    B cells are capable of independently eliciting rapid reactivation of encephalitogenic CD4 T cells in a murine model of multiple sclerosis

    <div><p>Recent success with B cell depletion therapies has revitalized efforts to understand the pathogenic role of B cells in Multiple Sclerosis (MS). Using the adoptive transfer system of experimental autoimmune encephalomyelitis (EAE), a murine model of MS, we have previously shown that mice in which B cells are the only MHCII-expressing antigen presenting cell (APC) are susceptible to EAE. However, a reproducible delay in the day of onset of disease driven by exclusive B cell antigen presentation suggests that B cells require optimal conditions to function as APCs in EAE. In this study, we utilize an <i>in vivo</i> genetic system to conditionally and temporally regulate expression of MHCII to test the hypothesis that B cell APCs mediate attenuated and delayed neuroinflammatory T cell responses during EAE. Remarkably, induction of MHCII on B cells following the transfer of encephalitogenic CD4 T cells induced a rapid and robust form of EAE, while no change in the time to disease onset occurred for recipient mice in which MHCII is induced on a normal complement of APC subsets. Changes in CD4 T cell activation over time did not account for more rapid onset of EAE symptoms in this new B cell-mediated EAE model. Our system represents a novel model to study how the timing of pathogenic cognate interactions between lymphocytes facilitates the development of autoimmune attacks within the CNS.</p></div

    Universal Fluctuations in Correlated Systems

    The probability density function (PDF) of a global measure in a large class of highly correlated systems has been suggested to be of the same functional form. Here, we identify the analytical form of the PDF of one such measure, the order parameter in the low temperature phase of the 2D-XY model. We demonstrate that this function describes the fluctuations of global quantities in other correlated, equilibrium and non-equilibrium systems. These include a coupled rotor model, Ising and percolation models, models of forest fires, sand-piles, avalanches and granular media in a self organized critical state. We discuss the relationship with both Gaussian and extremal statistics.Comment: 4 pages, 2 figure

    Evaluation of two interaction techniques for visualization of dynamic graphs

    Several techniques for visualization of dynamic graphs are based on different spatial arrangements of a temporal sequence of node-link diagrams. Many studies in the literature have investigated the importance of maintaining the user's mental map across this temporal sequence, but usually each layout is considered as a static graph drawing and the effect of user interaction is disregarded. We conducted a task-based controlled experiment to assess the effectiveness of two basic interaction techniques: the adjustment of the layout stability and the highlighting of adjacent nodes and edges. We found that generally both interaction techniques increase accuracy, sometimes at the cost of longer completion times, and that the highlighting outclasses the stability adjustment for many tasks except the most complex ones.Comment: Appears in the Proceedings of the 24th International Symposium on Graph Drawing and Network Visualization (GD 2016

    Background Rejection in the DMTPC Dark Matter Search Using Charge Signals

    The Dark Matter Time Projection Chamber (DMTPC) collaboration is developing low-pressure gas TPC detectors for measuring WIMP-nucleon interactions. Optical readout with CCD cameras allows for the detection for the daily modulation in the direction of the dark matter wind, while several charge readout channels allow for the measurement of additional recoil properties. In this article, we show that the addition of the charge readout analysis to the CCD allows us too obtain a statistics-limited 90% C.L. upper limit on the ee^- rejection factor of 5.6×1065.6\times10^{-6} for recoils with energies between 40 and 200 keVee_{\mathrm{ee}}. In addition, requiring coincidence between charge signals and light in the CCD reduces CCD-specific backgrounds by more than two orders of magnitude.Comment: 8 pages, 6 figures.     Tendinopathy—from basic science to treatment

    Chronic tendon pathology (tendinopathy), although common, is difficult to treat. Tendons possess a highly organized fibrillar matrix, consisting of type I collagen and various 'minor' collagens, proteoglycans and glycoproteins. The tendon matrix is maintained by the resident tenocytes, and there is evidence of a continuous process of matrix remodeling, although the rate of turnover varies at different sites. A change in remodeling activity is associated with the onset of tendinopathy. Major molecular changes include increased expression of type III collagen, fibronectin, tenascin C, aggrecan and biglycan. These changes are consistent with repair, but they might also be an adaptive response to changes in mechanical loading. Repeated minor strain is thought to be the major precipitating factor in tendinopathy, although further work is required to determine whether it is mechanical overstimulation or understimulation that leads to the change in tenocyte activity. Metalloproteinase enzymes have an important role in the tendon matrix, being responsible for the degradation of collagen and proteoglycan in both healthy patients and those with disease. Metalloproteinases that show increased expression in painful tendinopathy include ADAM (a disintegrin and metalloproteinase)-12 and MMP (matrix metalloproteinase)-23. The role of these enzymes in tendon pathology is unknown, and further work is required to identify novel and specific molecular targets for therapy

    Permeability Estimates of Self-Affine Fracture Faults Based on Generalization of the Bottle Neck Concept

    We propose a method for calculating the effective permeability of two-dimensional self-affine permeability fields based on generalizing the one-dimensional concept of a bottleneck. We test the method on fracture faults where the local permeability field is given by the cube of the aperture field. The method remains accurate even when there is substantial mechanical overlap between the two fracture surfaces. The computational efficiency of the method is comparable to calculating a simple average and is more than two orders of magnitude faster than solving the Reynolds equations using a finite-difference scheme

    First Dark Matter Search Results from a Surface Run of the 10-L DMTPC Directional Dark Matter Detector

    The Dark Matter Time Projection Chamber (DMTPC) is a low pressure (75 Torr CF4) 10 liter detector capable of measuring the vector direction of nuclear recoils with the goal of directional dark matter detection. In this paper we present the first dark matter limit from DMTPC. In an analysis window of 80-200 keV recoil energy, based on a 35.7 g-day exposure, we set a 90% C.L. upper limit on the spin-dependent WIMP-proton cross section of 2.0 x 10^{-33} cm^{2} for 115 GeV/c^2 dark matter particle mass.Comment: accepted for publication in Physics Letters
