viruses,
such as dengue virus (DENV), Zika virus (ZIKV), and yellow fever virus (YFV). The quest for
therapeutic targets to combat flavivirus infections requires a better understanding of the
kinetics of the virus-cell interplay during infections with wild-type viral strains.
Nevertheless, this is hindered by limitations of the current cell-based systems for
monitoring flavivirus infection by live-cell imaging. The present dissertation describes the
development and validation of fluorescence-activatable sensors to detect the activity of
flavivirus NS2B-NS3 serine proteases in living cells. The system consists of green fluorescent
protein (GFP)-based reporters that become fluorescent upon cleavage by recombinant
DENV-2/ZIKV proteases in vitro. A version of this sensor containing the flavivirus internal
NS3 cleavage site linker (AAQRRGRIG) reported the highest fluorescence activation in stably
transduced mammalian cells upon DENV-2/ZIKV infection. The onset of fluorescence
correlated with viral protease activity. Moreover, a far-red version of this flavivirus sensor
presented the best signal-to-noise ratio in a fluorescent Dulbecco’s plaque assay, leading to
the construction of a multireporter platform combining the flavivirus sensor with DNA
fluorescent dyes for the detection of virus-induced chromatin condensation and cell death
(cytophatic effect labeling). This enabled studies of viral plaque formation with a single-cell
resolution. This cytopathic effect labeling approach was conceptualized and validated
during the present work. Finally, the application of the multireporter platform also enabled
the study of kinetics of infection and cytophatic effect induction by DENV-2, ZIKV, and YFV
in cell-subpopulations. We anticipate that future studies of viral infection kinetics with our
reporter systems will enable basic investigations of virus-host cell interactions and will also
facilitate the screening of antiviral drugs to manage flavivirus infections.El género Flavivirus de la familia Flaviviridae incluye muchos virus de importancia médica,
como el virus del dengue (DENV), el virus Zika (ZIKV) y el virus de la fiebre amarilla (YFV). La
búsqueda de blancos terapéuticos para combatir las afecciones causadas por flavivirus
requiere un mejor entendimiento de la cinética de interacción virus-célula durante las
infecciones con cepas virales silvestres. Sin embargo, esto se ve obstaculizado por las
limitaciones de los sistemas celulares actuales para monitorear la infección por flavivirus
mediante imagenología de células vivas. La presente tesis describe el desarrollo y validación
de sensores fluorescentes activables para detectar la actividad de la serin proteasa flaviviral
NS2B-NS3 en células vivas. El sistema consta de reporteros basados en la proteína verde
fluorescente (GFP) que activan la fluorescencia al ser cortados por proteasas recombinantes
de DENV-2/ZIKV in vitro. Tras la infección por DENV-2/ZIKV, una versión de este sensor que
contiene el sitio de corte interno de la proteína NS3 de flavivirus (AAQRRGRIG) reportó la
mayor activación de fluorescencia en células de mamífero transducidas de manera estable.
La activación de la fluorescencia correlacionó con la actividad de la proteasa viral. Además,
una versión de color rojo lejano de este sensor de flavivirus presentó la mejor relación
señal/ruido en un ensayo de placas de Dulbecco fluorescentes, lo que llevó a la construcción
de una plataforma multireportero que combina el sensor de flavivirus con sondas
fluorescentes de intercalado en el ADN para la detección de condensación de cromatina y
muerte celular inducida por el virus (marcaje del efecto citopático). Esto permitió realizar
estudios de formación de placas virales con resolución a nivel de células individuales. Dicho
abordaje para el marcaje del efecto citopático fue conceptualizado y validado durante el
presente trabajo. Finalmente, la aplicación de la plataforma multireportero también
posibilitó el estudio de la cinética de infección a nivel de subpoblaciones celulares, así como
de la inducción del efecto citopático por DENV-2, ZIKV y YFV. Anticipamos que estudios
futuros de la cinética de infección viral con nuestros sistemas reporteros permitirán
investigaciones básicas de la interacción virus-célula huésped y facilitarán el tamizaje de

Generation and implementation of reporter BHK-21 cells for live imaging of flavivirus infection
The genus Flavivirus within the family Flaviviridae includes many viral species of medical
importance, such as yellow fever virus (YFV), Zika virus (ZIKV), and dengue virus (DENV), among
others. Presently, the identification of flavivirus-infected cells is based on either the immunolabeling of
viral proteins, the application of recombinant reporter replicons and viral genomes, or the use of cellbased molecular reporters of the flaviviral protease NS2B-NS3 activity. Among the latter, our flavivirusactivatable GFP and mNeptune reporters contain a quenching peptide (QP) joined to the fluorescent
protein by a linker consisting of a cleavage site for the flavivirus NS2B-NS3 proteases (AAQRRGRIG).
When the viral protease cleaves the linker, the quenching peptide is removed, and the fluorescent
protein adopts a conformation promoting fluorescence. Here we provide a detailed protocol for the
generation, selection and implementation of stable BHK-21 cells expressing our flavivirus geneticallyencoded molecular reporters, suitable to monitor the viral infection by live-cell imaging. We also describe
the image analysis procedures and provide the required software pipelines. Our reporter cells allow the
implementation of single-cell infection kinetics as well as plaque assays for both reference and native

The mast cell integrates the splanchnic and systemic inflammatory response in portal hypertension
Portal hypertension is a clinical syndrome that is difficult to study in an isolated manner since it is always associated with a greater or lesser degree of liver functional impairment. The aim of this review is to integrate the complications related to chronic liver disease by using both, the array of mast cell functions and mediators, since they possibly are involved in the pathophysiological mechanisms of these complications. The portal vein ligated rat is the experimental model most widely used to study this syndrome and it has been considered that a systemic inflammatory response is produced. This response is mediated among other inflammatory cells by mast cells and it evolves in three linked pathological functional systems. The nervous functional system presents ischemia-reperfusion and edema (oxidative stress) and would be responsible for hyperdynamic circulation; the immune functional system causes tissue infiltration by inflammatory cells, particularly mast cells and bacteria (enzymatic stress) and the endocrine functional system presents endothelial proliferation (antioxidative and antienzymatic stress) and angiogenesis. Mast cells could develop a key role in the expression of these three phenotypes because their mediators have the ability to produce all the aforementioned alterations, both at the splanchnic level (portal hypertensive enteropathy, mesenteric adenitis, liver steatosis) and the systemic level (portal hypertensive encephalopathy)
Inflammation: a way to understanding the evolution of portal hypertension
<p>Abstract</p> <p>Background</p> <p>Portal hypertension is a clinical syndrome that manifests as ascites, portosystemic encephalopathy and variceal hemorrhage, and these alterations often lead to death.</p> <p>Hypothesis</p> <p>Splanchnic and/or systemic responses to portal hypertension could have pathophysiological mechanisms similar to those involved in the post-traumatic inflammatory response.</p> <p>The splanchnic and systemic impairments produced throughout the evolution of experimental prehepatic portal hypertension could be considered to have an inflammatory origin. In portal vein ligated rats, portal hypertensive enteropathy, hepatic steatosis and portal hypertensive encephalopathy show phenotypes during their development that can be considered inflammatory, such as: ischemia-reperfusion (vasodilatory response), infiltration by inflammatory cells (mast cells) and bacteria (intestinal translocation of endotoxins and bacteria) and lastly, angiogenesis. Similar inflammatory phenotypes, worsened by chronic liver disease (with anti-oxidant and anti-enzymatic ability reduction) characterize the evolution of portal hypertension and its complications (hepatorenal syndrome, ascites and esophageal variceal hemorrhage) in humans.</p> <p>Conclusion</p> <p>Low-grade inflammation, related to prehepatic portal hypertension, switches to high-grade inflammation with the development of severe and life-threatening complications when associated with chronic liver disease.</p
A Fluorescent Real-Time Plaque Assay Enables Single-Cell Analysis of Virus-Induced Cytopathic Effect by Live-Cell Imaging
Conventional plaque assays rely on the use of overlays to restrict viral infection allowing
the formation of distinct foci that grow in time as the replication cycle continues leading to countable
plaques that are visualized with standard techniques such as crystal violet, neutral red, or immuno-
labeling. This classical approach takes several days until large enough plaques can be visualized
and counted with some variation due to subjectivity in plaque recognition. Since plaques are clonal
lesions produced by virus-induced cytopathic effect, we applied DNA fluorescent dyes with differen-
tial cell permeability to visualize them by live-cell imaging. We could observe different stages of that
cytopathic effect corresponding to an early wave of cells with chromatin-condensation followed by a
wave of dead cells with membrane permeabilization within plaques generated by different animal
viruses. This approach enables an automated plaque identification using image analysis to increase
single plaque resolution compared to crystal violet counterstaining and allows its application to
plaque tracking and plaque reduction assays to test compounds for both antiviral and cytotoxic
activities. This fluorescent real-time plaque assay sums to those next-generation technologies by
combining this robust classical method with modern fluorescence microscopy and image analysis

Aporte de la regulación metacognitiva al aprendizaje de competencias TIC
Los procesos educativos han incorporado las TIC en la enseñanza de las ciencias generado ambientes de grandes contribuciones, no solo a la enseñanza de los saberes científicos, sino también al fortalecimiento de las múltiples dimensiones del pensamiento crítico. Surgiendo la necesidad, en diferentes estudios, de estudiantes competentes en el manejo de estas tec-nologías. Es así como esta investigación pretende determinar el aporte de la regulación mtacognitiva al aprendizaje de las competencias TIC, en estudiantes de grado cuarto y quinto de educación básica por medio de la aplicación de una unidad didáctica en torno al manejo de las TIC, fomentando competencias en cuanto al manejo de la información y de la tecnología. Rediseño de la identidad corporativa de la empresa C&C fábrica textil y distribuciones
Este proyecto de titulación busca rediseñar el isologo de la empresa C&C Fábrica textil,
siguiendo la metodología de Ambroise Harris. Este proceso cuenta de siete etapas las cuales
son la definición del problema, investigación del mismo, ideación, prototipo, selección,
implementación y comprobación. Cada una de estas etapas se desarrolla con la ayuda de
herramientas creativas tales como el brainstorm, el moodboard, el bocetado y principalmente
el brief. El brief desempeña un papel fundamental en todo este proyecto debido a que es esta
herramienta que forma parte de la etapa de definición del problema, es en donde se obtendrá
la mayor cantidad de información que ayuda a la construcción de la propuesta. Finalmente, al
concluir todo el proceso creativo se realiza un manual de usuario que contará con mockups
de cómo se puede utilizar la propuesta gráfica en diferentes formatos, o aplicaciones prácticas.
Junto con el manual y la propuesta de identificador gráfico se realiza un feedback de todo el
proyecto y se obtiene una conclusión que denotará los resultados obtenidos a lo largo de todo
el proyecto.This degree project seeks to redesign the isologist of the company C&C Textile Factory,
following the methodology of Ambrose Harris. This process has seven stages which are the
definition of the problem, investigation of it, ideation, prototype, selection, implementation and
testing. Each of these stages is developed with the help of creative tools such as brainstorm,
moodboard, sketching and mainly the brief. The brief plays a fundamental role in this whole
project because it is this tool that is part of the problem definition stage, is where you will get
the most information that helps the construction of the proposal. Finally, at the end of the entire
creative process, a user manual is made that will have mockups of how the graphic proposal
can be used in different formats, or practical applications. Together with the manual and the
graphic identifier proposal, a feedback of the entire project is made and a conclusion is

Rediseño de la planta de producción de Bornes de Risaralda para el mejoramiento de sus procesos
El concepto de diseño de planta se ha utilizado durante décadas con el fin de conseguir un mejoramiento en los procesos y una mayor capacidad de producción, actualmente Bornes de Risaralda necesita de un rediseño de planta para poder suplir las necesidades del mercado nacional e intentar penetrar mercados internacionales vecinos como es el caso de Ecuador. En el desarrollo de este proyecto encontraremos los métodos utilizados durante varias décadas para resolver este problema de diseño de planta al igual que las normas básicas que se deben de tener en cuenta para una planta de fundición de metales como lo es la empresa Bornes de Risaralda. Del mismo modo se realiza un análisis a los procesos desarrollados dentro de la planta con el fin de darle una mejora que permita aumentar la producción de la planta. Luego de analizar la situación actual de la planta se empieza a determinar las variables y procesos que se deben cambiar, mejorar o implementar según la importancia que tenga dentro del proceso productivo. Durante el desarrollo del proyecto se trabaja con la metodología de Muther y su plan Layout para la distribución de planta, ya que ha sido el método más implementado para la solución de este tipo de problemas obteniendo buenos resultados para las organizaciones. Al tener el análisis actual de la empresa completo, es más sencillo establecer los puntos a cambiar y realizar un bosquejo de la distribución apropiada para la empresa teniendo como base su diagrama de flujo de materiales y como restricción el actual espacio de la planta, dado que el proyecto se realiza sobre unas instalaciones previamente diseñadas según las necesidades. Finalmente se obtiene una distribución que satisface las necesidades de los procesos de la planta de producción y logra empalmar el flujo de materiales con el principio de mínima distancia, para conseguir una distribución actualizada, moderna en algunos aspectos y primordialmente más segura y productiva para los empleados y la empresa. Esta nueva imagen de Bornes de Risaralda le permitirá aumentar su producción al disminuir enormemente las distancias entre estaciones y los largos tiempos de espera en las estaciones cuello de botella. Se espera que la empresa tome las recomendaciones dadas para así hacer de este proyecto más que un requisito de grado, una solución óptima a los problemas de distribución de la empresa.The plant design concept has been used for decades in order to achieve an improvement in processes and increased production capacity, Bornes de Risaralda currently needs a plant redesign to meet the needs of the domestic market and try to penetrate markets international neighbors as is the case of Ecuador. This new image of Bornes de Risaralda allow you to increase production to greatly reduce the distances between stations and the long waiting times at the bottleneck stations. Company is expected to take the recommendations given in order to make this project more than a degree requirement, an optimal solution to the problems of distribution of the company
